    Notes: Abstract Light-limited cultures of the planktonic diatom Ditylum brightwellii (West) Grunow were grown at 14‰ salinity. Cells were subjected to oxidative stress induced by copper, in the presence of zinc. In two continuous cultures with total Zn levels of 40 and 140 nM, respectively, dissolved Cu levels were increased from 3 to 126 nM. This resulted in an increased Cu adsorption capacity of the cell walls, probably due to an increase of surface area and roughness. Sexual reproduction (auxospore formation) was accelerated but was considered as a non-specific stress response. Cu-induced oxidative stress was indicated by a decrease of reduced glutathione (GSH), and a removal of superoxide anions monitored as an increasing activity of superoxide dismutase (SOD). Although Zn has no oxidative potential per se, cell division rates and chlorophyll c contents were lower in the culture with high Zn levels. In both cultures, the pro-oxidant Cu caused a decrease of chlorophyll a, decreasing photosynthetic O2 evolution and cell devision rates, and a growing number of deformed and broken cells.

    Notes: Abstract Vitamin B6 induces in vivo as well as in vitro the appearance of a puff at region 2–48C in Drosophila hydei. At concentrations of 10−2 M or lower, region 2–48C is the only region responding to vitamin B6 provided that oligomycin is present in the incubation medium. Pyridoxal phosphate and pyridoxamine phosphate supplied to medium containing oligomycin induce upon incubation of salivary glands a larger puff at 2–48C. — Puff 2–48C produces large quantities of a unique RNP-product; globular 140–220 Å particles which aggregate to stable complexes of 0.1–0.2 μ in diameter. Upon continuous in vitro incubation with vitamin B6, puff 2–48C becomes loaded with these aggregates which have never been observed in any other puff of Drosophila hydei.

    Notes: Abstract The giant puff Balbiani ring 2 (BR 2) in the salivary glands of Chironomus tentans is known to contain transcriptionally active 75S RNA genes. The corresponding chromosomal superstructure, the BR 2 chromomere, has been identified and characterized in the present study. It was demonstrated by cytological methods that BR 2 originates from the broad band IV-3B10, since all the bands except the 3B10 band could be detected as intact bands in the vicinity of BR 2. In situ hybridization of BR 2 RNA to squashed Malpighian tubule chromosomes lacking BR 2, showed that all or almost all the DNA sequences complementary to BR 2 RNA are located in the 3B10 band. The DNA amount in the 3B10 band, designated the BR 2 band, was measured in relation to the DNA content of the whole chromosome set by both direct and indirect microspectrophotometry of Feulgen-stained Malpighian tubule chromosomes. From a determination of the haploid DNA content in sperm cells (0.25 pg), the amount of DNA in a BR 2 chromomere could then be calculated to be 5.1×10−4 pg DNA or 470 kb DNA. A minimum value of the DNA compaction within the BR 2 chromomere was estimated to 380 from the B-form length of BR 2 DNA (160 μm) and the thickness of the BR 2 band (0.42 μm). Since there are only between 1 and 4 75S RNA genes, 37 kb in size, in a BR 2 chromomere, most of the BR 2 DNA must consist of DNA not coding for 75S RNA. The nature of this DNA is discussed in relation to the high average AT-content of Chironomus DNA. The organization of the chromosome fiber in the BR 2 chromomere is considered in relation to the gene activation process in the salivary glands, i.e. the formation of a 75S RNA transcription loop from a portion of the tightly packed chromosome fiber in the BR 2 chromomere.

    Notes: Abstract Morphology and banding pattern of the 4th chromosome in Chironomus tentans Malpighian tubules have been investigated by electron microscopy, using the squash and selection technique. The map we composed from our observations shows a remarkable increase (75%) in band numbers as compared to the map previously presented by Beermann for the 4th chromosome from salivary glands. Extrapolation of this increase to the entire genome would result in a total band number of about 3,500. The mean DNA content of bands can thus be calculated to be about 50 kb. Many bands show a complex structure, including the BR2 band. Some bands seem to result from fusion of smaller components. “Minibands” have also been observed. Some interbands contain RNP particles. In our material the interbands appeared to be made up of fibrils with a diameter of about 120 Å. On the basis of these results we estimate the DNA in the interbands as amounting to 2% of the entire genome. The results are discussed with respect to the organization of the polytene chromosomes and the functional significance of the banding pattern.

    Notes: Cortical microtubules (MTs) allegedly orient nascent cellulose microfibrils (CMFs) in plant cells. The frequently observed parallelism between them, and the effect of MT-depolymerizing agents, are the bases for this hypothesis. Data have, however, accumulated about cells in which MTs and CMFs are not in parallel alignment. These data will be reviewed. MT orientation cannot be the only factor determining CMF orientation, but MTs could overrule other factors in cells where, for instance, they are more tightly attached to the plasma membrane than in other cells. MT and CMF orientations could, however, both be controlled by a third factor, and CMFs may even impose orientation on MTs.

    Notes: The view is expressed that the well-known formulae for measuring changes in terms of trade cannot be used to conclude whether a country is better off or not. Other variables which reflect the impact upon production, employment, and the competitive position should also be taken into account.Similarly, changes in terms of trade between the industrial sectors of an economy cannot be used to conclude whether one sector is better off than others. Other factors which play a role include increases in productivity per man-hour. These may help to explain why in some sectors prices increase less than in others. An attempt is made to illustrate this point using some statistics for the Netherlands.

    Notes: Preliminary experiments are described which aimed to identify compounds that could inhibit the attachment of Flavobacterium branchiophilum strains LAB4a and ATCC 35035 to the gills of rainbow trout. Total inhibition was never achieved, regardless of the compound tested. Formalin-killed or acetone-killed F. branchiophilum cells retained at least some of their adherent nature, relative to untreated (live) cells. Adherence was reduced by 22–33% following immersion of fish in one litre of water containing 0.21 mg of a homologous crude fimbrial extract. When fish were immersed in water containing hyperimmune rainbow trout antiLAB4a serum, a dose-dependent decrease in attachment (a reduction of 15% to 63%) of LAB4a to the gills was observed. Rainbow trout anti-LAB4a serum also reduced the attachment of ATCC 35035 to the gills, but this reduction was not significant. Adherence of LAB4a was not inhibited following exposure of fish to group 1 carbohydrates (arabinose, mannose and xylose), group 2 carbohydrates (dextrose, galactose and lactose), group 3 carbohydrates (galactosamine, glucosamine and fucose) or group 4 carbohydrates (N-acetyl-D-glucosamine, N-acetyl-D-galactosamine, N-acetyl-neuraminic acid and the globoceramide glycolipid from human erythrocytes). In contrast, when rainbow trout erythrocytes were incubated with the bacteria prior to bath challenge, this resulted in an 87% and 53% reduction in gill-associated LAB4a and ATCC 35035 antigen, respectively, following immersion of rainbow trout in this suspension.
