    Notes: An active oolitic sand wave was monitored for a period of 37 days in order to address the relationship between the direction and strength of tidal currents and the resultant geometry, and amount and direction of migration of bedforms in carbonate sands. The study area is situated in a tidal channel near Lee Stocking Island (Exumas, Bahamas) containing an estimated 5.5 to 6 × 105 m3 of mobile oolitic sand. Tidal ranges within the inlet are microtidal and the maximum current velocity at the studied site is 0.6 m s−1. At least 300–400 m3 of mostly oolitic sand are formed within, or brought into, the channel area every year. The tidal inlet is subdivided into an ocean-orientated segment, in which sand waves are shaped by both flood and ebb tides, and a platform-orientated segment, where sand waves are mainly shaped by flood tides. The studied sand wave lies on the platformward flood-tide dominated segment in a water depth of 3.5.4.5 m. During the 37 days of observation, the oolitic and bioclastic sand wave migrated 4 m in the direction of the dominant flood current. The increments of migration were directly related to the strength of the tide.During each tidal cycle, bedforms formed depending on the strength of the tidal current, tidal range and their location on the sand wave. During flood tides, a steep lee and a gentle stoss side formed and current ripples and small dunes developed on the crest of the sand wave, while the trough developed only ripples. The average lee slope of the sand wave is 24.2°, and therefore steeper than typical siliciclastic sand waves. During ebb tides, portions of the crest are eroded creating a convex upward ebb stoss side, covered with climbing cuspate and linguoid ripples and composite dunes. The area between the ebb-lee side and the trough is covered with fan systems, sinuous ripples and dunes.The migration of all bedforms deviated to a variable degree from the main current direction, reflecting complex flow patterns in the tidal inlet. Small bedforms displayed the largest deviation, migrating at an angle of up to 90° and more to the dominant current direction during spring tides.

    Notes: We have cloned the H1 histone gene (hhoA) of Aspergillus nidulans. This single-copy gene codes for a typical linker histone with one central globular domain. The open reading frame is interrupted by six introns. The position of the first intron is identical to that of introns found in some plant histones. An H1–GFP fusion shows exclusive nuclear localization, whereas chromosomal localization can be observed during condensation at mitosis. Surprisingly, the deletion of hhoA results in no obvious phenotype. The nucleosomal repeat length and susceptibility to micrococcal nuclease digestion of A. nidulans chromatin are unchanged in the deleted strain. The nucleosomal organization of a number of promoters, including in particular the strictly regulated niiA-niaD bidirectional promoter is not affected.

    Notes: The potential of several alternative genetic engineering based strategies in order to accelerate Saccharomyces cerevisiae autolysis for wine production has been studied. Both constitutively autophagic and defective in autophagy strains have been studied. Although both alternatives lead to impaired survival under starvation conditions, only constitutively autophagic strains, carrying a multicopy plasmid with the csc1-1 allele under the control of the TDH3 promoter, undergo accelerated autolysis in the experimental conditions tested. Fermentation performance is impaired in the autolytic strains, but industrial strains carrying the above-mentioned construction are still able to complete second fermentation of a model base wine. We suggest the construction of industrial yeasts showing a constitutive autophagic phenotype as a way to obtain second fermentation yeast strains undergoing accelerated autolysis.

    Notes: We have investigated the effect of disruption of the bgl1-(β-glucosidase l-encoding) gene of Trichoderma reesei on the formation of other β-glucosidase activities and on the induction of cellulases. To this end the bgl1 locus was disrupted by insertion of the Aspergillus nidulans amdS (acetamidase-encoding) gene. The bgl1-disrupted strain did not produce the 75kDa extracellular β-glucosidase on cellulose or lactose, but still formed β-glucosidase activity on glucose, cellobiose, xylan or β-1,3-glucan, suggesting that the enzyme(s) exhibiting this β-glucosidase activity is (are) not encoded by bgl1. The cellulose-inducer sophorose induced the bgl1-encoded β-glucosidase, whereas the remaining β-glucosidase activity was induced by methyl-β-D-glucoside. The bgl1-gene product was mainly secreted into the medium, whereas the other β-glucosidase activity was mainly associated with the cells. A bgl1-multicopy strain formed higher amounts of cellulases than the parent strain. Nonsaturating concentrations of sophorose efficiently induced cellobiohydrolase I formation in the bgl1-multicopy strain, but less efficiently in the bgl1-disrupted strain. The multicopy strain and the parent strain were comparably efficient at saturating sophorose concentrations. The β-glucosidase inhibitor nojirimycin strongly inhibited induction in all strains. These data suggest that the bgl1-encoded β-glucosidase is not identical to the plasma-membrane-bound, constitutive, methyl-β-glucoside inducible β-glucosidase, but represents an extracellular cellulose-induced enzyme. Both enzymes contribute to rapid induction of cellulases by modifying the inducer sophorose.

    Notes: Abstract Mid-infrared (MIR) and near-infrared (NIR) spectroscopy were used to determine water in lubricating oils with high additive contents that introduce large errors in determinations by the Karl-Fischer and hydride methods. MIR spectra were obtained in the attenuated total reflectance (ATR) mode and exhibited water specific band absorption in the region 3100–3700cm−1, which facilitated calibration. Multivariate (partial least-squares regression, PLSR) and univariate calibration (based on peak height and band area as independent variables) were tested. Both led to errors of prediction less than 5%. NIRS determinations rely on absorbance and first-derivative spectra, in addition to two different types of multivariate calibration,viz. inverse multiple linear regression (MLR) and partial least-squares regression (PLSR). Both approaches gave similar results, with errors of prediction less than 2%. For none of the proposed approaches any sample pretreatment for recording spectra is required.

    Notes: The thermodynamic compatibility of the diffuse layer theory with the chemical potential of ionic species in the solution bulk is introduced. From the electrocapillary equation, a compatibility equation, which relates the potential drop across the diffuse layer to the chemical potential of the ionic species in the solution bulk, is deduced for a binary electrolyte. When the Gouy-Chapman (GC) theory is introduced in the compatibility equation, the latter only becomes an identity if the ionic activity is taken as the ionic concentration, as expected. A compatible theory of the diffuse layer which has GC as the limit case at dilute solutions and fulfills the compatibility equation for whatever physical description of the ionic activity coefficients is stated. The significance of the compatible theory is that the electrode reaction rates calculated by means of the electrode kinetic equation with the Frumkin correction using this theory differ from those calculated using GC. In order to test both theories, we have studied the electroreduction of peroxodisulfate at an amalgamated copper rotating disk electrode. The experimental results fit our compatible theory better than GC does.

    Notes: Acute inflammation was induced in pigs using a single subcutaneous turpentine injection. The acute phase serum protein response was analyzed using crossed immunoelectrophoresis and immunodiffusion. The concentration of C reactive protein and haptoglobin increases 5-7 times 48 h after the injection, whereas the concentration of an α2-globulin, named pig major acute phase protein (pig-MAP), increases at least 15-fold. A molecular mass of 115 kDa for pig-MAP was determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. This protein did not crossreact with antisera to human hemopexin, ceruloplasmin, H-kininogen and complement factor C3. Albumin and α-lipoprotein were negative acute phase proteins because their concentration significantly decreased during inflammation. Finally, the concentration of α1-acid glycoprotein, fetuin, α1-protease inhibitor, transferrin and α2-macro-globulins, as well as total proteins, did not change significantly during inflammation.
