    Notes: The potential impact of optical fibre sensors embedded into medical textiles for thecontinuous monitoring of the patient during Magnetic Resonance Imaging (MRI) is presented. Inthat way, we report on several pure optical sensing technologies for pulse oximetry and respiratorymovements monitoring. The technique for pulse oximetry measurement is known as NIRS (NearInfra-Red Spectroscopy) in a reflectance mode. In parallel, we tested two different optical sensorbased fabric designs breathing activity detection – a macro bending sensor and a fibre Bragg gratingsensor consisting in respiratory frequency measurement by intensity variation detection and opticalspectral analysis

    Notes: SYNOPSIS. Naegleria fowleri strains HB-1 and KUL, pathogenic for humans, Naegleria gruberi strain 1518/1e, and 3 strains (Vm1, LvH1, and LvH2) of Naegleria isolated from a body of water polluted with thermal effluents were compared in an attempt at specific identifications of the latter strains. The 3 environmental isolates were morphologically almost identical with N. fowleri and had almost the same temperature tolerance, although at 37 and 42 C the growth rates of LvH1 and LvH2 were higher than those of the human pathogen, N. fowleri, and of isolate Vm1, which was pathogenic for mice. Serologic examinations by indirect fluorescent antibody method revealed a very close relationship of the new isolates with the human pathogens. While Vm1 was indistinguishable from N. fowleri, LvH1 and LvH2 were not, when cross-absorbed antisera were used. Of all the strains examined, only the 2 LvH isolates were not inhibited by amphotericin B, while only N. gruberi was not inhibited by fumagillin. The cytopathic effect in Vero cell cultures suggested that the LvH strains could have a certain degree of virulence, although this was not confirmed by intranasal and intracerebral inoculations of mice. The cytopathic effects of the human pathogens and of the isolate pathogenic for mice were related to their virulence for mice. It is concluded that there exists an intermediate form between N. gruberi and N. fowleri, with a strong relationship to the latter species. We refer to such strains as nonpathogenic variants of N. fowleri. Further research is needed to reveal their place in the taxonomy.

    Notes: Small subunit ribosomal DNA (SSU rDNA) comparisons have revealed that not only species but even genus identification based on morphology is unreliable in the family Vahlkampfiidae, and SSU rDNA sequences have been used to redefine vahlkampfiid genera. However, it is time consuming to use SSU rDNA sequences for species identification. The ITS region (including the 5.8S rDNA) has the advantage that it is much shorter than the SSU rDNA and thus easier and quicker to obtain. Initially, 29 strains of 13 species belonging to seven vahlkampfiid genera were investigated. It was found that each species has a specific ITS sequence, which appears to be conserved within a species. The same had previously been found in the vahlkampfiid genus Naegleria. Consequently, we have demonstrated that the ITS region (including the 5.8S rDNA) can be used to identify vahlkampfiid amoebae. In the current investigation, the ITS sequences of 16 undescribed vahlkampfiid isolates were determined. Based on these sequences, seven isolates were identified as belonging to described species, while nine represent six new species. Four of the new species belong to the genus Tetramitus, and one each to the genera Vahlkampfia and Paravahlkampfia.

    Notes: Eight isolates of Naegleria australiensis were obtained from a small lake in Tulsa, Oklahoma. The eight strains were isolated during the hot summer months of July through September, when water temperatures ranged from 27 to 33°C. All eight isolates were pathogenic for mice. The mean time to death for mice was 10 days (range 6–13 days). This pathogenic free-living ameba has not before been reported from the United States or the Western Hemisphere.

    Notes: Summary For the first time three strains of the newly described species of pathogenic Naegleria,N. australiensis De Jonckheere 1981 were isolated from a thermally polluted canal in Western Germany. They were pathogenic for mice with a lower mortality and a longer incubation period compared to mice infected withN. fowleri (Carter 1970). By the immunofluorescence-method these strains could be identified asN. australiensis.

    Notes: Abstract The nonpathogenic amoebaNaegleria lovaniensis is closely related to the human pathogenN. Fowleri. Both grow at a maximal temperature of 45°C and, therefore, are often found together in the environment. As they are morphologically inseparable at the light-microscope level, refined techniques are necessary to separate the two species. I have used restriction-fragment-length polymorphism analysis of the polymerase chain reaction (PCR)-amplified ribosomal RNA gene, or riboprinting, to distinguish between the differentNaegleria spp. Riboprints generated from the small subunit and the large subunit separateN. fowleri fromN. lovaniensis. To examine the taxonomic relationships among allNaegleria spp., analysis of the large subunit has to be performed; the small subunit contains a 1.3-kb group I intron, which interferes with tree building based upon restriction sites.

    Notes: A method has been developed for quantitative determination of pinaverium-bromide, a quaternary ammonium derivative with papaverine-like activity, in human serum. The method involves a chloroform extraction of serum spiked with N-(6,6-dimethyl bicyclo[3.1.1]2-heptenyl-ethoxyethyl)perhydro-1,4-oxazine as internal standard. After evaporation of the solvent, and reduction of the residue with Raney-Nickel, the internal standard and the reduced pinaverium-bromide are re-extracted from the reaction mixture with toluene and analysed isothermally on a fused silica column coated with OV-101. Although chemical ionization with methane revealed intense protonated molecular ions for both pinaverium-bromide and the internal standard, selectivity and sensitivity were significantly lower in comparison with electron impact ionization at 70 eV. Therefore, quantification was performed in the electron impact mode by single ion monitoring of the common fragment ion at m/z 100.2. A linear detector response was observed up to 160 ng ml-1. A within-run assay precision better than 2% CV (n=5) was found, and a detection limit of 1 ng pinaverium-bromide ml-1 of serum was attained.

    Notes: As a substantial part in the development of a reference material in clinical chemistry, a highly accurate and precise isotope dilution mass spectrometric method has been worked out for the determination of cortisol in human serum. The candidate definitive method consists of addition of 4-[14C]cortisol and, following equilibration, solvent extraction of cortisol and its internal standard from the serum matrix. After conversion into methoxime-trimethylsilyl derivatives, the extract is purified by gel chromatography. Measurements are made by combined capillary gas chromatography mass spectrometry. The ratio of peak heights at m/z 605 and 607 for each sample and calibration mixtures is used for quantification. To ensure maximum accuracy each set of calibration mixtures was composed from two stock solutions and closely bracketed the anticipated serum concentration. An analysis of variance, involving comparison of within-run and between-run variability, gave a total coefficient of variation (CV) of 0.38% for a serum pool containing 90.79 ng cortisol ml-1 serum.
