    Description: We present a spectroscopic analysis of the black hole binary Nova Muscae 1991 in quiescence using data obtained in 2009 with Magellan Echelette on the Magellan Clay telescope and in 2010 with IMACS on the Magellan Baade telescope at the Las Campanas Observatory. Emission from the disc is observed in Hα, Hβ and Ca  ii (8662). A prominent hotspot is observed in the Doppler maps of all three emission lines. The existence of this spot establishes ongoing mass transfer from the donor star in 2009–2010 and, given its absence in the 1993–1995 observations, demonstrates the presence of a variable hotspot in the system. We find the radial distance to the hotspot from the black hole to be consistent with the circularization radius. Our tomograms are suggestive of stream–disc overflow in the system. We also detect possible Ca  ii (8662) absorption from the donor star.

    Description: DNA mimic proteins are unique factors that control the DNA binding activity of target proteins by directly occupying their DNA binding sites. The extremely divergent amino acid sequences of the DNA mimics make these proteins hard to predict, and although they are likely to be ubiquitous, to date, only a few have been reported and functionally analyzed. Here we used a bioinformatic approach to look for potential DNA mimic proteins among previously reported protein structures. From ~14 candidates, we selected the Staphylococcus conserved hypothetical protein SSP0047, and used proteomic and structural approaches to show that it is a novel DNA mimic protein. In Staphylococcus aureus , we found that this protein acts as a uracil-DNA glycosylase inhibitor, and therefore named it S. aureus uracil-DNA glycosylase inhibitor (SAUGI). We also determined and analyzed the complex structure of SAUGI and S. aureus uracil-DNA glycosylase (SAUDG). Subsequent BIAcore studies further showed that SAUGI has a high binding affinity to both S. aureus and human UDG. The two uracil-DNA glycosylase inhibitors (UGI and p56) previously known to science were both found in Bacillus phages, and this is the first report of a bacterial DNA mimic that may regulate SAUDG’s functional roles in DNA repair and host defense.

    Description: Body fat distribution is a heritable trait and a well-established predictor of adverse metabolic outcomes, independent of overall adiposity. To increase our understanding of the genetic basis of body fat distribution and its molecular links to cardiometabolic traits, here we conduct genome-wide association meta-analyses of traits related to waist and hip circumferences in up to 224,459 individuals. We identify 49 loci (33 new) associated with waist-to-hip ratio adjusted for body mass index (BMI), and an additional 19 loci newly associated with related waist and hip circumference measures (P 〈 5 x 10(-8)). In total, 20 of the 49 waist-to-hip ratio adjusted for BMI loci show significant sexual dimorphism, 19 of which display a stronger effect in women. The identified loci were enriched for genes expressed in adipose tissue and for putative regulatory elements in adipocytes. Pathway analyses implicated adipogenesis, angiogenesis, transcriptional regulation and insulin resistance as processes affecting fat distribution, providing insight into potential pathophysiological     Description: Rett syndrome (RTT) is a neurodevelopmental disorder caused primarily by mutations of the X-linked MECP2 gene. Although the loss of MeCP2 function affects many neural systems, impairments of catecholaminergic function have been hypothesized to underlie several of the cardinal behavioral deficits of RTT patients and Mecp2-deficient mice. Although recent Mecp2 reactivation studies indicate that RTT may be a reversible condition, it remains unclear whether specifically preserving Mecp2 function within a specific system will be sufficient to convey beneficial effects. Here, we test whether the selective preservation of Mecp2 within catecholaminergic cells will improve the phenotype of Mecp2-deficient mice. Our results show that this targeted preservation of Mecp2 significantly improves the lifespan, phenotypic severity and cortical epileptiform discharge activity of both male and female Mecp2-deficient mice. Further, we found that the catecholaminergic preservation of Mecp2 also improves the ambulatory rate, rearing activity, motor coordination, anxiety and nest-building performances of Mecp2-deficient mice of each gender. Interestingly, our results also revealed a gender-specific improvement, as specific cortical and hippocampal electroencephalographic abnormalities were significantly improved in male, but not female, rescue mice. Collectively, these results support the role of the catecholaminergic system in the pathogenesis of RTT and provide proof-of-principle that restoring MeCP2 function within this specific system could represent a treatment strategy for RTT.

    Notes: Densely sintered Mn–Zn ferrites with grain size varying between 1 and 6 μm were obtained. Then the effect of grain size was investigated in detail by power loss analysis and scanning electron microscopy examination of microstructure. It is found that the power loss is seen to be lowered from 260 to 120 kW/m3 at 500 kHz, 50 m T and 90 °C with grain size decreased in the order of 3 μm. It is assumed that the residual loss from domain wall resonance is a dominant factor of power loss from 500 kHz to 1 MHz. Therefore we conclude that Mn–Zn ferrite display a grain size dependent power loss with transition at grain size about 3 μm due to the absence of intragranular domain wall and these fine core materials can be prepared by low sintering temperature and the optimum control of atmosphere containing 0.5% O2. © 1996 American Institute of Physics.

    Description: In the practice of vortex flow controls, the most important factor is that the persistency and obstinacy of a concentrated vortex depend on its stability and dissipation. In this paper, the modern nonlinear stability theory for circulation-preserving flows is summarized, and the dissipation for general viscous flows is analyzed in terms of the evolution of total enstrophy. These analyses provide a theoretical base for understanding relevant physics of vortex flows, and lead to some guiding principles and methods for their controls. Case studies taken from various theoretical and/or experimental works of vortex controls, due to the present authors as well as others, confirm the feasibility of the recommended principles and methods.

    Description: A new instrument has been developed for direct measurement of wall shear stress. This instrument is simple and symmetric in design with small moving mass and no internal friction. Features employed in the design of this instrument eliminate most of the difficulties associated with the traditional floating element balances. Vibration problems associated with the floating element skin friction balances have been found to be minimized by the design features and optional damping provided. The unique design of this instrument eliminates or reduces the errors associated with conventional floating-element devices: such as errors due to gaps, pressure gradient, acceleration, heat transfer and temperature change. The instrument is equipped with various sensing systems and the output signal is a linear function of the wall shear stress. Measurement made in three different tunnels show good agreement with theory and data obtained by the floating element devices.
