    Description: Background: The human malaria parasite Plasmodium falciparum has a complex and multi-stage life cycle that requires extensive and precise gene regulation to allow invasion and hijacking of host cells, transmission, and immune escape. To date, the regulatory elements orchestrating these critical parasite processes remain largely unknown. Yet it is becoming increasingly clear that long non-coding RNAs (lncRNAs) could represent a missing regulatory layer across a broad range of organisms. Results: To investigate the regulatory capacity of lncRNA in P. falciparum, we harvested fifteen samples from two time-courses. Our sample set profiled 56 h of P. falciparum blood stage development. We then developed and validated strand-specific, non-polyA-selected RNA sequencing methods, and pursued the first assembly of P. falciparum strand-specific transcript structures from RNA sequencing data. This approach enabled the annotation of over one thousand lncRNA transcript models and their comprehensive global analysis: coding prediction, periodicity, stage-specificity, correlation, GC content, length, location relative to annotated transcripts, and splicing. We validated the complete splicing structure of three lncRNAs with compelling properties. Non-polyA-selected deep sequencing also enabled the prediction of hundreds of intriguing P. falciparum circular RNAs, six of which we validated experimentally. Conclusions: We found that a subset of lncRNAs, including all subtelomeric lncRNAs, strongly peaked in expression during invasion. By contrast, antisense transcript levels significantly dropped during invasion. As compared to neighboring mRNAs, the expression of antisense-sense pairs was significantly anti-correlated during blood stage development, indicating transcriptional interference. We also validated that P. falciparum produces circRNAs, which is notable given the lack of RNA interference in the organism, and discovered that a highly expressed, five-exon antisense RNA is poised to regulate P. falciparum gametocyte development 1 (PfGDV1), a gene required for early sexual commitment events.

    Description: Forests are a substantial terrestrial carbon sink, but anthropogenic changes in land \nuse and climate have considerably reduced the scale of this system1 \n. Remote-sensing \nestimates to quantify carbon losses from global forests2\xe2\x80\x935 \n are characterized by \nconsiderable uncertainty and we lack a comprehensive ground-sourced evaluation to \nbenchmark these estimates. Here we combine several ground-sourced6 \n and satellitederived approaches2,7,8 \n to evaluate the scale of the global forest carbon potential \noutside agricultural and urban lands. Despite regional variation, the predictions \ndemonstrated remarkable consistency at a global scale, with only a 12% diference \nbetween the ground-sourced and satellite-derived estimates. At present, global forest \ncarbon storage is markedly under the natural potential, with a total defcit of 226\xe2\x80\x89Gt \n(model range\xe2\x80\x89=\xe2\x80\x89151\xe2\x80\x93363\xe2\x80\x89Gt) in areas with low human footprint. Most (61%, 139\xe2\x80\x89Gt\xe2\x80\x89C) \nof this potential is in areas with existing forests, in which ecosystem protection can \nallow forests to recover to maturity. The remaining 39% (87\xe2\x80\x89Gt\xe2\x80\x89C) of potential lies in \nregions in which forests have been removed or fragmented. Although forests cannot \nbe a substitute for emissions reductions, our results support the idea2,3,9 \n that the \nconservation, restoration and sustainable management of diverse forests ofer \nvaluable contributions to meeting global climate and biodiversity targets.

    Description: Understanding what controls global leaf type variation in trees is crucial for \ncomprehending their role in terrestrial ecosystems, including carbon, water \nand nutrient dynamics. Yet our understanding of the factors infuencing \nforest leaf types remains incomplete, leaving us uncertain about the global \nproportions of needle-leaved, broadleaved, evergreen and deciduous \ntrees. To address these gaps, we conducted a global, ground-sourced \nassessment of forest leaf-type variation by integrating forest inventory \ndata with comprehensive leaf form (broadleaf vs needle-leaf) and habit \n(evergreen vs deciduous) records. We found that global variation in leaf \nhabit is primarily driven by isothermality and soil characteristics, while leaf \nform is predominantly driven by temperature. Given these relationships, \nwe estimate that 38% of global tree individuals are needle-leaved evergreen, \n29% are broadleaved evergreen, 27% are broadleaved deciduous and \n5% are needle-leaved deciduous. The aboveground biomass distribution \namong these tree types is approximately 21% (126.4\xe2\x80\x89Gt), 54% (335.7\xe2\x80\x89Gt), 22% \n(136.2\xe2\x80\x89Gt) and 3% (18.7\xe2\x80\x89Gt), respectively. We further project that, depending \non future emissions pathways, 17\xe2\x80\x9334% of forested areas will experience \nclimate conditions by the end of the century that currently support a \ndiferent forest type, highlighting the intensifcation of climatic stress on \nexisting forests. By quantifying the distribution of tree leaf types and their \ncorresponding biomass, and identifying regions where climate change will \nexert greatest pressure on current leaf types, our results can help improve \npredictions of future terrestrial ecosystem functioning and carbon cycling.

    Notes: The first order difference method of neutron diffraction and isotopic substitution has been applied to argon dissolved under pressure in heavy water. The results show that argon atoms possess a well-defined nearest neighbor hydration shell composed of 16(2) water molecules in the range 2.8≤r (A(ring))≤5.4. Also evident is a weaker second shell which extends to ∼8 A(ring). The results are in broad agreement with those obtained from Monte Carlo and M.D. computer simulation of models in which an argon atom interacts with a number of water molecules via pairwise additive forces. Significant differences are discernible when attempts are made to match quantitatively the theoretical results over the entire range of the experimental information.

    Notes: Far-reaching managerial changes to the public sector in the UK have been occurring in the last few years. Broadly described as the ‘new public management’ (Hood 1991, 1995) these changes have reached new levels of intensity ever since the Conservative Government came to power in 1979. The article argues that the government should be accountable for these changes but not in a way which simply justifies its actions to the body politic but rather by going beyond this to a complete critical evaluation to assess their merit and worth. It is this exposure which will provide its real accountability. This article addresses the contextual factors surrounding the possibility of conducting such a wide-ranging evaluation. It traces a range of different examples which highlight the resistance of the previous government to an open-ended evaluation tracing the sizeable investment in implementation rather than careful policy experimentation and evaluation that has occurred. Finally, and most importantly the article explores the constitutional position of the Comptroller and Auditor General (C&AG) and the National Audit Office (NAO) who could conduct the wide-ranging evaluation which is deemed necessary. The conclusion of the article is that there is a need for an evaluation and any resistance of the government could be overcome if the C&AG and NAO exercised their constitutional right to conduct an evaluation of the reforms.

    Notes: A rapid thermal anneal (RTA) in an NH3 ambient has been found to increase the thermal stability of W films chemically vapor deposited (CVD) on Si. W films deposited onto single-crystal Si by low-pressure CVD were rapid thermal annealed at temperatures between 500 and 1100 °C in NH3 and Ar ambients. The reactions were studied using Rutherford backscattering spectrometry, x-ray diffraction, Auger electron spectroscopy, transmission electron microscopy, and four-point resistivity probe. High-temperature (≥1000 °C) RTA in Ar completely converted W into the low resistivity (31 μΩ cm) tetragonal WSi2 phase. In contrast, after a prior 900 °C RTA in NH3, N inclusion within the W film and at the W/Si interface almost completely suppressed the W-Si reaction. Detailed examination, however, revealed some patches of WSi2 formed at the interface accompanied by long tunnels extending into the substrate, and some crystalline precipitates in the substrate close to the interface. The associated interfacial contact resistance was only slightly altered by the 900 °C NH3 anneal. The NH3-treated W film acted as a diffusion barrier in an Al/W/Si contact metallurgy up to at least 550 °C, at which point some increase in contact resistance was measured.
