    Notes: Vibrational state distributions of the A 2Σ+ excited state of HCl+ were measured by dispersed fluorescence following resonant photoionization. Autoionization of levels excited at the NeI resonance line strongly influence the vibrational branching ratios of the A 2Σ+ state although not in accord with the propensity rule expected for vibrational autoionization. Other measurements utilizing total fluorescence yields and synchrotron radiation confirm the presence of competing dissociation channels for autoionizing Rydberg states converging to the A 2Σ+ limit. These results are discussed in terms of the multichannel interactions responsible for determining the observed ion and fragment product distributions.

    Notes: The domain of validity of the weak-phase-object (WPO) approximation is evaluated for high-energy electrons (100 keV, 500 keV and 1 MeV) scattered by crystalline biological macromolecules. Cytochrome b5 is used as an example in which calculated dynamical diffraction intensities are used to simulate observed diffraction intensities, which are then compared with intensities calculated by the weak-phase-object approximation. Three criteria of validity are used, namely the crystallographic residual (R value), the interpretability of difference Patterson maps, and the results of phasing by the heavy-atom isomorphous replacement method. The present calculations indicate that the error associated with the WPO approximation is quite acceptable up to a specimen thickness of 200 Å for 100 keV electrons, which is two to four times the thickness limit for crystalline organic structures with much smaller unit-cell dimensions. An equally acceptable thickness limit at 500keV and 1 MeV is about 300-350 Å.

    Notes: The conditions under which direct phasing methods might be used to determine the crystallographic phases from electron diffraction intensity data are investigated through numerical calculations for two organic crystals. The Cowley-Moodie multislice formulation of dynamical diffraction theory is used to calculate the diffraction intensity data for various crystal thicknesses, at electron energies of 100 keV and 1.0 MeV. The direct phasing method is found to give generally correct phases up to a crystal thickness of about 75 Å at 100 keV. The use of higher electron energy (i.e. 1.0 MeV) produces a significant improvement in the success of the direct phasing approach at crystal thicknesses greater than 75 Å.

    Notes: In order to fast and economically purify MTGase from Streptoverticillium ladakanum, a stepwise elution method was developed and compared with linear gradient elution method. MTGase was purified to electrophoretical homogeneity by using CM Sepharose CL-6B and Blue Sepharose Fast Flow chromatographies by linear gradient or stepwise methods. The recovery of MTGase by linear gradient and stepwise methods were 68.4% and 81.0%, respectively. The optimal temperature and pH were 40 °C and 5.5, respectively. It was stable at pH 5.0 to 7.0 and had a rate constant (KD) of 6.21 °o 10-5 min-1 for thermal inactivation at 45 °C. The purified MTGase was activated by K+ Na+, Ca2+, Mn2+, and Mg2+, not affected by Fe3+, EDTA, but inhibited by Cu2+, Zn2+, Hg2+, Ni2+, Co2+, Cd2+, PCMB, NEM, IAA, and PMSF. A simple stepwise method was developed for the purification of MTGase from S. ladakanum.

    Notes: : The properties of surimi gels from threadfin-bream and pollack surimi set at 30 °C or 45 °C with microbial transglutaminase (MTGase) from Streptoverticillium ladakanum were determined. The optimal amounts of MTGase and setting conditions were: 0.3 unit/g surimi either at 30 °C for 90 min or at 45 °C for 20 min for threadfin-bream, and 0.2 unit/g surimi at 30 °C for 60 min for pollack. The strength of golden threadfin-bream surimi gels with 0.35 unit MTGase set at 30 °C for 90 min or 45 °C for 20 min was 3400 g cm, almost 3-fold of the control. SDS-PAGE analyses indicated that inter- and/or intramolecular cross-linking formed in the myosin heavy chain of MTGase-containing surimi gels.

    Notes: The application of NADPH-sulfite reductase, produced from Escherichia coli, on mackerel surimi was investigated. The activity of this reductase was very stable. There was about 70%, 80%, and 90% activity left even after 8–wk storage at 25 °C and 12–wk storage at 4 or -30 °C, respectively. Increase in mackerel-surimi gel properties containing reductase was consistent with the amounts of reactive sulfhydryl groups detected. SDS-PAGE indicated that NADPH-sulfite reductase could reconstitute myosin heavy-chain and increase soluble actomyosin. From the data obtained, E. coli NADPH-sulfite reductase effectively improved the gel properties of mackerel surimi.

    Notes: A combination of microbial transglutaminase (MTGase), reducing agent and protease inhibitor was employed to improve the quality of underutilized fish surimi. SDS-PAGE indicated that cross-linking of myosin heavy chain (MHC) occurred in MTGase-contained samples, while MHC of samples without MTGase disappeared after 120 min incubation at 45°C. Although the gel-forming ability increased with MTGase added up to 0.6 unit/g, it was still too low to be commercially acceptable. However, the combined use of 0.1% NaHSO3, 0.01 mM transepoxysuccinyl-L-leucylamido-(4-guanidino)butane (E-64) and 0.35 units MTGase/g substantially improved the quality of hairtail surimi. Based on this result, the combined use of E-64, MTGase and NaHSO3 seemed to be a better way to improve gel-forming ability of hairtail surimi.
