    Notes: Summary Previous transfection experiments have shown that 162 base pairs (bp) of the 5′ flanking sequence of the chicken αA-crystallin gene are required for promoter activity in primary chicken lens epithelial cells (PLE), while only 111 by of the 5′ flanking sequence are needed for activity of the mouse αA-crystallin promoter in transfected chicken PLE cells or in a SV40 T-antigen-transformed transfected mouse lens epithelial cell line (αTN4-1). The effect of site-directed mutations covering positions −111 to −34 of the mouse αA-crystallin promoter fused to the bacterial chloramphenicol acetyltransferase (CAT) gene was compared in transfected chicken PLE cells and mouse αTN4-1 cells; selected mutations were also examined in a nontransformed rabbit lens epithelial cell line (N/N1003A). In general, the same mutations reduced promoter activity in the transfected lens cells from all three species, although differences were noted. The mutations severely affected regions −111/−106 and −69/−40 regions in all the transfected cells examined; by contrast, mutations at positions −105/−99 and −87/−70 had a somewhat greater effect in the chicken PLE than the mouse αTN4-1 cells, while mutations of the −93/−88 sequence reduced expression in the αTN4-1 but not the PLE cells. A partial cDNA with sequence similarity to αA-CRYPB 1 of the mouse has been isolated from a chicken lens library; mouse αA-CRYBP1 is a putative transcription factor which binds to the −66/−55 sequence of the mouse αA-crystallin promoter. Thus, despite differences in the molecular mechanisms of expression of the chicken and mouse αA-crystallin gene, the present results indicate numerous similarities in the behavior of the mouse promoter in the transfected lens cells of chicken, mouse, and rabbit.

    Notes: : An essential component to the ground water budget for the Las Vegas Valley (LVV) in southern Nevada is discharge from the ground water system. Discharge for the LW has been based on estimates made more than 50 years ago of 35,524,224 m3 per year as evapotranspiration (ET) and 0 m3 per year as subsurface outflow. Newly published values for recharge based on a more robust data set (70,308,360 m3) indicate a large imbalance associated with the earlier discharge estimates, providing the basis for the reevaluation conducted in this study. ET estimates in this study, as opposed to previous studies, were assigned a range in values that included an approach that assigned higher weight to the unique soil, plant, water, and climatic conditions that existed in predevelopment (1905) LW. The earlier discharge estimates also assumed that the basin was hydrologically closed; however, based on our evaluation, a range in yearly discharge by subsurface outflow from 1,480,176 m3 to 19,735,680 m3 could be assigned. Likewise, a range in yearly ET from 20,475,768 m3 to 78,819,372 m3 could be assigned. Based on newly published recharge values, closure can only occur if higher values are assigned to both the subsurface outflow and/or ET components of ground water discharge. We cannot provide a complete water balance closure with our ground water discharge estimate of 64,140,960 m3. However our reevaluation gives support to the higher recharge estimates and provides the rationale for future studies to be conducted based on a more rigorous scientific assessment.

    Notes: : Published estimates of natural recharge in Las Vegas Valley range between 21,000 and 35,000 acre-feet per year. This study examined the underlying assumptions of previous investigations and evaluated the altitude-precipitation relationships. Period-of-record averages from high altitude precipitation gages established in the 1940s through the 1990s, were used to determine strong local altitude-precipitation relationships that indicate new total precipitation and natural recharge amounts and a new spatial distribution of that recharge. This investigation calculated about 51,000 acre-feet per year of natural recharge in the Las Vegas Hydrographic Basin, with an additional 6,000 acre-feet per year from areas tributary to Las Vegas Valley, for a total of 57,000 acre-feet per year. The total amount of natural recharge is greater than estimates from earlier investigations and is consistent with a companion study of natural discharge, which estimated 53,000 acre-feet per year of outflow. The hydrologic implications of greater recharge in Las Vegas Valley infer a more accurate ground-water budget and a better understanding of ground-water recharge that will be represented in a ground-water model. Thus model based ground-water management scenarios will more realistically access impacts to the ground-water system.

    Notes: Summary A diploid strain of the yeast Saccharomyces cerevisiae has been constructed that has one copy of the ribosomal protein gene CYH2 completely deleted and replaced with the TRP1 gene using the method of Rothstein (1983). There are only small differences in growth rate and no detectable difference in steady state level of CYH2 mRNA between the diploid that is heterozygous for the CYH2 deletion and the parent diploid with two normal copies of this gene. This suggests that the diploid must partially compensate for the loss of one CYH2 gene. Tetrad dissection shows that haploid spores lacking the CYH2 gene cannot germinate. The lethality of this deletion can be rescued by a CYH2 cDNA on a low copy vector. Haploids which lack the genomic copy of the CYH2 gene, but contain a plasmid copy of the CYH2 cDNA are able to grow normally. These CYH2 deleted yeast haploids should be useful to analyze mutationally altered CYH2 genes and genes homologous to CYH2 from other organisms without interference from a genomic copy.

    Notes: The regulation and possible function of the preproenkephalin gene in testis were studied in vivo in transgenic mice containing: (1) bases -193 to +210 of the human proenkephalin gene and an additional one kilobase of 3' proenkephalin flanking sequence driving expression of bacterial chloramphenicol acetyltransferase (CAT), and (2) the same promoter and flanking sequences driving expression of a rat proenkephalin cDNA. Five lines of mice, designated HEC1-5, expressed the first construct and 10, HER1-10, the second. Each HEC male and many HER males showed dramatic expression of the transgene in the testis, although much lower expression was observed in the brain and other enkephalin-producing tissues. High levels of expression in testis can thus be achieved with a very short promoter region and do not require intron A sequences previously considered necessary. Altered enkephalin expression may affect testicular function. One founder, HER8, displayed grossly abnormal testicular morphology and was completely infertile. A second founder, HER6, had low sperm motility. Two offspring from other lines also displayed subnormal fertility. These studies support a role for specific promoter sequences in testis expression and may further support a significant role for proenkephalin in testicular function. © 1994 Wiley-Liss, Inc.

    Description: Abstract This study presents observations of the two‐dimensional velocities and field‐aligned currents of magnetospheric field line resonances. Knowledge of the two‐dimensional plasma flows is essential for determining the field‐aligned current structure of the field line resonance and for understanding the role that field line resonances play in the generation of some auroral arc forms. In this work the full velocity vectors are determined from a high resolution fit to the Super Dual Auroral Radar Network (SuperDARN) line‐of‐sight data. Two discrete field line resonance events are presented, one at 4 mHz (4‐5 min period) and the other at 0.6 mHz (30 min period). Bandpass filtering the north‐south and east‐west components of the total convection velocity vectors reveals the two‐dimensional velocity field specific to an individual field line resonance. The field line resonance velocities exhibit periodic vorticity which correspond to periodic poleward moving bands of enhanced field‐aligned current. Assuming a constant ionospheric Pedersen conductance of 10 Siemens, peak field‐aligned current values are estimated to be 2‐4 μA/m2. The field‐aligned currents alternate between upward and downward directed and peak in magnitude ~0.5‐1 degrees poleward and/or equatorward of the peak in field line resonance flow velocity. A Swarm satellite overpass occurs directly over the 0.6 mHz field line resonance. The SuperDARN field aligned current profile along the Swarm trajectory is found to be similar in magnitude and structure to that determined from Swarm‐A and Swarm‐C in situ measurements.

    Description: Background: CCN2/CTGF is an established effector of TGFbeta driven responses in diabetic nephropathy. We have identified an interaction between CCN2 and TGFbeta leading to altered phenotypic differentiation and inhibited cellular migration. Here we determine the gene expression profile associated with this phenotype and define a transcriptional basis for differential actin related gene expression and cytoskeletal function. Results: From a panel of genes regulated by TGFbeta and CCN2, we used co-inertia analysis to identify and then experimentally verify a subset of transcription factors, E2F1 and CREB, that regulate an expression fingerprint implicated in altered actin dynamics and cell hypertrophy. Importantly, actin related genes containing E2F1 and CREB binding sites, stratified by expression profile within the dataset. Further analysis of actin and cytoskeletal related genes from patients with diabetic nephropathy suggests recapitulation of this programme during the development of renal disease. The Rho family member Cdc42 was also found uniquely to be activated in cells treated with TGFbeta and CCN2; Cdc42 interacting genes were differentially regulated in diabetic nephropathy. Conclusions: TGFbeta and CCN2 attenuate CREB and augment E2F1 transcriptional activation with the likely effect of altering actin cytoskeletal and cell growth/hypertrophic gene activity with implications for cell dysfunction in diabetic kidney disease. The cytoskeletal regulator Cdc42 may play a role in this signalling response.
