    Notes: Abstract Staphylococcus aureus α-toxin forms ionic channels of large size in lipid bilayer membranes. We have developed two methods for studying the mechanism of pore formation. One is based on measurement of the ionic current flowing through a planar lipid membrane after exposure to the toxin; the other is based on measuring the release of the fluorescent complex Tb-Dipicolinic acid from large unilamellar vesicles under similar conditions. Both methods indicate that the pore formation process is complex, showing an initial delay followed by non-linear kinetics. The power dependence of the pore formation rate on the toxin concentration in planar bilayers indicates that an aggregation mechanism underlies the channel assembly. Arrhenius plots, obtained with both techniques, show no deviation from linearity up to 50°C and the derived activation energies are found to be comparable to those for the binding and the lysis of rabbit erythrocytes by the same toxin. The temperature dependence of the conductance induced in planar bilayers by a large number of toxin channels indicates that the pores are filled with aqueous solution. The analysis of single conductance events shows that a heterogeneous population of pores exist and that smaller channels are preferred at low temperature. We attribute this heterogeneity to the existence of pores resulting from the aggregation of different numbers of monomers.

    Notes: The main determinants of soil respiration were investigated in 11 forest types distributed along an altitudinal and thermal gradient in the southern Italian Alps (altitudinal range 1520 m, range in mean annual temperature 7.8°C). Soil respiration, soil carbon content and principal stand characteristics were measured with standardized methods. Soil CO2 fluxes were measured at each site every 15–20 days with a closed dynamic system (LI-COR 6400) using soil collars from spring 2000 to spring 2002. At the same time, soil temperature at a depth of 10 cm and soil water content (m3 m−3) were measured at each collar. Soil samples were collected to a depth of 30 cm and stones, root content and bulk density were determined in order to obtain reliable estimates of carbon content per unit area (kg C m−2). Soil respiration and temperature data were fitted with a simple logistic model separately for each site, so that base respiration rates and mean annual soil respiration were estimated. Then the same regression model was applied to all sites simultaneously, with each model parameter being expressed as a linear function of site variables. The general model explained about 86% of the intersite variability of soil respiration. In particular, soil mean annual temperature explained the most of the variance of the model (0.41), followed by soil temperature interquartlile range (0.24), soil carbon content (0.16) and soil water content (0.05).

    Notes: We present carbon stable isotope, δ13C, results from air and organic matter samples collected during 98 individual field campaigns across a network of Carboeuroflux forest sites in 2001 (14 sites) and 2002 (16 sites). Using these data, we tested the hypothesis that δ13C values derived from large-scale atmospheric measurements and models, which are routinely used to partition carbon fluxes between land and ocean, and potentially between respiration and photosynthesis on land, are consistent with directly measured ecosystem-scale δ13C values. In this framework, we also tested the potential of δ13C in canopy air and plant organic matter to record regional-scale ecophysiological patterns.Our network estimates for the mean δ13C of ecosystem respired CO2 and the related ‘discrimination’ of ecosystem respiration, δer and Δer, respectively, were −25.6±1.9‰ and 17.8 ±2.0‰ in 2001 and −26.6±1.5‰ and 19.0±1.6‰ in 2002. The results were in close agreement with δ13C values derived from regional-scale atmospheric measurement programs for 2001, but less so in 2002, which had an unusual precipitation pattern. This suggests that regional-scale atmospheric sampling programs generally capture ecosystem δ13C signals over Europe, but may be limited in capturing some of the interannual variations.In 2001, but less so in 2002, there were discernable longitudinal and seasonal trends in δer. From west to east, across the network, there was a general enrichment in 13C (∼3‰ and ∼1‰ for the 2 years, respectively) consistent with increasing Gorczynski continentality index for warmer and drier conditions. In 2001 only, seasonal 13C enrichment between July and September, followed by depletion in November (from about −26.0‰ to −24.5‰ to −30.0‰), was also observed. In 2001, July and August δer values across the network were significantly related to average daytime vapor pressure deficit (VPD), relative humidity (RH), and, to a lesser degree, air temperature (Ta), but not significantly with monthly average precipitation (Pm). In contrast, in 2002 (a much wetter peak season), δer was significantly related with Ta, but not significantly with VPD and RH. The important role of plant physiological processes on δer in 2001 was emphasized by a relatively rapid turnover (between 1 and 6 days) of assimilated carbon inferred from time-lag analyses of δer vs. meteorological parameters. However, this was not evident in 2002. These analyses also noted corresponding diurnal cycles of δer and meteorological parameters in 2001, indicating a rapid transmission of daytime meteorology, via physiological responses, to the δer signal during this season.Organic matter δ13C results showed progressive 13C enrichment from leaves, through stems and roots to soil organic matter, which may be explained by 13C fractionation during respiration. This enrichment was species dependent and was prominent in angiosperms but not in gymnosperms. δ13C values of organic matter of any of the plant components did not well represent short-term δer values during the seasonal cycle, and could not be used to partition ecosystem respiration into autotrophic and heterotrophic components.

    Notes: Leaf age-dependent changes in structure, nitrogen content, internal mesophyll diffusion conductance (gm), the capacity for photosynthetic electron transport (Jmax) and the maximum carboxylase activity of Rubisco (Vcmax) were investigated in mature non-senescent leaves of Laurus nobilis L., Olea europea L. and Quercus ilex L. to test the hypothesis that the relative significance of biochemical and diffusion limitations of photosynthesis changes with leaf age. The leaf life-span was up to 3 years in L. nobilis and O. europea and 6 years in Q. ilex. Increases in leaf age resulted in enhanced leaf dry mass per unit area (MA), larger leaf dry to fresh mass ratio, and lower nitrogen contents per dry mass (NM) in all species, and lower nitrogen contents per area (NA) in L. nobilis and Q. ilex. Older leaves had lower gm, Jmax and Vcmax. Due to the age-dependent increase in MA, mass-based gm, Jmax and Vcmax declined more strongly (7- to 10-fold) with age than area-based (5- to 7-fold) characteristics. Diffusion conductance was positively associated with foliage photosynthetic potentials. However, this correlation was curvilinear, leading to lower ratio of chloroplastic to internal CO2 concentration (Cc/Ci) and larger drawdown of CO2 from leaf internal air space to chloroplasts (ΔC) in older leaves with lower gm. Overall the age-dependent decreases in photosynthetic potentials were associated with decreases in NM and in the fraction of N in photosynthetic proteins, whereas decreases in gm were associated with increases in MA and the fraction of cell walls. These age-dependent modifications altered the functional scaling of foliage photosynthetic potentials with MA, NM, and NA. The species primarily differed in the rate of age-dependent modifications in foliage structural and functional characteristics, but also in the degree of age-dependent changes in various variables. Stomatal openness was weakly associated with leaf age, but due to species differences in stomatal openness, the distribution of total diffusion limitation between stomata and mesophyll varied among species. These data collectively demonstrate that in Mediterranean evergreens, structural limitations of photosynthesis strongly interact with biochemical limitations. Age-dependent changes in gm and photosynthetic capacities do not occur in a co-ordinated manner in these species such that mesophyll diffusion constraints curb photosynthesis more in older than in younger leaves.

    Notes: Shoot architecture has been investigated using the ratio of mean shoot silhouette area to total needle area ( 〈inlineGraphic alt="inline image" href="urn:x-wiley:01407791:PCE974:PCE_974_mu1" location="image_n/PCE_974_mu1.gif"/〉) as a structural index of needle clumping in shoot space, and as the effective extinction coefficient of needle area. Although 〈inlineGraphic alt="inline image" href="urn:x-wiley:01407791:PCE974:PCE_974_mu1" location="image_n/PCE_974_mu1.gif"/〉 can be used effectively for the prediction of canopy gap fraction, it does not provide information about the within-shoot radiative regime. For this purpose, the estimation of three architectural properties of the shoots is required: needle area density, angular distribution and spatial aggregation. To estimate these features, we developed a method based on the inversion of a Markov three-dimensional interception model. This approach is based on the turbid medium approximation for needle area in the shoot volume, and assumes an ellipsoidal angular distribution of the normals to the needle area. Observed shoot dimensions and silhouette areas for different vertical and azimuth angles (AS) are used as model inputs. The shape coefficient of the ellipsoidal distribution (c) and the Markov clumping index (λ0) are estimated by a least square procedure, in order to minimize the differences between model prediction and measurements of AS. This methodology was applied to silver fir (Abies alba Mill.) shoots collected in a mixed fir–beech–spruce forest in the Italian Alps. The model worked effectively over the entire range of shoot morphologies: c ranged from 1 to 8 and λ0 from 0·3 to 1 moving from the top to the base of the canopy. Finally, the shoot model was applied to reconstruct the within-shoot light regime, and the potential of this technique in upscaling photosynthesis to the canopy level is discussed.

Die Zahl der mit Pheromon-Fallen eingefangenenL. monacha-Falter war umgekehrt proportional der Höhenstufe und bewegte sich innerhalb der bekannten Bereiche der latenten Bevölkerungsdichte. Die folgenden Arten von Blattwespen der GattungCephalcia wurden in genannter Reihenfolge in abnehmender Dichte angetroffen:C. arvensis, C. fallenii, C. abietis, C. alashanica. Sie wiesen unterschiedliche Flugperioden auf: als erste Art erschienC. fallenii im Frühling, gefolgt vonC. arvensis, C. abietis, C. alashanica und erneutC. arvensis. Das Problem der Flugperiodik wurde unter dem Aspekt der neuesten Entdekkungen in der Tschechoslowakei untersucht. Die angewandten Kontrollmethoden führten bezüglich des Geschlechtsanteiles und der Fangmenge zu verschiedenen Ergebnissen. Hinweise über die Anziehung zwischen den Geschlechtern wurden nachgewiesen. Die Fangmenge der am häufigsten vorkommenden Arten wurde stark beeinflußt von der Höhenlage und dem Charakter des Waldbestandes.L. monacha scheint niedrigere Höhen mit vorwiegend kleineren Bäumen vorzuziehen.C. arvensis undC. fallenii waren in kalten Gegenden an Bäumen von mittlerem Durchmesser. zahlreicher vorhanden. Die Ergebnisse wurden mit den Waldeigenschaften in Massenwechselgebieten verglichen.
    Notes: Abstract The life history and the ecology ofLymantria monacha andCephalcia spp. were studied in spruce (Picea abies) stands under latent conditions. Methods were based on the use of different monitoring devices, whose efficiency was evaluated. The abundance ofL. monacha monitored by pheromone traps, was inversely proportional to the altitude and the catches fell always within the range known for the latent populations. The following species of sawflies of the genusCephalcia were found, in order of decreasing abundance:C. arvensis, C. fallenii, C. abietis, C. alashanica. They presented different flight period,C. fallenii being the first emerging species in spring, followed byC. arvensis, C. abietis, C. alashanica andC. arvensis again. The problem of the peaks in the flight ofC. arvensis is discussed in the light of the recent findings in Czechoslovakia. The devices used in monitoring (tree band, yellow board, emergence trap) supplied different results for the sex ratio and the level of catches. Evidence for attraction between individuals is given. The catches of the most abundant species were strongly influenced by the altitude and the stand characteristics.L. monacha seems to prefer the lower sites, with a large number of small trees.C. arvensis andC. fallenii were most abundant in the colder sites, characterized by a medium (C. arvensis) or a medium-high (C. fallenii) diameter and by a low number of trees. The results are compared with the stand characteristics of the outbreak areas.

    Notes: Summary Staphylococcus aureus α-toxin causes cell damage by forming an amphiphilic hexamer that inserts into the cell membrane and generates a hydrophilic pore. To investigate the role of the three histidine residues of this toxin we modified them with diethylpyrocarbonate, obtaining N-carbethoxy-histidine whose appearance may be followed spectrophotometrically. Despite the statistical nature of random chemical modification, it was possible to establish that modification of any one of the three histidines was enough to impair α-toxin activity on red blood cells and platelets. Two out of three histidines were essential for the interaction of the toxin with model membranes such as lipid vesicles and planar bilayers. Loss of lytic activity in both natural and model membranes was due both to defective binding and to defective oligomerization. When α-toxin hexamers inserted into lipid vesicles were assayed for chemical modifiability two histidines per monomer were found to be protected from diethylpyrocarbonate modification, whereas only one was protected after delipidation of the oligomer with a detergent. A possible model for the role of each histidine in the monomer is presented.

    Notes: Summary Staphylococcus aureaus α-toxin opens an ion channel in planar phospholipid bilayers which is selective for anions over cations, supposedly because of the presence of positively charged groups along the ion pathway. To remove some positive charges of this protein toxin, we chemically modified part of its lysine residues either with diethylpyrocarbonate, followed by histidine regeneration with hydroxylamine, or with trinitrobenzenesulfonic acid. The extent of chemical modification can be followed accurately by native polyacrylamide gel electrophoresis and isoelectric focusing. Ethoxyformilation of two to three lysine residues per toxin monomer does not impair hemolysis of rabbit red blood cells nor formation of pores in model membranes. It reduces the conductance and the anion selectivity of the channel and changes the shape of its current-voltage characteristic. This indicates that positively charged lysine residues are actually important in determining the electrical properties of the pore. Ethoxyformilation of channels preassembled in planar bilayers produces the same changes as modification of toxin monomers before channel formation. Furthermore, it can be performed by adding diethylpyrocarbonate on either side of the bilayer. This suggests that the lysine residues relevant for the electrical properties of the pore are located inside its lumen where they can be reached by diethylpyrocarbonate diffusing from either entrance of the channel.

    Description: Significant climate risks are associated with a positive carbon–temperature feedback in northern latitude carbon-rich ecosystems, making an accurate analysis of human impacts on the net greenhouse gas balance of wetlands a priority. Here, we provide a coherent assessment of the climate footprint of a network of wetland sites based on simultaneous and quasi-continuous ecosystem observations of CO2 and CH4 fluxes. Experimental areas are located both in natural and in managed wetlands and cover a wide range of climatic regions, ecosystem types, and management practices. Based on direct observations we predict that sustained CH4 emissions in natural ecosystems are in the long term (i.e., several centuries) typically offset by CO2 uptake, although with large spatiotemporal variability. Using a space-for-time analogy across ecological and climatic gradients, we represent the chronosequence from natural to managed conditions to quantify the “cost” of CH4 emissions for the benefit of net carbon sequestration. With a sustained pulse–response radiative forcing model, we found a significant increase in atmospheric forcing due to land management, in particular for wetland converted to cropland. Our results quantify the role of human activities on the climate footprint of northern wetlands and call for development of active mitigation strategies for managed wetlands and new guidelines of the Intergovernmental Panel on Climate Change (IPCC) accounting for both sustained CH4 emissions and cumulative CO2 exchange.
