    Notes: Abstract: A survey was made of the presence, copy number and location of the Salmonella-specific DNA insertion element IS200, within the genomes of the 27 phage type strains of Salmonella enteritidis. All the phage type strains contained copies of IS200 revealed by genomic Southern blot hybridizations with a 300-bp DNA probe internal to the element. Restriction site variation around IS200 insertion sites was examined. Three fundamental patterns of hybridization corresponding to chromosomal IS200 loci were found. In terms of population genetics, these ‘IS200 profiles’ correspond to clonal lineages of recent evolutionary origin, and underlike the phage-typing scheme for epidemiological subdivision of S. enteritidis. The molecular analysis is consistent with genetic selection pressures which are apparent in the observed epidemiological distribution of S. enteritidis, since each clonal lineage contained one of the phage types of major clinical importance in the U.K.

    Notes: Six epidemiologically distinct ancestral strains of Salmonella enteritidis and 5 of S. typhimurium from the preantibiotic era were examined for plasmid content, and for presence of plasmid genes implicated in mouse-virulence. Five sizes of plasmid were detected in S. enteritidis varying from 1 to 60 MDa. Two sizes of plasmid were found in S. typhimurium, 28 and 60 MDa. Plasmids of the same size were not common to both serovars. The HindIII restriction patterns of 3 of the ancestral S. enteritidis plasmids were identical to the modern 38 MDa plasmid, while all contained identical bands of 3.5, 2.7 and 1.9 kb. All the 60-MDa S. typhimurium plasmids, ancestral and contemporary, has an identical restriction pattern. Three different sized S. enteritidis plasmids and one size S. typhimurium plasmid contained a 3.5-kb DNA fragment carrying the virulence locus VirA. The VirB virulence locus was located on a 2.7-kb DNA fragment in S. enteritidis and on a 2.5-kb fragment in S. typhimurium. Both loci were precisely conserved between the ancestral strains and the modern representatives of both serovars.

    Notes: Manual sexing of redclaw crayfish Cherar quadricurinatus was undertaken to evaluate the feasibility of monosex culture and to establish growth and yield characteristics of single sex populations. Although the all-male population exhibited greater mean weight after 10 mo of growout, all-female and mixed sex populations achieved greater total yields due to significant juvenile recruitment. Commercial cultivation of all-male populations would be advantageous as in-pond reproduction is greatly abated, growth is superior and marketable yield is higher. Apparent sexual lability of juveniles combined with the laborious nature of manual sexing may render manual sexing unsuitable for commercial applications.

    Notes: An experiment was designed to assess the relative performance of several shelter types on the production of redclaw crayfish Cherar quadricarinatus under earthen pond conditions. Juvenile crayfish with a mean stocking weight of 12.76 g were cultured in 24 net pens at 12.5 m2 within a 0.2 ha earthen pond, and were provided with one of five shelter types or no shelter over 162 d. The various shelter types assessed were: mesh bundles, pipe stacks, car tires, elevated cement/fiber-board sheets, and cement/fiber-board sheets laid flat on the substrate. Crayfish in each pen were fed a formulated pellet diet three times each week. Treatments were assessed on the basis of crayfish growth, survival, harvest biomass and berry rate among females at harvest. There was no significant effect (P 〉 0.05) of shelter type on growth; however, there was a highly significant effect (P 〈 0.001) on survival with mesh bundles (75.1%), tires (51.4%), and pipe stacks (43.25%) performing significantly better than the no shelter control (15%). The consequence of equal growth and differing survival among treatments produced significant differences in harvest biomass (P 〈 0.01). There were also significant differences in berry rate for the different treatments (P= 0.002), and a significant negative correlation of berry rate on survival for those treatments where shelter was provided. This experiment showed that the shelter types provided during pond culture had a significant effect on harvested biomass, primarily through its effect on survival. The efficacy of the mesh bundle shelters may be attributable to the variability in the size of the spaces provided, and the ability of the mesh to separate many individuals relative to the overall volume of the shelter.

    Notes: Why do microbes make secondary products? That question has been the subject of intense debate for many decades. There are two extreme opinions. Some argue that most secondary metabolites play no role in increasing the fitness of an organism. The opposite view, now widely held, is that every secondary metabolite is made because it possesses (or did possess at some stage in evolution) a biological activity that endows the producer with increased fitness. These opposing views can be reconciled by recognizing that, because of the principles governing molecular interactions, potent biological activity is a rare property for any molecule to possess. Consequently, in order for an organism to evolve the rare potent, biologically active molecule, a great many chemical structures have to be generated, most of which will possess no useful biological activity. Thus, the two sides of the debate about the role and evolution of secondary metabolism can be accommodated within the view that the possession of secondary metabolism can enhance fitness, but that many products of secondary metabolism will not enhance the fitness of the producer. It is proposed that secondary metabolism will have evolved such that traits that optimize the production and retention of chemical diversity at minimum cost will have been selected. Evidence exists for some of these predicted traits. Opportunities now exist to exploit these unique properties of secondary metabolism to enhance secondary product diversity and to devise new strategies for biotransformation and bioremediation.
