    Notes: The sequential changes in serum oestradiol 17β (measured by specific radioimmunoassay), vitellogenin (measured by phosphoprotein phosphorus content) and total calcium (measured by fluorimetry) in male and female rainbow trout under a simulated natural photoperiod cycle were investigated. Resting levels of 130 pg/ml serum oestradiol in both male and female fish were found in April and May. In the female, levels reached a peak of 4800 pg/ml in October, and almost returned to resting levels just prior to spawning in mid-January. No significant change from resting levels was seen in the male fish.In both male and female fish levels of 25 μg/ml serum phosphoprotein phosphorus and 10–14 mg % total serum calcium were found from April to July. Coincident with the rise in oestradiol 17β, in the females serum levels of phospho-protein phosphorus and total calcium increased to 400 uβg/ml and 58 mg% respectively just prior to spawning. In the males no significant change in either of these values was observed throughout the cycle. These results strongly support the hypothesis that photoperiod is the major factor in the environmental control of reproductive activity in the rainbow trout.

    Notes: Plasma melatonin synthesis in juvenile Atlantic salmon Salmo salar decreased with increasing light intensity. Melatonin profiles reflected accurately the photoperiod under which the salmon were maintained. Groups maintained at 12°C showed significantly higher (P 〈 0·01) levels of dark phase plasma melatonin compared with the groups maintained at 4°C.

    Notes: Abstract. The bacterial flora of rainbow, Salmo gairdneri Richardson, and brown trout, Salmo trutta L., eggs was studied during incubation. Few bacteria were considered to exist internally, although Pseudomonas species and Aeromonas hydrophila were isolated. During incubation, large numbers of bacteria approaching 500 colony forming units/mm2 accumulated around egg surfaces. Both rainbow and brown trout eggs placed in a through-flow system were colonized mainly by Pseudomonas sp. and A. hydrophila, whilst rainbow trout eggs in a separate recycling system were dominated by a Cytophaga species. Glass beads were also incubated (as an inert surface) and found to support significantly lower numbers of bacteria. Egg mortalities were recorded daily and found to differ significantly between both fish species and water conditions. Statistical analysis indicated a potentially significant correlation between hatching success and numbers of surface bacteria.

    Notes: Abstract. Eggs of female rainbow trout, Oncorhynchus mykiss (Walbaum), were fertilized and subsequently exposed to high eoncentrations of Pseudomonas fluorescens or Cytophaga sp. (106 cells/ml) for 45 min during water hardening. Eggs were placed in horizontal hatchery trays alongside sterile water-treated eggs and mortality rates were observed daily throughout incubation. Numbers and species of bacteria present on egg surfaces and within egg contents were monitored at the green stage (unfertilized), after hardening, and at 12h, 7 days, 14 days, 21 days and 28 days incubation. Surfaces of the incubating eggs of all groups, irrespective of treatment at water hardening, were eventually colonized by considerable numbers of Cytophaga sp., P. fluorescens and to a lesser extent Pseudomonas sp. and Aeromonas hydrophila. Few bacteria were recovered from inside eggs. Overall, no correlation was found between bacteria on the egg surface and egg death. However, a significantly higher ‘rate’ of egg death during early stages of incubation was found amongst eggs exposed to P. fluorescens at water hardening. Thus, high numbers of P. fluorescens on egg surfaces during initial stages of incubation might pose a potential threat to egg survival. Therefore, it is suggested that strenuous efforts are made at time of hardening to obtain the purest available source of water.

    Notes: Abstract. Lipid class and fatty acid analyses were carried out on eight batches of pre-fertilized eggs of Atlantic halibut, Hippoglossus hippoglossus (L.). These batches were classified as ‘viable’ or ‘non-viable’ according to fertilization rate, blastomere symmetry and incubation success. The lipid class and fatty acid compositions were very similar between egg categories with exception of cholesterol which was shown to be significantly greater in the ‘non-viable’ eggs. Polar lipid classes predominated (67·1% and 67·0% of the total lipid for ‘viable’ and ‘non-viable’ egg batches respectively), with phosphatidylcholine (PC) being the major class (43·2% and 43·5%) and triacylglycerol (TAG) the main neutral lipid class (18·8% and 17·8%). Gas liquid chromatography of fatty acid methyl esters from the total lipid revealed no significant differences in fatty acid composition. The fatty acid profiles of viable egg batches showed five major fatty acids 16:0 (17·3%), 18:0 (4·8%), 18:1n-9 (9%), 20:5n-3 (119%) and 22:6n-3 (25·9%). The results are discussed in terms of the potential role of lipids as egg quality determinants in fish.

    Notes: Abstract Plasma melatonin levels were measured at hourly intervals in rainbow trout, Oncorhynchus mykiss (Walbaum), maitained under three different skeleton photoperiods: 8L:2D:2L:12D;8L:7D:2L:7D; and 8L:12D:2L:2D. Blood samples were taken before, during and after the 2-h light pulses. Melatonin levels increased rapidly to mean scotophase (dark period) values of 261 ± 7 pg/ml after the first light-dark transition and had returned to mean photophase (light period) values of 51 ± 2 pg/ml 30min after the end of the scotophase. Light pulses during the early, mid- and late scotophase elicited similar reductions in melatonin levels, followed by rapid increases after the light pulse, reaching previous scotophase values within 90 min. The inability of any of the light pulses to modulate or truncate melatonin secretion under a LD 8:16 photoperiod provides further support for the hypothesis that melatonin secretion in the rainbow trout is a direct response to darkness, and is not under endogenous circadian control as in other vertebrates examined.

Measurements of egg number, egg size and total egg volume in relation to post-stripped weight, made on the eggs stripped from 12 separately maintained farm stocks of 4-year-old rainbow trout. Oncorhynchus mykiss Richardson, revealed highly significant differences between populations when analysed by regression and covariance statistical techniques.All of the stocks showed increases in fecundity with increasing fish size, with 61% (coefficient of determination or r2) of the variation in fecundity of the 12 stocks explained by the common regression on parental fish size. Comparison of the regressions of the different stocks revealed that the rate of increase of fecundity with increasing fish weight was constant throughout the weight range (i.e. the regressions had similar slopes) although there were highly significant differences in elevation, with some stocks producing almost twice as many eggs as some of the others.By contrast, the relationship of egg size with fish weight was much more variable with three of the stocks showing no significant regression of egg size on fish weight and a further two stocks with data poorly fitted to the regressions. Of the remaining seven stocks only one showed a significant difference in slope of the regression with modest increases in fish weight being associated with large increases in egg size. Only four of the remaining stocks of fish showed significant differences in the elevation of their regressions, i.e. the egg sizes were significantly different after the potentially conflicting influence of fish size was partitioned using covariance techniques.Assessments of total egg volume produced far improved regressions on post-strip weight with 75% (r2) of the variation in the pooled data of the 12 stocks being described by the line of the common regression. In general, there appeared to be a “trade-off” between egg number and size with the most and least fecund fish producing somewhat smaller and larger eggs respectively. All 12 regressions of totai egg volume on fish weight showed a common slope. However, there were significant differences in elevation of the regressions, with some of the stocks of trout producing up to a 55% greater volume of eggs.There appeared to be no relationship between the differences in egg number, size or total egg volume and the type or geograpfiical location of the farms on which the stocks were held.Although it remains to be determined to what extent these differences in reproductive performance are Inheritable, it is recommended that these characteristics should be taken into account by hatcheries in tbe design of long-term broodstock selection programmes for rainbow trout.
