    Description: We here report on specific chemical and microbial compositions observed at the Archaean hydrothermal site in the Southern Mariana backarc spreading center, which produces two remarkably different hydrothermal fluids. One was black smoker hydrothermal fluid at 340 °C containing a low concentration of methane with a relatively high carbon isotope ratio (d13C of methane relative to VPDB), -7.8 per mil, indicating that methane originated from a magmatic source; thus, this is a fairly ordinary hydrothermal fluid for basalt-hosted hydrothermal activity. In contrast, the other fluid was clear smoker hydrothermal fluid at 117 °C containing a high concentration of methane with a very low carbon isotope ratio (-49.7 per mil). The host rock of the Archaean hydrothermal site is basalt, and therefore Fischer-Tropsch-type reactions resulting from serpentinization of mantle rocks are not feasible as a source for the "light" carbon in methane. Additionally, the carbon isotope ratio of carbon dioxide demonstrated that the sources of CO2 and CH4 were not organic materials. The remaining possibility, biogenic methane, was confirmed by cultivation of hyperthermophilic hydrogen-oxidizing methanogens from both rocks covering the vent of clear smoker hydrothermal fluids and in situ cultivation systems. Although the dominance and abundance of methanogens were very low in this ecosystem, the number was consistent with the relationship between hydrogen concentration and methanogen abundance (Takai et al., 2015). These results suggest that phase separation led to concentration of hydrogen and subsequent persistence of a hyperthermophilic subsurface lithoautotrophic microbial ecosystem in both serpentine-hosted and basalt-hosted hydrothermal systems. This type of ecosystem may occur in similar settings elsewhere on modern earth; in addition, similar communities may have existed in other types of deep-sea hydrothermal systems in the geological past.

    Description: We have measured the 3He/4He and 4He/20Ne ratios and chemical compositions of gases exsolved from deep-sea sediments at two sites (798 and 799) in the Japan Sea. The 3He/4He and 4He/20Ne ratios vary from 0.642 Ratm (where Ratm is the atmospheric 3He/4He ratio of 1.393*10**-6) to 0.840 Ratm, and from 0.41 to 4.5, respectively. Helium in the samples can be explained by the mixing between atmospheric helium dissolved in bottom water of the Japan Sea and crustal helium in the sediment. The sedimentary helium is enriched in mantle-derived 3He compared with those from the Japan Trench and the Nankai Trough. This suggests that the basement of the Japan Sea has relatively large remnants of mantle-derived helium compared with that of the Pacific. Major chemical compositions of the samples are methane and nitrogen. There is a positive correlation between methane content and helium content corrected for air component. Based on the 3He/4He-Sum C/3He diagram, the major part of methane can be attributed to crustal and/or organic origin.

    Notes: After excavation using a portable submarine driller near deep-sea hydrothermal vents in the Suiyo Seamount, Izu-Bonin Arc, microbial diversity was examined in samples collected from inside the boreholes using an in situ growth chamber called a vent catheter. This instrument, which we devised for this study, consists of a heat-tolerant pipe tipped with a titanium mesh entrapment capsule that is packed with sterilized inorganic porous grains, which serve as an adhesion substrate. After this instrument was deployed inside each of the boreholes, as well as a natural vent, for 3–10 days in the vicinity of hot vent fluids (maxima: 156–305°C), DNA was extracted from the adhesion grains, 16S rDNA was amplified, and randomly selected clones were sequenced. In phylogenetic analysis of more than 120 clones, several novel phylotypes were detected within the ε-Proteobacteria, photosynthetic bacteria (PSB)-related α-Proteobacteria, and Euryarchaeota clusters. Members of ε-Proteobacteria were frequently encountered. Half of these were classified between two known groups, Corre's B and D. The other half of the clones were assigned to new groups, SSSV-BE1 and SSSV-BE2 (Suiyo Seamount sub-vent origin, Bacteria domain, ε-Proteobacteria, groups 1 and 2). From this hydrothermal vent field, we detected a novel lineage within the PSB cluster, SSNV-BA1 (Suiyo Seamount natural vent origin, Bacteria domain, α-Proteobacteria, group 1), which is closely related to Rhodopila globiformis isolated from a hot spring. A number of archaeal clones were also detected from the borehole samples. These clones formed a novel monophyletic clade, SSSV-AE1 (Suiyo Seamount sub-vent origin, Archaea domain, Euryarchaeota, group 1), approximately between methanogenic hyperthermophilic members of Methanococcales and environmental clone members of DHVE Group II. Thus, this hydrothermal vent environment appears to be a noteworthy microbial and genetic resource. It is also noteworthy that some of the findings presented here were made possible by the application of the in situ growth chamber into the hot fluids deep inside the boreholes.

    Notes: Phase-separation and -segregation (boiling/distillation of subseafloor hydrothermal fluids) represent the primary mechanisms causing intra-field variations in vent fluid compositions. To determine whether this geochemical process affects the formation of microbial communities, we examined the microbial communities at three different vent sites located within a few tens meters of one another. In addition to chimney structures, colonization devices capturing subseafloor communities entrained by the vent fluids were studied, using culture-dependent and -independent methods. Microbiological analyses demonstrated the occurrence of distinctive microbial communities in each of the hydrothermal niches. Within a chimney structure, there was a transition from a mixed community of mesophiles and thermophiles in the exterior parts to thermophiles in the interior. Beside the transition within a chimney structure, intra-field variations in microbial communities in vent fluids were apparent. Geochemical analysis demonstrated that different vent fluids have distinctive end-member compositions as a consequence of subseafloor phase-separation and -segregation, which were designated gas-depleted, normal and gas-enriched fluids. In comparison to gas-depleted and normal fluids, gas-enriched fluids harbored more abundant chemolithoautotrophs with gaseous component-dependent energy metabolism, such as hydrogenotrophic methanogenesis. Subseafloor phase-separation and -segregation may play a key role in supplying energy and carbon sources to vent-associated chemolithoautotrophs and subvent microbial communities.
