Description: 〈span〉The northern Tibetan Plateau, north of the Qaidam Basin and south of the Hexi Corridor (China), consists of a series of WNW- to NW-trending elongated mountain ranges. Deciphering the time-space deformation pattern of these ranges is central to understanding the mechanism of plateau formation and to the controversial issue of whether Tibet has undergone progressive northward growth or synchronous growth since the India-Eurasia collision. Here, we report new constraints on the timing of accelerated uplift of the Tuolai Shan, one of the elongated mountain ranges in the northern Tibetan Plateau. New apatite fission-track data from an elevation transect in the Tuolai Shan provide a definitive tie to rapid cooling that began at 17–15 Ma. We attribute this rapid cooling to accelerated exhumation resulting from thrusting in the hanging wall of the Haiyuan fault in response to progressive northward growth of the plateau. Combining these fission-track data and the published geologic, sedimentological, and thermochronologic data from the northern Qilian Shan and Hexi Corridor, we propose a progressively north-northeastward growth model for the northernmost part of Tibet, suggesting that deformation in the inner Qilian Shan occurred synchronously in the middle Miocene, and subsequently, increasingly further north.〈/span〉

Description: The process and mechanisms of secondary hydrocarbon migration in the Tazhong uplift, Tarim Basin, were investigated based on the analysis of the regional structure and by integrating geologic, hydrodynamic, and geochemical parameters. Parameters successfully analyzed included the fluid potential, fluid properties, production outputs, and diamantane index. The results indicated that hydrocarbons migrated into the Tazhong uplift from the northern part of the Manjiaer depression through a series of injection points (IPs) during four orogenies, that is, the early Caledonian (510 Ma), the late Caledonian (439 Ma), the late Hercynian–Indosinian (290 Ma), and the Yanshanian–Himalayan (208 Ma). A total of six IPs were identified at the intersections of the northeast-trending faults and the northwest-trending flower strike faults. The hydrocarbons migrated from the IPs into traps along regional trends from northwest to southeast and from northeast to southwest. The hydrocarbon migration process and patterns determined the distribution of hydrocarbon properties and production rates in the Tazhong uplift. With increasing distance from the IPs, daily hydrocarbon production decreases, and the hydrocarbons become progressively heavier and display lower gas:oil ratios.

Description: Seven hydrocarbon reservoirs have been discovered to date in the Upper Ordovician of the Tazhong Area, a region in which hydrocarbon phase distribution is complex. In the present study, the genesis and controlling factors of the hydrocarbons with complex phase in the Tazhong Area were investigated on the basis of the geological and geochemical conditions required for the formation and distribution of hydrocarbon reservoirs, integrated with the source rock geochemistry, natural gas and oil properties, and oil and gas reservoir fluid tests PVT (i.e., pressure, volume, and temperature tests). The results indicate that hydrocarbon reservoir types in the Upper Ordovician of the Tazhong Area transition from unsaturated to saturated condensate-gas reservoirs from west to east and from condensate-gas reservoirs to unsaturated-oil reservoirs from north to south. The crude oil in the region originated primarily from the mixing of Lower–Middle Cambrian and Middle–Upper Ordovician source rocks, while the natural gas was sourced primarily from the cracking gas of Lower–Middle Cambrian crude oil. This hydrocarbon-phase distribution was controlled primarily by temperature and pressure and has been affected by multiple periods of hydrocarbon accumulation and alteration. The high-quality Lower–Middle Cambrian reservoir–cap assemblage may be an important target for future exploration of natural gas in the Tazhong Area.

Description: Ongoing mouse studies of a proposed therapy for cocaine abuse based on viral gene transfer of butyrylcholinesterase (BChE) mutated for accelerated cocaine hydrolysis have yielded surprising effects on aggression. Further investigation has linked these effects to a reduction in circulating ghrelin, driven by BChE at levels ∼100-fold above normal. Tests...

Description: As an important mechanism for adaptation to heterogeneous environment, plastic responses of correlated traits to environmental alteration may also be genetically correlated, but less is known about the underlying genetic basis. We describe a statistical model for mapping specific quantitative trait loci (QTLs) that control the interrelationship of phenotypic plasticity between different traits. The model is constructed by a bivariate mixture setting, implemented with the EM algorithm to estimate the genetic effects of QTLs on correlative plastic response. We provide a series of procedure that test (1) how a QTL controls the phenotypic plasticity of a single trait; and (2) how the QTL determines the correlation of environment-induced changes of different traits. The model is readily extended to test how epistatic interactions among QTLs play a part in the correlations of different plastic traits. The model was validated through computer simulation and used to analyse multi-environment data of genetic mapping in winter wheat, showing its utilization in practice.

Description: The Syrian hamster embryo (SHE) cell transformation assay (pH 6.7) has utility in the assessment of potential chemical carcinogenicity (both genotoxic and non-genotoxic mechanisms of action). The assay uses morphological transformation as an end point and has a reported sensitivity of 87%, specificity of 83% and overall concordance of 85% with in vivo rodent bioassay data. However, the scoring of morphologically transformed SHE cells is subjective. We treated SHE cells grown on low-E reflective slides with benzo[ a ]pyrene, 3-methylcholanthrene, anthracene, N -nitroso- N -methylnitroguanidine, ortho-toluidine HCl, 2,4-diaminotoluene or D -mannitol for 7 days before fixation with methanol. Identified colonies were interrogated by acquiring a minimum of five infrared (IR) spectra per colony using attenuated total reflection Fourier-transform IR spectroscopy. Individual IR spectra were acquired over a spatial area of approximately 250 x 250 μm. Resultant data were analysed using Fisher’s linear discriminant analysis and feature histogram algorithms to extract classifying biomarkers of test agent-specific effects or transformation in SHE cells. Clustering of spectral points suggested co-segregation or discrimination of test agent categories based on mechanism of action. Towards transformation, unifying alterations were associated with alterations in the Amide I and Amide II peaks; these were consistently major classifying biomarkers for transformed versus non-transformed SHE cells. Our approach highlights a novel method towards objectively screening and classifying SHE cells, be it to ascertain test agent treatment based on mechanism of action or transformation.

Description: The ability of prions to infect some species and not others is determined by the transmission barrier. This unexplained phenomenon has led to the belief that certain species were not susceptible to transmissible spongiform encephalopathies (TSEs) and therefore represented negligible risk to human health if consumed. Using the protein misfolding cyclic amplification (PMCA) technique, we were able to overcome the species barrier in rabbits, which have been classified as TSE resistant for four decades. Rabbit brain homogenate, either unseeded or seeded in vitro with disease-related prions obtained from different species, was subjected to serial rounds of PMCA. De novo rabbit prions produced in vitro from unseeded material were tested for infectivity in rabbits, with one of three intracerebrally challenged animals succumbing to disease at 766 d and displaying all of the characteristics of a TSE, thereby demonstrating that leporids are not resistant to prion infection. Material from the brain of the clinically affected rabbit containing abnormal prion protein resulted in a 100% attack rate after its inoculation in transgenic mice overexpressing rabbit PrP. Transmissibility to rabbits (〉470 d) has been confirmed in 2 of 10 rabbits after intracerebral challenge. Despite rabbits no longer being able to be classified as resistant to TSEs, an outbreak of “mad rabbit disease” is unlikely.
