    Notes: Summary Full wing disc evagination requires about 10 h of continuous exposure to 20-hydroxyecdysone. The synthesis of two polypeptides is increased when wing discs are subjected to short exposure (4 h) to the hormone, and their synthesis is dependent on hormone. A second group of proteins increased in synthesis only after longer hormonal treatment (12 h); however, the increased synthesis of these proteins can be induced by withdrawing hormone after short exposure. The results of this study are consistent with the model of sequential gene activation by 20-hydroxyecdysone proposed by Ashburner et al. (1974).

    Notes: Abstract Lipid analogues and glycosylphosphati-dylinositol (GPI)-anchored proteins incorporated in glass-supported phospholipid bilayers (SBL) were coupled to small (30 nm diameter) fluorescent beads whose motion in the liquid phase was tracked by intensified fluorescence video microscopy. Streptavidin (St), covalently attached to the carboxyl modified surface of the polystyrene bead, bound either the biotinylated membrane component, or a biotinylated monoclonal antibody (mAb) directed against a specific membrane constituent. The positions of the beads tethered to randomly diffusing membrane molecules were recorded at 0.2 sec intervals for about l min. The mean square displacement (ϱ) of the beads was found to be a linear function of diffusion time t, and the diffusion coefficient, D, was derived from the relation, ϱ(t) = 4Dt. The values of D for biotinylated phosphatidylethanolamine (Bi-PE) dispersed in an egg lecithin: cholesterol (80:20%) bilayer obtained by this methodology range from 0.05 to 0.6 μm2/sec with an average of 〈D〉 = 0.26 μm2/sec, similar to the value of 〈D〉 = 0.24 μm2/sec for fluorescein-conjugated phosphati-dylethanolamine (Fl-PE) linked to St-coupled beads by the anti-fluorescein mAb 4-4-20 or its Fab fragment. These values of D are comparable to those reported for Fl-PE linked to 30 nm gold particles but are several times lower than that of Fl-PE in the same planar bilayer as measured by fluorescence photobleaching recovery, D = 1.3 μm2sec. The mobilities of two GPI-anchored proteins in similar SBL were also determined by use of the appropriate biotinylated mAb and were found to be 〈D〉 = 0.25 and 0.56 μm2/sec for the decay accelerating factor (DAF, CD55) and the human FcγRIIIB (CD16) receptors, respectively. The methodology described here is suitable for tracking any accessible membrane component.

    Notes: The monoatomic hydride layer on silicon was used as a prototype for resistless electron beam lithography. Arbitrary patterns with linewidths below 60 nm have been achieved. The variation of the linewidth with electron energy, electron dose, and substrate thickness was studied to determine the mechanisms that govern surface hydrogen desorption and subsequent pattern formation. Unlike in resist based lithography, no resolution enhancement was observed with decreasing substrate thickness. The experimental data in combination with Monte Carlo simulations of the backscattered and transmitted electrons suggest that surface hydrogen desorption and pattern formation are not strongly related to the backscattered electrons and the secondary electrons (energies 〈50 eV) associated with the backscattered electrons. © 1998 American Institute of Physics.

    Notes: Abstract The integration of representation and recognition of rigid solid objects is becoming increasingly important in computer-aided design (CAD), computer-aided manufacturing (CAM), computer graphics, computer vision, and other fields that deal with spatial phenomena. The mathematical framework used for modeling solid objects is mathematical morphology, which is based on set-theoretic concept. The mathematical characteristics of these operators are investigated in order to achieve a formal theory. Using mathematical morphology as a tool, our theoretical research aims at studying the representation schemes for the dimension and tolerance of the geometric structure. Object features can be also extracted by using the mathematical morphology approach. Through a distance transformation, we can obtain the shape number, significant points database, and skeleton. We have also developed the object recognition, localization, and corner and circle detection algorithms.

    Notes: Abstract Direct in situ introduction of retroviral producer cells might provide a form of treatment for localized tumors. A possible undesirable consequence of this treatment could be uncontrolled proliferation of the injected producer cells. To test this possibility, severe combined immunodeficiency (SCID) mice were reconstituted with human peripheral blood lymphocytes which were marked with a retroviral vector using a coculture method. Although specific measures were taken to remove the possible contaminating producer cells, a high percentage of mice developed fibrosarcoma 2–6 weeks after reconstitution. We hypothesized that tumors arose from a small number of contaminating producer cells in the inoculum. Tumor cells were consistently DNA tetraploid, a characteristic of the producer cell line. DNA extracted from tumor tissue was found to contain the gene (neomycin phosphotransferase) used to mark the producer cell line. Furthermore, SCID mice injected with 1 × 104 producer cells developed tumors with analogous characteristics. This report indicates that the retroviral producer cell line is tumorigenic in immune-deficient animals.
