    Notes: Nanocrystalline diamond films (NDFs) were deposited on mirror-polished silicon (100)substrates using a graphite-grid assisted hot filament chemical vapor deposition (HFCVD)technique. The evidence of nanocrystallinity, smoothness and purity was obtained by characterizingthe sample with various advanced analyses. A graphite-grid was used as the DC electrode, whichwas pre-coated by diamond films and could emit electron when a negative bias was applied to theunder electrode. The results show that the film consists of nanocrystalline diamond grains with sizesof about 7-15nm. The Raman spectroscopy, XRD pattern, HR-TEM image and SAED pattern of thefilms indicate the presence of nanocrystalline diamond. Surface roughness is measured as Ra〈20nmby the profilometry scans. A large quantity of electrons emission from the graphite-grid and positiveions bombardment to the graphite-grid results in an enormous enhancement of the generation ofdiamond nuclei. Density of a diamond nuclei as high as 1010~1011/cm2 can be attained with thismethod. NDFs can be deposited on mirror-polished Si substrate surfaces without damaging surfacepretreatment for nucleation enhancement. These ultra-smooth films will display excellentperformances, which make them the best candidates for semiconductor and MEMS applications

    Notes: The Si–H stretching–bending overtones in SiHCl3 were investigated employing theoretically calculated potential energy surfaces (PES) and dipole moment surfaces (DMS). The coupled cluster method CCSD(T) was utilized to generate both one-dimensional (1D) and three-dimensional (3D) surfaces. An empirical 3D PES was also taken into consideration. The computed energy levels and band intensities agree reasonably well with observation for most of the bands. Comparison of CCSD(T) and density functional results for the very weak 2ν1 band shows that it is essential to calculate the DMS at a high level of quantum-chemical theory when cancellation of linear and quadratic contributions to the DMS is significant. The 3D ab initio PES yields more accurate band intensities than the empirical PES and therefore appears to be more realistic.© 2002 American Institute of Physics.

    Notes: The infrared spectra of PH3 molecule were recorded on a Bruker IFS 120HR Fourier transform spectrometer from 4000 to 9500 cm−1. The P–H stretching vibrational frequencies and intensities were derived from the experimental data. The Morse oscillator parameters De and α in the anharmonically coupled anharmonic oscillator local mode model were determined by the least-squares fitting with the observed vibrational band centers. The ab initio three-dimensional P–H stretching dipole moment surfaces were calculated by the density functional theory method. The dipole moment vectors were projected to three kinds of molecule-fixed reference systems. The corresponding dipole moment components were fitted to polynomial functions in terms of the P–H bond length displacements with the molecular symmetry taken into account. The absolute band intensities were obtained and then compared with the experimental data. The results showed that a proposed improved bond dipole model can predict the absolute band intensities within a factor of 2 for most of the observed transitions, indicating a reasonably good agreement. © 2001 American Institute of Physics.

    Notes: Band strengths of the Fermi resonance polyads involving C–H stretching and bending vibrations for the CHBr3 molecule are calculated with a one-dimensional dipole function and a two-dimensional dipole surface. These are obtained by the ab initio density functional method. Both treatments reproduce the interpolyad band strengths quite well, while the two-dimensional dipole moment surface is superior to the one-dimensional one in modeling the intrapolyad band strength pattern, indicating the importance of the bending vibrations in dipole moment surface and intensities calculations. The different behavior of interpolyad band strengths of CHBr3 compared to CHCl3, with an intensity anomaly occurring in the latter case, is illustrated by "dipole–weighted" overlap integrals. © 2001 American Institute of Physics.

    Notes: Relative absorption intensities of the Fermi resonance polyads of isolated CH chromophore for the CHCl3 molecule were calculated with one-dimensional and two-dimensional dipole moment surfaces, which are obtained by the ab initio density functional method. The calculation showed an unusual strong absorption at the second Fermi resonance polyad, which agrees well with the experimental data. Such an intensities anomaly can be attributed to the nonlinearity of the dipole moment surface in the vicinity of the equilibrium configuration. By taking advantage of the two-dimensional dipole moment surface where both CH stretching and bending vibrations are taken into consideration, the ν1 and 2ν4 bands which constitute the first Fermi resonance polyad and have little wave function mixture, are found to be of almost equal intensity. Such a calculation agrees with observations. © 2000 American Institute of Physics.

    Notes: Abstract Among many interesting and sophisticated mechanisms used by bacterial pathogens to subvert eukaryotic hosts is a class of specialized protein secretion systems (known as type III protein secretion systems) that deliver bacterial virulence proteins directly into the host cell. Recent studies have revealed four important features of these secretion systems. First, they are widespread among plant and animal bacterial pathogens, and mutations affecting type III protein secretion often eliminate bacterial virulence completely. Second, at least eight type III secretion components share sequence similarities with those of the flagellar assembly machinery and flagellum-like structures are associated with type III secretion, raising the possibility that these secretion systems are derived from the presumably more ancient flagellar assembly apparatus. Third, type III secretion is activated in vivo upon contact with host cells. Fourth, the type III secretion mechanism is Sec-independent and the effector proteins may possess mRNA-based targeting signals. This review highlights the similarities and differences among type III secretion systems of selected model plant and animal pathogenic bacteria.

    Notes: Pili are required for protein and/or DNA transfer from bacteria to recipient plant or bacterial cells, based on genetic evidence. However, it has never been shown directly that the effector proteins or DNA are localized along or inside the pili in situ. Failure to visualize an association of effector proteins/DNA with pili is the central issue in the debate regarding the exact function of pili in protein and DNA transfer. In this study, a newly developed in situ immunogold labelling procedure enabled visualization of the specific localization of type III effector proteins of Erwinia amylovora and Pseudomonas syringae pv. tomato along the Hrp pilus, but not along the flagellum or randomly in the intercellular space. In contrast, PelE, a pectate lyase secreted via the type II protein secretion system, was not associated with the Hrp pilus. These results provide direct evidence that type III secretion occurs only at the site of Hrp pilus assembly and that the Hrp pilus guides the transfer of effector proteins outside the bacterial cell, favouring the ‘conduit/guiding filament’ model.

    Notes: An 8551-bp plasmid, pFQ11, from Frankia alni strain CpI1 was sequenced. Its sequence was found to be very similar to that presented for pFQ31 from strain ArI3. Six potential protein-encoding open reading frames (ORFs) were identified, and transcriptional activity was shown within four of those regions of the plasmid by reverse transcriptase-polymerase chain reaction (RT-PCR) analysis. An earlier study reported that ORF EF of pFQ31, which is nearly identical to the 3′ 45% of ORF1 of pFQ11, is significantly similar to RepF. We found no such similarity. ORF2 and ORF3 predict products that are similar to a repressor protein and a partition protein, respectively. We found inverted repeats within and covering the start codon of ORF3; palindromic sequences and direct repeats between ORF3 and ORF4; and 3′ from ORF3, an AT-rich sequence that extensively overlaps the promoter region of a uvrB homolog in strain ArI3.
