    Description: A set of thermodynamic models is presented that, for the first time, allows partial melting equilibria to be calculated for metabasic rocks. The models consist of new activity-composition relations combined with end-member thermodynamic properties from the Holland & Powell dataset, version 6. They allow for forward modelling in the system Na2O-CaO-K2O-FeO-MgO-Al2O3-SiO2-H2O-TiO2-Fe2O3. In particular, new activity-composition relations are presented for silicate melt of broadly trondhjemitic-tonalitic composition, and for augitic clinopyroxene with Si-Al mixing on the tetrahedral sites, while existing activity-composition relations for hornblende are extended to include K2O and TiO2. Calibration of the activity-composition relations was carried out with the aim of reproducing major experimental phase-in/phase-out boundaries that define the amphibolite-granulite transition, across a range of bulk compositions, at ≤ 13 kbar.     Description: Rocks of basic and intermediate bulk composition occur in orogenic terranes from all geological time periods and are thought to represent significant petrological components of the middle and lower continental crust. However, the former lack of appropriate thermodynamic models for silicate melt, amphibole, and clinopyroxene that can be applied to such lithologies at high temperature has inhibited effective phase equilibrium modelling of their petrological evolution during amphibolite- and granulite-facies metamorphism. In this work, we present phase diagrams calculated in the Na2O–CaO–K2O–FeO–MgO–Al2O3–SiO2–H2O–TiO2–O2 (NCKFMASHTO) compositional system for a range of natural basic and intermediate bulk compositions for conditions of 2–12 kbar and 600–1050 ◦ C using newly parameterised activity–composition relationships detailed in a companion paper in this issue (Green et al ., 2016). Particular attention is given to mid-ocean ridge basalt (MORB) and diorite protolith bulk compositions. Calculated subsolidus mineral assemblages in all basic and intermediate rock types are modally dominated by hornblende and plagioclase, with variable proportions of epidote, clinopyroxene, garnet, biotite, muscovite, quartz, sphene, or ilmenite present at different pressures. The H2O-saturated (wet) solidus has a negative P – T slope and occurs between ∼ 620–690 ◦ C at mid- to lower-crustal pressures of 5–10 kbar. The lowest- T melts generated close to the wet solidus are calculated to have granitic major-element oxide compositions. Melting at higher temperature is attributed primarily to multivariate hydrate-breakdown reactions involving biotite and/or hornblende. Partial melt compositions calculated at 800–1050 ◦ C for MORB show good correlation with analysed compositions of experimental glasses produced via hydrate-breakdown melting of natural and synthetic basic protoliths, with Niggli norms indicating that they would crystallise to trondhjemite or tonalite. Diorite is shown to be significantly more fertile than MORB and is calculated to produce high- T melts ( 〉 800 ◦ C) of granodioritic composition. Subsolidus and suprasolidus mineral assemblages show no significant variation between different members of the basalt family, although the P – T conditions at which orthopyroxene stabilises, thus defining the prograde amphibolite–granulite transition, is strongly dependent on bulk-rock oxidation state and water content. The petrological effects of open- and closed-system processes on the mineral assemblages produced during prograde metamorphism and preserved during retrograde metamorphism are also examined via a case-study analysis of a natural Archaean amphibolite from the Lewisian Complex, northwest Scotland.     Description: We present the design and capabilities of a high-resolution, decade-spanning ASynchronous OPtical Sampling (ASOPS)-based TeraHertz Time-Domain Spectroscopy (THz-TDS) instrument. Our system employs dual mode-locked femtosecond Ti:Sapphire oscillators with repetition rates offset locked at 100 Hz via a Phase-Locked Loop (PLL) operating at the 60th harmonic of the ∼80 MHz oscillator repetition rates. The respective time delays of the individual laser pulses are scanned across a 12.5 ns window in a laboratory scan time of 10 ms, supporting a time delay resolution as fine as 15.6 fs. The repetition rate of the pump oscillator is synchronized to a Rb frequency standard via a PLL operating at the 12th harmonic of the oscillator repetition rate, achieving milliHertz (mHz) stability. We characterize the timing jitter of the system using an air-spaced etalon, an optical cross correlator, and the phase noise spectrum of the PLL. Spectroscopic applications of ASOPS-THz-TDS are demonstrated by measuring water vapor absorption lines from 0.55 to 3.35 THz and acetonitrile absorption lines from 0.13 to 1.39 THz in a short pathlength gas cell. With 70 min of data acquisition, a 50 dB signal-to-noise ratio is achieved. The achieved root-mean-square deviation is 14.6 MHz, with a mean deviation of 11.6 MHz, for the measured water line center frequencies as compared to the JPL molecular spectroscopy database. Further, with the same instrument and data acquisition hardware, we use the ability to control the repetition rate of the pump oscillator to enable THz frequency comb spectroscopy (THz-FCS). Here, a frequency comb with a tooth width of 5 MHz is generated and used to fully resolve the pure rotational spectrum of acetonitrile with Doppler-limited precision. The oscillator repetition rate stability achieved by our PLL lock circuits enables sub-MHz tooth width generation, if desired. This instrument provides unprecedented decade-spanning, tunable resolution, from 80 MHz down to sub-MHz, and heralds a new generation of gas-phase spectroscopic tools in the THz region.

    Description: Strain hardening capability is critical for metallic materials to achieve high ductility during plastic deformation. A majority of nanocrystalline metals, however, have inherently low work hardening capability with few exceptions. Interpretations on work hardening mechanisms in nanocrystalline metals are still controversial due to the lack of in situ experimental evidence. Here we report, by using an in situ transmission electron microscope nanoindentation tool, the direct observation of dynamic work hardening event in nanocrystalline nickel. During strain hardening stage, abundant Lomer-Cottrell (L-C) locks formed both within nanograins and against twin boundaries. Two major mechanisms were identified during interactions between L-C locks and twin boundaries. Quantitative nanoindentation experiments recorded show an increase of yield strength from 1.64 to 2.29 GPa during multiple loading-unloading cycles. This study provides both the evidence to explain the roots of work hardening at small length scales and the insight for future design of ductile nanocrystalline metals.     Description: The recent publication of an updated thermodynamic dataset for petrological calculations provides an opportunity to illustrate the relationship between experimental data and the dataset, in the context of a new set of activity–composition models for several key minerals. These models represent orthopyroxene, clinopyroxene and garnet in the system CaO–MgO–Al 2 O 3 –SiO 2 (CMAS), and are valid up to 50 kbar and at least 1800 °C; they are the first high-temperature models for these phases to be developed for the Holland & Powell dataset. The models are calibrated with reference to phase-relation data in the subsystems CaO–MgO–SiO 2 (CMS) and MgO–Al 2 O 3 –SiO 2 (MAS), and will themselves form the basis of models in larger systems, suitable for calculating phase equilibria in the crust and mantle. In the course of calibrating the models, it was necessary to consider the reaction orthopyroxene + clinopyroxene + spinel = garnet + forsterite in CMAS, representing a univariant transition between simple spinel and garnet lherzolite assemblages. The high-temperature segment of this reaction has been much disputed. We offer a powerful thermodynamic argument relating this reaction to the equivalent reaction in MAS, that forces us to choose between good model fits to the data in MAS or to the more recent data in CMAS. We favour the fit to the MAS data, preserving conformity with a large body of experimental and thermodynamic data that are incorporated as constraints on the activity–composition modelling via the internally consistent thermodynamic dataset.

    Description: A fully thermodynamic model for mafic melt in CaO–MgO–Al 2 O 3 –SiO 2 (CMAS) has been calibrated, for calculation of melting equilibria in the pressure range 0–50 kbar. It is intended as a preliminary step towards a large-system melt model, suitable for exploring melting, melt loss and crystallization processes in a wide range of natural rock compositions. Calibration was performed with attention to the model's behaviour in its compositional subsystems, as a rigorous test of model structure and parameterization. The model is consistent with the latest Holland & Powell thermodynamic data set, and can therefore be used to calculate phase relations in conjunction with the many solid-phase activity–composition models written for the data set. Model calculations successfully reproduce experimental melting reactions in CMAS spinel lherzolite and garnet lherzolite assemblages, as well as sapphirine- and kyanite-bearing assemblages, at moderate to high pressure. Thermodynamically sensitive features, such as thermal divides are also recovered. However, some changes to the model structure will be required before the model can describe the full range of mafic and ultramafic melt compositions known from experiment at low pressures.

    Description: A novel approach has been developed for the isolation and maturation of human antibodies that replicates key features of the adaptive immune system by coupling in vitro somatic hypermutation (SHM) with mammalian cell display. SHM is dependent on the action of the B cell specific enzyme, activation-induced cytidine deaminase (AID), and can be replicated in non-B cells through expression of recombinant AID. A library of human antibodies, based on germline V-gene segments with recombined human regions was used to isolate low-affinity antibodies to human β nerve growth factor (hβNGF). These antibodies, initially naïve to SHM, were subjected to AID-directed SHM in vitro and selected using the same mammalian cell display system, as illustrated by the maturation of one of the antibodies to low pM KD. This approach overcomes many of the previous limitations of mammalian cell display, enabling direct selection and maturation of antibodies as full-length, glycosylated IgGs.
