    Notes: Abstract. Growth and cytology of root apical meristems of Chinese Spring wheat were studied in relation to temperature. The maximum rate of growth increased with temperature, a marked rise occurring between 10°C and 12°C. At all temperatures studied nucleolar volume increased to a maximum and then declined. The maximum nucleolar size achieved showed particular temperature sensitivity, higher volumes being attained at lower temperatures. The peak at 5°C was 70% higher than at 20°C. However, in comparison, cell and nuclear volumes were only 38% and 47% larger, respectively. Ultrastructural analysis of the nucleoli revealed a temperature-dependent relationship between the proportion of granular component and dense fibrillar component. The results are discussed in relation to the regulation of ribosome synthesis and function during growth and development at different temperatures.

    Notes: Abstract Serial sectioning of wheat roots prepared for electron microscopy was used to count the number of fibrillar centres per nucleus and nucleolus and to calculate their sizes. After growth at 35 ° C for two days the nucleoli became segregated and a one to one relationship was evident between chromosomal nucleolar organizers and fibrillar centres. This was confirmed using an aneuploid line carrying an additional pair of organizers. Quantitative studies showed that the fibrillar centres occupied a volume 0.24% of the total chromatin reticulum. From this it was calculated that only about 1/3 of the fibrillar centre material was likely to be nucleolar organizer chromatin. The other material was considered to be the protein revealed in silver staining studies. The importance of this was shown by its constant ratio to the size of all nucleoli in a given nucleus. — Evidence was found for the movement and fusion of organizer flanking regions during growth at 35 ° C. The number of junctions between chromatin and nucleolar organizers drops by about half giving one per organizer after segregation, and serial sectioning demonstrated such junctions in close proximity, an arrangement suggestive of incipient fusion.

    Notes: Summary Wheat roots from germinating seedlings of Chinese spring wheatTriticum aestivum grown for 36 hours at 20°C were examined by conventional thin-section electron microscopy. Virus-like particles were seen inside a large cytoplasmic intrusion into the nucleus having the appearence of a nucleolar vacuole. The particles were isometric and about 50 nm in diameter with a membrane-like coat and a small core. The cytoplasmic intrusion was bounded by nuclear envelope with pores apparent where it abutted nucleoplasm. The particles are similar to previously reported solitary particles “S bodies” from a range of plants but are also similar in size and morphology to the retroposon particles associated with copia like elements in other organisms. The position of the virus-like particles in the young wheat roots is discussed in relation to interactions with components of the cell skeleton.

    Notes: Summary Nuclear bodies are found in interphase nuclei of root apices of a number of plant species. They often show differences in structure and position relative to the nucleolus and this has led to an attempt to define two classes of body. However, in some species their separation into two classes on structural grounds alone breaks down, indicating that although they may occupy different positions within the nucleus they may in these particular cases be only different forms of the same body. The two extremes of the range of bodies examined represent what have been called “nucleolus-associated body” (karyosome) and “dense body”. The nucleolus-associated body is typically attached to, or adjacent to, the nucleolus. It is composed of fibrils 4–8 nm wide and often has an open structure showing compound threads or fibrils separated from each other by electron-lucent spaces. The dense body is more compact in structure and typically lies free in the nucleoplasm. Both types of body have an affinity for silver ions which, together with their staining reaction following treatment with EDTA, indicates that they consist of ribonucleoprotein. The characteristics of nuclear bodies found in different plant species have some relationship with the structure and DNA content of the interphase nucleus. Nucleolus-associated bodies are characteristic of species with an areticulate nuclear structure (2 C DNA content 〈6 pg), while dense bodies are common in species with a reticulate nuclear structure (2 C DNA content 〉6 pg). The possible functions of the two forms of nuclear body are discussed.

    Notes: Summary Vacuole-like structures were found in the nuclei of root tip cells ofBrassica napus. The cells containing the unusual nuclear inclusions were found to be adjacent to zones of degenerating cells. Such groups of cells occurred irregularly in the meristematic regions of the young root tips. The possibility that they represent changes which have occurred in old seeds is discussed. The “vacuole-like” structures seen in the cells adjacent to the degenerating zones were bounded by a membranous layer 12 nm thick. This is thicker than most cellular membranes. The “vacuoles” frequently contained inclusions and showed similarities to protein bodies reported elsewhere. The structures are thought to represent rearrangements of cell products which may have accumulated through an imbalance of metabolism in consequence of the imminent cell degeneration.
