Zusammenfassung 1. Es wurden die Veränderungen der proteolytischen Aktivität des Mitteldarms von Galleria mellonella im Laufe der Larvenentwicklung des vorletzten und letzten Stadiums und während der Puppenentwicklung untersucht. 2. Die proteolytische Aktivität des Mitteldarms erhöht sich im vorletzten Larvenstadium bis zur Häutungsperiode. Zur Zeit der Häutung steigt die Aktivität nicht an; sie nimmt erst während der ersten Hälfte des letzten Larvenstadiums wieder zu und sinkt in der zweiten Hälfte dieses Stadiums bis zum Ende der Larvenentwicklung rasch ab. 3. Die Veränderungen proteolytischer Aktivität des Mitteldarms stehen in enger Beziehung zum Körperwachstum und zur Nahrungsaufnahme. Demgegenüber muß die Beziehung zwischen der proteolytischen Aktivität und dem Darmtrockengewicht im Laufe der Larvenentwicklung nicht immer eindeutig sein. 4. Während der Metamorphose sinkt die proteolytisohe Aktivität des Mitteldarms zunächst sehr rasch ab (vom Anfang der Einspinnung der Larven an bis zur Entstehung der Puppe). Nach der Verpuppung kommt es noch zur weiteren Herabsetzung der Darmproteasenaktivität, die dann bis zum Ende der Puppenentwicklung auf einem sehr niedrigen Niveau stehenbleibt. 5. Der Vergleich zwischen der proteolytischen Aktivität, der in der Darmwand und der im Darminhalt enthaltenen Proteinmenge im Laufe der Metamorphose des Mitteldarms deutet auf eine enge Beziehung zwischen der Darmproteasenaktivität und den mit dem Umbau des Darmes in dieser Periode verbundenen Vorgängen hin. Es scheint, daß die Darmproteasen an der Zerstörung des larvalen Darmes bedeutend beteiligt sind.

Summary 1. The proteolytic activity of Galleria mellonella midgut was followed in the course of the penultimate and last larval instars and during the pupal instar. 2. The activity increases in the penultimate instar and in the first half of the last larval instar; the increase is temporarily interrupted during the interjacent moulting period. In the second half of the latter instar, the activity continuously decreases. 3. These changes in proteolytic activity seem to be in a close relation to body growth and food consumption. They do not always correlate, however, with alterations of the midgut dry matter. 4. During metamorphosis of the midgut, which commences when the last instar larvae begin to spin their cocoons, the proteolytic activity enormously decreases. The initial rapid decrease slows down after the larval-pupal ecdysis and later levels off; a low level of activity is maintained until the emergence of adults. 5. From comparing during metamorphosis the changes in proteolytic activity, in amount of midgut-wall proteins, and in quantity of proteins in the gut content it follows that there is a tight interrelationship between the proteolytic activity and the process of the midgut transformation. The midgut proteases appear to participate in breaking down and digesting the larval gut.
