Notes: We have re-examined the analysis of centrifugal consolidation experiments for the determination of drained compressibility. Existing models for centrifugation of ceramic powder suspensions are prone to error, because they do not fully account for spatial variations of both the centrifugal acceleration and packing density. We present a new model that accounts for these two effects. We also derive dimensionless parameters that separately describe the error contributions that result from the heterogeneities of both the centrifugal acceleration and packing density. These dimensionless parameters make it no longer necessary to determine drained compressibility by the complicated method of nondestructively measuring the density profiles of centrifuge cakes and then substituting these profiles into a complex mathematical equation that is difficult to solve. Instead, one can evaluate drained compressibility by applying a correction factor to a simple experimental method of measuring the cake height during multispeed centrifugation. In turn, this procedure provides a valuable, new tool to characterize the effects of ceramic processing variables on their tendency to produce density gradients during powder consolidation.

Notes: Green and partially sintered compacts of alpha-Al2O3 powder were made by filtration of aqueous suspensions under three conditions: (i) electrostatic stabilization without any organic additive, (ii) strong flocculation near the isoelectric point without any organic additive, and (iii) weak flocculation by the use of maltodextrin or oxalic acid additives. We evaluated relationships between the macroscopic and interparticle mechanical behavior of these compacts using model correlations with measurements of diametral compression, ultrasonic velocity, and ultrasonic attenuation. Although type iii green specimens were less dense than type i, type iii exhibited significant increases in velocity, macroscopic Young's modulus, interparticle-contact stiffness, and diametral compressive strength, suggesting that the mechanism of stiffening/strengthening entailed interparticle bridging of maltodextrin or oxalic acid. These properties were significantly reduced upon heating type iii specimens to 500°C, suggesting that pyrolysis of surface-adsorbed maltodextrin and oxalic acid may have reduced the interparticle stiffness and strength. In contrast, negligible changes in these properties occurred upon heating type i specimens to the same temperature. Despite small increases in packing density, significant decreases in attenuation and significant increases in velocity, interparticle-contact stiffness, and Young's modulus occurred upon heating all specimens to greaterthan equal to700°C, suggesting the formation of interparticle necks by solid-state sintering.

Notes: Abstract The hypothesis that rates of litter turnover in mangroves are controlled by local geophysical processes such as tides has been studied at sites with mostly small tides (〈1 m) and minor crab consumption of leaf litter. Our study describes litter dynamics of three riverine mangrove sites (M1, M2, M3), inhabited by the mangrove crab Ucides occidentalis, located in a macrotidal (〉3 m) river-dominated tropical estuary in Ecuador (2.5°S latitude). There were statistical effects of site and depth on soil salinities, but all mean salinities were 〈17 g kg−1. Daily rates of leaf litter fall were higher in the rainy compared to the dry season, although no seasonal effects were observed for other components of litter fall. Annual total litter fall rates were significantly different among sites at 10.64, 6.47, and 7.87 Mg ha−1 year−1 for M1, M2, and M3, respectively. There were significant site (M3 〉 M2 〉 M1) and season (rainy 〉 dry) effects on leaf degradation, and both effects were related to differences in the initial nitrogen content of senescent leaves. Mean leaf litter standing crop among the sites ranged from 1.53 to 9.18 g m−2, but amounts were strongly seasonal with peak values during September in both years of our study (no significant year effect) at all three sites. Leaf turnover rates based on leaf fall estimates and litter standing crop were 10- to 20-fold higher than estimated from rates of leaf degradation, indicating the significant effect of leaf transport by tides and crabs. Field experiments demonstrated that the mangrove crab can remove daily additions of leaf material within 1 h at all three sites, except during August–October, when the crab is inactive on the forest floor. Even though there is seasonally elevated leaf accumulation on the forest floor during this time, leaf turnover rates are much higher than expected based on leaf degradation, demonstrating the importance of tidal export. This is the first description of how crabs influence litter dynamics in the New World tropics, and results are similar to higher rates of crab transport of leaf litter in the Old World tropics. Even in riverine mangroves with high geophysical energies, patterns of litter dynamics can be influenced by ecological processes such as crab transport.

Notes: Abstract In order to define the active domain for lipid binding in CETP (cholesteryl ester transfer protein), our study discusses some fundamental physicochemical properties of this molecule such as hydrophobic moment, protein active surface and helix amphipathicity, in comparison to the properties reported for a series of apoproteins including apoAI, apoAII, apoCI, CII, CIII and apoE. Our study suggests that CETP corresponds to a protein with an active surface slightly lower than the one calculated for the exchangeable apoproteins AI, AII, CI, CII, CIII and E. Arrays type (i, i + 3) and (i, i + 4) were found in the region associated to lipid binding in these apoproteins. Seven such arrays located in the amphipathic a-helices of CETP are also suggested to contribute to the overall lipid binding activity as a consequence of α-helix stability. It is proposed that for lipid binding to occur in both types of molecules, the possibility of a conformational specificity given by a redundant stereochemi cal code can be actively operating.
Description: This work was into the Network of Environmental Monitoring, Institutional Operational Plan 2002 IMARPE. In the first period (April 26-30) 16 sea sample points, and in the second one (July 24-27), 19 points were fixed; in both cases, 12 beach sample stations, one in Lacramarca river and other in Santa river, were set. Indicators of the marine environment and of the pollution from land were evaluated. In El Ferrol Bay the sewage are discharged through four pumping chambers in the north, and in the south, the Lacramarca river flows into the sea. The Coishco bay receives agricultural and domestic runoff, forming water mixes with the Santa river, with a typical salinity. The temperature corresponded to the área and season studied. Dissolved oxygen levels were low in July, with anoxia off fish factories in El Ferrol Bay, and hypoxia in some áreas into Coishco bay. Silicates and phosphates were high, mainly due to the influence of Santa river to Coishco. The total suspended solids (TSS) were within the permissible limits of the Peruvian General Waters Law. The Hydrogen sulfide concentrations were low. Oils and fats reached significant levels in some points of El Ferrol Bay southeast. In Coishco bay, the values of thermotolerant coliforms were high mainly in the central área; in El Ferrol bay, in July, those values exceeded the LGA for IV, V and VI classes. The Cd, Pb and Zn, trace metals had the lowest average concentration in the last two years. Hydrocarbon concentrations in sediments were generally low. The water quality of Santa river, at the end of the lower basin showed high concentrations of silicate and nitrate inputs; besides mainly thermo tolerant coliform contamination, mainly in April, and BO5D within the permissible levels.

Description: Nitric oxide (NO) produced by endothelial nitric oxide synthase (eNOS) is a critical mediator of vascular function. eNOS is tightly regulated at various levels, including transcription, co- and posttranslational modifications, and by various protein–protein interactions. Using stable isotope labeling with amino acids in cell culture (SILAC) and mass spectrometry (MS), we identified several eNOS interactors, including the protein plasminogen activator inhibitor-1 (PAI-1). In cultured human umbilical vein endothelial cells (HUVECs), PAI-1 and eNOS colocalize and proximity ligation assays demonstrate a protein–protein interaction between PAI-1 and eNOS. Knockdown of PAI-1 or eNOS eliminates the proximity ligation assay (PLA) signal in endothelial cells. Overexpression of eNOS and HA-tagged PAI-1 in COS7 cells confirmed the colocalization observations in HUVECs. Furthermore, the source of intracellular PAI-1 interacting with eNOS was shown to be endocytosis derived. The interaction between PAI-1 and eNOS is a direct interaction as supported in experiments with purified proteins. Moreover, PAI-1 directly inhibits eNOS activity, reducing NO synthesis, and the knockdown or antagonism of PAI-1 increases NO bioavailability. Taken together, these findings place PAI-1 as a negative regulator of eNOS and disruptions in eNOS–PAI-1 binding promote increases in NO production and enhance vasodilation in vivo.
