    Description / Table of Contents: Vibration grinding of high-molecular pectic acid results in preparations of nearly constant composition and decreased average molecular weight in dependence of grinding time. By using this favourable procedure it is generally possible to produce standardized pectic acid preparations with better and more reproducible solubility properties. Such preparations are required, e.g., for comparative enzymatic investigations. For evaluating polygalacturonase activity it is proposed to use pectic acid preparations with the following properties: viscosity number [η] (intrinsic viscosity) in the range of 50-100 ml/g, mineral content lower than 1 %, and anhydrogalacturonic acid content in the ash-free dry substance minimum 80 %.
    Notes: Durch Schwingmahlung hochmolekularer Pektinsäure von unterschiedlicher Dauer gelangt man zu Präparaten weitgehend konstanter Zusammensetzung und vermindertem Durchschnitts-Molekulargewicht. Diese vorteilhafte Methode ermöglicht generell die Herstellung von standardisierten Pektinsäurepräparaten mit verbessertem und reproduzierbarem Lösungsverhalten, wie sie beispielsweise als Substrate für vergleichende enzymatische Untersuchungen benötigt werden. In diesem Zusammenhang wird zur Ermittlung der Polygalakturonase-Aktivität empfohlen, Pektinsäurepräparate mit einer Viskositätszahl [η] im Bereich von 50 bis 100 ml/g und einem unter 1% liegenden Aschegehalt sowie mit einem Mindestanteil von 80% Anhydrogalakturonsäure in der aschefreien Trockensubstanz zu verwenden.

    Description / Table of Contents: On vibration grinding of air-dried pectin preparations with a hydrolytic cleavage of the α(1←4) glycosidic linkage the degree of esterification remains constant. The random distribution of free and of esterified carboxyl groups remains unchanged. The affinity of the carboxyl groups to Ca2+ ions is changing only when oligomers are formed. The limit of a significant weakening of the ionic linkage being dependent on the chain lenght, will be shifted to higher viscosity numbers and to higher degrees of polymerization, respectively, with an increasing degree of esterification. Moreover, at a proceeding mechanolysis there is a characteristic decrease of the width of the distribution of molecular weights. This could be proved by gel chromatography. An enrichment of the neutral saccharides still present in the pectin preparations together with the D-galacturonic acid in spite of an intense purification, takes place independently of the grinding time and of the degree of degradation, respectively, as well in the low-molecular as in the high-molecular fractions of the gel chromatograms.
    Notes: Beim Schwingmahlabbau von lufttrockenen Pektinpräparaten bleibt unter der Bedingung einer hydrolytischen Spaltung der α(1←4)-glykosidischen Bindungen der Verseterungsgrad konstant. Die zu Beginn vorliegende statistische Verteilung der freien und der veresterten Carboxylgruppen wird unverändert beibehalten. Die Affinität der Carboxygruppen zu Ca2+-Ionen ändert sich erst im Bereich der vermehrten Buildung von oligomeren Abbauprodukten. Die von der Kettenlänge abhängige Grenze einer deutlichen Abschwächung der Ionenbindung verschiebt sich mit zunehmendem Veresterungsgrad zu höheren Viskositätszahlen bzw. Polymerisationsgraden. Mit fortschreitender Mechanolyse wird außerdem eine charakteristische Vereinheitlichung in der Molmassenverteilung gelchromatographisch nachgewiesen. Eine Anreicherung der die D-Galakturonsäure trotz intensiver Reingingung der Pektinpräparate noch begleitenden neutralen Saccharide tritt unabhängig von der Mahldauer bzw. dem Abbaugrad jeweils in den niedermolekularen und in den hochmolekularen Fraktionen der Gelchromatogramme auf.

    Description / Table of Contents: In percolating aqueous apple pectin solutions on the colour adsorber „Wofatit EZ“, especially the fractions with low molecular weight are adsorbed. Contrarily to galacturonic acid as well as to high-molecular pectin these fractions cannot be regained by washing with water. The lower the weight-average molecular weight of the tested pectins, the higher is the adsorption effect. Moreover the percolates show a decreased content in substances reducing 3,5-dinitrosalicylic acid. They contain pectins of a slightly increased degree of esterification. Therefore, preparative separation of degraded fractions from high-molecular pectin preparations by means of highly porous discolouring resins seems to be possible. However, during regeneration of Wofatit EZ by alkali and acid the adsorbed pectin fractions are desorbed to a great extent.
    Notes: Bei der Perkolation von wäßrigen Apfelpektinlösungen über den Farbadsorber „Wofatit EZ“ werden vorwiegend Fraktionen von niederem Molekulargewicht adsorptiv gebunden, die sich im Gegensatz zur Galakturonsäure und zum hochmolekularen Pektin durch Waschen mit Wasser nicht zurückgewinnen lassen. Dieser Adsorptionseffekt ist um so stärker ausgeprägt, je niedriger das Gewichtsdurchschnitts-Molekulargewicht der Testpektine liegt. Die Perkolate weisen außerdem einen geringeren Gehalt an 3,5-Dinitrosalicylsäure-reduzierenden Substanzen auf, und sie enthalten Pektine mit schwach erhöhtem Veresterungsgrad. Dadurch erscheint eine präparative Abtrennung von abgebauten Fraktionen aus hochmolekularen Pektinpräparaten mit Hilfe hochporöser Entfärbungsharze möglich. Im Prozeß der Regenerierug von Wofatit EZ mittels Lauge und Säre werden die adsorptiv gebundenen Pektinfraktionen hingegen wieder weitgehend desorbiert.

    Description / Table of Contents: Vibration grinding of metal pectates increases the destruction effect depending on the bound metallic cation in the sequence K+ 〈 Mg2+ 〈 Ca2+ ≪ Cu2+. Therefore, the mechanolysis of Cu-pectate can be used for preparing highly destructed galacturonans. Cu2+-ions destabilize the pectate molecule also during storage, consequently a conversion of the degradation products into the stable K+-form is necessary.
    Notes: Beim Schwingmahlabbau von Metallpektaten erhöht sich die Abbauwirkung mit dem gebundenen Metallkation in der Reihenfolge K+ 〈 Mg2+ 〈 Ca2+ ≪ Cu2+. Die Mechanolyse von Cu-Pektat kann daher zur Präparierung von stark abgebauten Galakturonanen eingesetzt werden. Cu2+-Ionen destabilisieren das Pektatmolekül auch bei der Lagerung, so daß eine Überführung der Abbauprodukte in die lagerstabile K+-Form erforderlich ist.
