    Notes: Abstract. Easy-to-use audio/video authoring tools play a crucial role in moving multimedia software from research curiosity to mainstream applications. However, research in multimedia authoring systems has rarely been documented in the literature. This paper describes the design and implementation of an interactive video authoring system called Zodiac, which employs an innovative edit history abstraction to support several unique editing features not found in existing commercial and research video editing systems. Zodiac provides users a conceptually clean and semantically powerful branching history model of edit operations to organize the authoring process, and to navigate among versions of authored documents. In addition, by analyzing the edit history, Zodiac is able to reliably detect a composed video stream's shot and scene boundaries, which facilitates interactive video browsing. Zodiac also features a video object annotation capability that allows users to associate annotations to moving objects in a video sequence. The annotations themselves could be text, image, audio, or video. Zodiac is built on top of MMFS, a file system specifically designed for interactive multimedia development environments, and implements an internal buffer manager that supports transparent lossless compression/decompression. Shot/scene detection, video object annotation, and buffer management all exploit the edit history information for performance optimization.

    Notes: Abstract Computer analysis of protein phosphorylation site sequences revealed that transcriptional factors and viral oncoproteins are prime targets for regulation of proline-directed protein phosphorylation, suggesting an association of the proline-directed protein kinase (PDPK) family with neoplastic transformation and tumorigenesis. In this report, an immunoprecipitate activity assay of proline-directed protein kinase FA/glycogen synthase kinase-3α (PDPK FA/GSK-3α) has been optimized to demonstrate significantly increased (p 〈 0.01) activity in poorly differentiated human prostate carcinoma PC-3 cells (55.5 ± 3.8 units/mg) when compared to well-differentiated LNCaP cells (28.1 ± 2.3 units/mg). Immunoblotting analysis revealed that increased activity of this PDPK. in PC-3 cells is due not to overexpression of the protein, but to enhanced tyrosine phosphorylation of the kinase. When treated with genistein (a protein tyrosine kinase PTK inhibitor), the enhanced tyrosine phosphorylation/activation of the kinase in PC-3 cells can be blocked. Conversely, when treated with vanadate (a protein tyrosine phosphatase PTP inhibitor), the phosphotyrosine content of PDPK FA/GSK-3α in LNCaP cells can be promoted to the level of PC-3 cells. In sharp contrast, the PTK inhibitor has little effect on the tyrosine phosphorylation level of the kinase in LNCaP cells, whereas the PTP inhibitor has little effect on the tyrosine phosphorylation level of the kinase in PC-3 cells. Taken together, the results provide initial evidence that the tyrosine phosphorylation/activation levels of this oncogenic PDPK can be differentially regulated in well- and poorly differentiated prostate carcinoma cells.

    Notes: Abstract The single-spin-flip Metropolis algorithm is applied to an Ising ferromagnet with mixed spins ofS=1/2 andS=1 on the square lattice. The critical temperature obtained from our Monte Carlo simulation is very close to the high temperature series expansion result. The finite size scaling results for the exponents yield the two dimensional Ising values, which are in good agreement with those suggested by the universality hypothesis.

In this report, an immunoprecipitate activity assay of protein kinase FA/glycogen synthase kinase-3α (kinase FA/GSK-3α) (a particular member of PDPK family) has been optimized for human cervical tissue and used to demonstrate for the first time significantly increased (P 〈 0.001) activity in poorly differentiated cervical carcinoma (82.8 ± 6.6 U/mg of protein), moderately differentiated carcinoma (36.2 ± 3.4 U/mg of protein), and well-differentiated carcinoma (18.3 ± 2.4 U/mg of protein) from 36 human cervical carcinoma samples when compared to 12 normal controls (4.9 ± 0.6 U/mg of protein). Immunoblotting analysis further revealed that increased activity of kinase FA/GSK-3α in cervical carcinoma is due to overexpression of protein synthesis of the kinase. Taken together, the results provide initial evidence that overexpression of protein synthesis of the kinase. Taken together, the results provide initial evidence that overexpression of protein synthesis and cellular activity of kinase FA/GSK-3α may be involved in human cervical carcinoma dedifferentiation/progression, supporting an association of proline-directed protein kinase with neoplastic transformation and tumorigenesis. Since protein kinase FA/GSK-3α may function as a possible regulator of transcription factors/proto-oncogenes, the results further suggest that kinase FA/GSK-3α may play a potential role in human cervical carcinogenesis, especially in its dedifferentiation and progression. © 1995 Wiley-Liss, Inc.

Immunoblotting analysis revealed that increased activity of kinase FA/GSK-3α is due to overexpression of the protein. Elevated kinase FA/GSK-3α expression in human hepatoma biopsies relative to normal liver tissue was found to be even more profound. This kinase appeared to be ∼fivefold overexpressed in well differentiated hepatoma and ∼13-fold overexpressed in poorly differentiated hepatoma when compared to normal liver tissue. Taken together, the results provide initial evidence that overexpression of kinase FA/GSK-3α is involved in human hepatoma dedifferentiation/progression. Since kinase FA/GSK-3α is a PDPK, the results further support a potential role of this kinase in human liver tumorigenesis, especially in its dedifferentiation/progression. © 1996 Wiley-Liss, Inc.

    Notes: The molecular structure and polarizabilities of poly(aryl ether ketone)s were investigated with quantum chemical calculations, based on the fully optimized structures of model compounds (S5: ArCOArOArOArCOAr and BS6: ArOArCOArArCOArOAr) for their chain segments. The results showed that the average dihedral angle between their aromatic rings was in the range of 36°-37° relative to the molecular zig-zag plane. Especially, when a biphenyl group was introduced into the molecular chain of this polymer, its two aromatic rings were distorted by 48.0° to each other. Also, the calculation results of refractive indices for pure crystals obtained by using the structural parameters were in good accordance with those reported in the literature.

    Notes: As compared to normal people, the lymphocytes of patients with schizophrenia were found to have an impairment of ATP. Mg-dependent protein phosphatase activation. More importantly, the impaired protein phosphatase activation in the lymphocytes of schizophrenic patients could be consistently and completely restored to normal by exogenous pure protein kinase FA/glycogen synthase kinase-3α (kinase FA/GSK-3α) (the activating factor of ATP.Mg-dependent protein phosphatase), indicating that the molecular mechanism for the impaired protein phosphatase activation in schizophrenic patients may be due to a functional loss of kinase FA/GSK-3α immunoblotting and kinase activity analysis in an anti-kinase FA/GSK-3α immunoprecipitate further demonstrate that both cellular activities and protein levels of kinase FA/GSK-3α in the lymphocytes of schizophrenic patients were greatly impared as compared to normal controls. Statistical analysis revealed that the lymphocytes isolated from 37 normal people contain kinase FA/GSK-3α activity in the high levels of 14.8 ± 2.4 units/mg of cell protein, whereas the lymphocytes of 48 patients with schizophrenic disorder contain kinase FA/GSK-3α activity in the low levels of 2.8 ± 1.6 units/mg, indicating that the different levels of kinase FA/GSK-3α activity between schizophrenic patients and normal people are statistically significant. Taken together, the results provide intial evidence that patients with schizophrenic disorder may have a common impairment in the protein levels and cellular activities of kinase FA/GSK-3α, a multisubstrate protein kinase and a multisubstrate protein phosphatase activator in their lymphocytes. © 1995 Wiley-Liss, Inc.

    Notes: The molecular structure of a series of model compounds for poly(aryl ether ketone)s was investigated by means of 13C NMR spectroscopy, quantum chemical calculation, and wide-angle X-ray scattering. The comparison of the chemical shifts for the model compounds dissolved in 98% H2SO4 and CDCl3 (or CDCl3 + DMSO-d6) suggested that their carbonyl groups are partially protonated by sulfuric acid and the induced positive charges are delocalized in their aromatic rings. This result was supported by the charge distribution obtained by quantum chemical calculations. The model compounds were also used to characterize the molecular structure of poly(aryl ether ketone)s. The length of the repeating unit of the polymer chain, which was calculated based on the whole chain length or fragment chain length of the model compounds obtained by semiempirical PM3 method, is compared with the c-axis of the crystal unit cell of the polymer. For poly(aryl ether ketone)s containing biphenyl moieties, the length of the c-axis of the unit cell is approximately twice that of the repeating unit of the macromolecule.

    Notes: Poly(aryl ether ether ketone ketone)s (PEEKK) containing meta-phenyl links and their series of copolymers were synthesized and investigated by both X-ray and differential scanning calorimetry (DSC) methods. Results showed that the heat properties of this kind of copolymer depended greatly on the content of meta-phenyl links in the copolymer system, in which occurred the lowest melting point. Results from X-rays showed that PEEKK containing meta-phenyl links had no (111) crystal face diffraction. These proved that meta-phenyl links had introduced asymmetrical factors, which had produced poor crystal structure and difficulty in crystallization. Even so, the modification of PEEKK by introducing the meta-phenyl links improved the polymer composite performances, e.g., the copolymer M2, which kept performances close to PEEKK but better than PEEK. DSC results of M2 showed that its Avrami number (n) was 1.5 and its crystal grew fibrously from isothermal crystallization of the melting state, while for the nonisothermal crystallization from the melting state, n was 4.4 to the spherical crystal growth, and the activation energy (ΔE) of crystallization was 184 kJ/mol, which was less than the ΔE of 296 kJ/mol for PEEKK crystallized from the nonisothermal melting state. When M2 was isothermally crystallized from the rubber state, its n was 2 to the disklike crystal growth, while its n was 4.6 to the spherulitic crystal growth for the nonisothermal crystallization state of melting. The isothermal crystallization process was different from the nonisothermal process in the crystal nucleation and growth for M2. © 1996 John Wiley & Sons, Inc.
