    Description: Mafic granulites of the Dunhuang block, eastern Tarim Craton, record high-pressure granulite-facies metamorphism followed by a medium- to low-pressure granulite-facies metamorphic overprint, suggesting a clockwise P–T path and implying an environment of collisional orogenesis. Zircon U–Pb ages of two samples are 1834 ± 12 Ma and 1842 ± 5 Ma. Mineral inclusions in zircon indicate that these ages record the high-pressure granulite-facies event, suggesting that the Tarim Craton may be related to the c . 1.85 Ga assembly of the Columbia supercontinent. Similar c . 1.85 Ga metamorphism in both the Dunhuang block and the Alxa block of the North China Craton supports their correlation across the Altyn Tagh fault.     Description: As high-strength cotton fibers are critical components of high quality cotton, developing cotton cultivars with high-strength fibers as well as high yield is a top priority for cotton development. Recently, chromosome segment substitution lines (CSSLs) have been developed from high-yield Upland cotton ( Gossypium hirsutum ) crossed with high-quality Sea Island cotton ( G. barbadense ). Here, we constructed a CSSL population by crossing CCRI45, a high-yield Upland cotton cultivar, with Hai1, a Sea Island cotton cultivar with superior fiber quality. We then selected two CSSLs with significantly higher fiber strength than CCRI45 (MBI7747 and MBI7561), and one CSSL with lower fiber strength than CCRI45 (MBI7285), for further analysis. We sequenced all four transcriptomes at four different time points postanthesis, and clustered the 44,678 identified genes by function. We identified 2200 common differentially-expressed genes (DEGs): those that were found in both high quality CSSLs (MBI7747 and MBI7561), but not in the low quality CSSL (MBI7285). Many of these genes were associated with various metabolic pathways that affect fiber strength. Upregulated DEGs were associated with polysaccharide metabolic regulation, single-organism localization, cell wall organization, and biogenesis, while the downregulated DEGs were associated with microtubule regulation, the cellular response to stress, and the cell cycle. Further analyses indicated that three genes, XLOC_036333 [mannosyl-oligosaccharide-α-mannosidase ( MNS1 )], XLOC_029945 ( FLA8 ), and XLOC_075372 ( snakin-1 ), were potentially important for the regulation of cotton fiber strength. Our results suggest that these genes may be good candidates for future investigation of the molecular mechanisms of fiber strength formation and for the improvement of cotton fiber quality through molecular breeding.

    Description: A cloud pattern matching technique is applied to polar mesospheric cloud (PMC) images taken by the Cloud Imaging and Particle Size instrument (CIPS) to infer the wind velocities in the mesopause region. CIPS measurements are analyzed to detect patterns that repeat from one orbit to the next, but are displaced in location; the displacement provides a measure of the wind velocity. Pattern matching is achieved by re-sampling the CIPS data to longitude and latitude grids with the grid-box size forced at ~5km in both directions. The correlated patterns are searched within a geographic region referred to as a " frame" of ~500 km in longitude × 400 km in latitude. The histograms of the derived velocities indicate that easterly winds prevail, with a mean zonal wind of −20 ~ −15 m/s. Mean meridional winds are overall small, but in late summer the histogram indicated a poleward wind of ~20-30m/s. The variability of CIPS cloud albedo on consecutive orbits is also examined at fixed geolocations. The statistical results suggest that ~86% of pairs underwent mean cloud albedo variation of 〈 50% on consecutive orbits, suggesting a moderate change. It is also found that the correlation of the cloud structures between two consecutive orbits at a fixed location is generally poor. These findings suggest that cloud patterns are subject to wind advection, but the cloud patches are more extended in size than the movement that occurs. Cloud voids are found to be more likely to remain at the same geolocations.

    Description: The rpoS mRNA, which encodes the master regulator S of general stress response, requires Hfq-facilitated base pairing with DsrA small RNA for efficient translation at low temperatures. It has recently been proposed that one mechanism underlying Hfq action is to bridge a transient ternary complex by simultaneously binding to rpoS and DsrA. However, no structural evidence of Hfq simultaneously bound to different RNAs has been reported. We detected simultaneous binding of Hfq to rpoS and DsrA fragments. Crystal structures of AU6A•Hfq•A7 and Hfq•A7 complexes were resolved using 1.8- and 1.9-Å resolution, respectively. Ternary complex has been further verified in solution by NMR. In vivo , activation of rpoS translation requires intact Hfq, which is capable of bridging rpoS and DsrA simultaneously into ternary complex. This ternary complex possibly corresponds to a meta-stable transition state in Hfq-facilitated small RNA–mRNA annealing process.

    Description: Abstract High‐speed solar wind streams that originate from coronal holes play an important role in space weather disturbances, especially during the declining phase of the solar cycle. Space weather forecasters attempt to find good coronal hole indices that can be used to predict high‐speed streams days in advance. Several indices related to the coronal hole area, brightness, or magnetic field expansion factor have been reported in the literature. Empirical solar wind forecast models have been developed and used in operational service by several organizations by constructing prediction functions that relate the coronal hole index to the solar wind speed. In this paper, we present a new empirical modeling method and test its validity by comparing it with a previously reported method when applied to different coronal hole indices. In total, six empirical models are tested for a long period of time (2011–2018), with a 27‐day persistence model as a comparison benchmark. The results show that while all these empirical models can capture the temporal patterns of the solar wind observations well, the new modeling method and utilization of a composite coronal hole index PCH as an input parameter indeed improves the forecast accuracy. The high‐speed streams can be predicted approximately 3 days in advance, with a probability of detection of 0.78, a positive predictive value of 0.73, and a threat score of 0.61. The uncertainty of the high‐speed stream arrival time is approximately 1 day and the uncertainty of the peak speed is approximately 80 km/s.

    Description: Mutations in Fused in sarcoma (FUS) gene cause a subset of familial amyotrophic lateral sclerosis (ALS), a fatal motor neuron degenerative disease. Wild-type FUS is largely localized in the nucleus, but mutant FUS accumulates in the cytoplasm and forms inclusions. It is unclear whether FUS depletion from the nucleus or FUS inclusions in the cytoplasm triggers motor neuron degeneration. In this study, we revealed that the nuclear and cytoplasmic FUS proteins form distinct local distribution patterns. The nuclear FUS forms oligomers and appears granular under confocal microscope. In contrast, the cytoplasmic FUS forms inclusions with no oligomers detected. These patterns are determined by the subcellular localization of FUS, regardless of wild-type or mutant protein. Moreover, mutant FUS remained or re-directed in the nucleus can oligomerize and behave similarly to the wild-type FUS protein. We further found that nuclear RNAs are critical to its oligomerization. Interestingly, the formation of cytoplasmic FUS inclusions is also dependent on RNA binding. Since the ALS mutations disrupt the nuclear localization sequence, mutant FUS is likely retained in the cytoplasm after translation and interacts with cytoplasmic RNAs. We therefore propose that local RNA molecules interacting with the FUS protein in different subcellular compartments play a fundamental role in determining FUS protein architecture and function.

    Description: The human Y box-binding protein-1 (YB-1) is a deoxyribonucleic acid (DNA)/ribonucleic acid (RNA)-binding protein with pleiotropic functions. Besides its roles in the regulation of transcription and translation, several recent studies indicate that YB-1 is a spliceosome-associated protein and is involved in alternative splicing, but the underlying mechanism has remained elusive. Here, we define both CAUC and CACC as high-affinity binding motifs for YB-1 by systematic evolution of ligands by exponential enrichment (SELEX) and demonstrate that these newly defined motifs function as splicing enhancers. Interestingly, on the endogenous CD44 gene, YB-1 appears to mediate a network interaction to activate exon v5 inclusion via multiple CAUC motifs in both the alternative exon and its upstream polypyrimidine tract. We provide evidence that YB-1 activates splicing by facilitating the recruitment of U2AF65 to weak polypyrimidine tracts through direct protein–protein interactions. Together, these findings suggest a vital role of YB-1 in activating a subset of weak 3' splice sites in mammalian cells.
