    Notes: Abstract The design and use of a marine heat probe with capability for measuring thermal conductivity insitu with high accuracy, and providing digital acoustic transmission of data to the ship, is described. The instrument employs the ‘violin bow’ strength member and parallel sensor string configuration suggested by C. R. B. Lister. Several hundred measurements have been made in the deep ocean on multipenetration or ‘pogostick’ profiles using a 3 m probe and in deep inlets of western Canada using a 7 m probe. The insitu thermal conductivity technique using a calibrated heat pulse has been studied in detail through laboratory calibration of the probe in materials of known conductivity, through numerical models, and through comparison of insitu measurements with needle probe measurements on sediment cores taken from the same sites. The insitu technique permits a conductivity accuracy of better than ±5% with a recording time of 7 minutes following 7 minutes in the bottom to establish the geothermal gradient. The pulse heating is also more energy efficient than the conventional continuous heating technique.

    Notes: Summary In recent years, distinct changes in regulatory peptides have been found in a number of gastrointestinal diseases. Grass sickness is a fatal disease of horses for which the etiology has yet to be fully ascertained. In this study, the peptide-containing nerves and ganglionic and mucosal endocrine cells of the ileum, colon and rectum were investigated in horses with sub-acute or chronic grass sickness and compared with normal controls using immunocytochemistry, at both the light and electron microscopical levels, and radioimmunoassay. A substantial loss of both peptide-containing cells and nerves was found in all of the sick horses, particularly in the ileum. Electron microscopy revealed marked degeneration of nerves in the gut wall. fibers containing granules immunostained for substance P or VIP, using the immunogold staining technique, underwent extensive degranulation in grass sickness, with the formation of multiple vacuoles. Radioimmunoassay of peptide content also showed that the most drastic changes occurred in the ileum. For example, VIP content was significantly reduced from 109±19.8 (mean±SEM) pmoles/g in controls to 6.8±1.4 pmoles/g in grass sickness (p〈0.001) and substance P from 65.9±8.1 to 31.3±9.5 (p〈0.02). These results may have applications in the diagnosis and treatment of grass sickness.

    Notes: Abstract The effect of mevastatin and mevinolin on the fusion of L6 myoblasts was studied. Both compounds were potent inhibitors of myoblast fusion at concentrations as low as 0.25 μM, but fusion was restored when the inhibitors were removed. Both compounds resulted in decreased binding of conA and WGA to cell surface oligosaccharides showing they were causing a reduction in N-linked cell surface glycoproteins. There was a reduction in creatine phosphokinase activities in the presence of both compounds showing that they were affecting biochemical differentiation. The presence of both compounds inhibited the incorporation of labeled mannose from GDP-mannose into lipid-sugar and N-linked glycoprotein, but the inhibition was reversed by addition of exogenous dolichol phosphate to the incorporation mixture. The main conclusion from these studies is that mevinolin and mevastatin are inhibiting myoblast fusion by affecting the synthesis of fusogenic cell surface N-linked glycoproteins probably by affecting the synthesis of dolichol phosphate containing oligosaccharides that are required as intermediates in N-linked glycoprotein biosynthesis.

    Description / Table of Contents: Résumé En appliquant des précontraintes plastiques sur des barreaux de traction entaillés, on a créé des cavités au voisinage de particules de carbure dans des aciers traités. Les cavités se forment préférentiellement dans la région centrale du barreau. Dans le cas d'un acier allié trempé et revenu, il s'est produit une formation d'une cavité en feuille tandis que dans le cas d'un acier doux, on ne pouvait voir que des cavités isolées. On a ensuite usiné une petite éprouvette de traction hors de la zone des barreaux entaillés qui était soumise à déformation de manière à ce que la densité maximum de cavité corresponde à la région centrale. Après avoir opéré un recuit sous vide, cette région a été chargée électrolytiquement par de l'hydrogène et a fait montre d'un accroissement dans l'absorption d'hydrogène. Le chargement d'hydrogène a causé une réduction dans la limite d'élasticité, dans la charge de rupture et dans la ductilité des aciers. Des micrographies à balayage électronique des surfaces de rupture de l'acier au C ont révélé une réduction de la dimension des cupules dans la région centrale. Par contre, l'acier faiblement allié sévèrement fragilisé faisait montre d'une rupture sensiblement par clivage. La transition de la rupture ductile en rupture quasi par clivage suit un micromécanisme fonction de la quantité d'hydrogène absorbé dans l'acier. L'absorption d'hydrogène est relié à la taille et à la distribution des cavités associées à la prédéformation.
    Notes: Abstract Cavities were created near carbide particles in heat treated steels by various plastic pre-strains on notched tensile bars. The cavities formed preferentially in the central region of the bar. In the quenched and tempered alloy steel “void sheet” formation occurred; whereas only isolated cavities were seen in the mild steel. Small tensile specimens were then machined from the strained notched bars so that the maximum cavity density corresponded to the central region. After vacuum annealing, this region was electrolytically charged with hydrogen and showed an increase in hydrogen absorption. Hydrogen charging caused a reduction in the yield, ultimate and fracture strength and the ductility of the steels. Scanning electron micrographs of the fracture surfaces of the plain carbon steel revealed a reduction in dimple size in the central region. The severely embrittled low alloy steel showed quasi-cleavage fracture. The fracture transition from ductile to quasi-cleavage micromechanism is a function of the amount of hydrogen absorbed into the steel. Hydrogen absorption is related to the size and distribution of the pre-strain cavities.

    Notes: Mutants of mouse L-cells which are temperature-sensitive for growth have been obtained by using both selective and nonselective isolation procedures on populations treated with the mutagen nitrosoguanidine. Selective isolation was carried out by utilizing a five-day treatment with 3H-TdR and ara-C as selective agents at the nonpermissive temperature. Nonselective isolation was performed by isolating 1400 clones in the absence of selective agents and then testing them for temperature-sensitivity. From this experiment we obtained a minimum estimate of 6 × 10-3 for the frequency of mutants in the mutagentreated population. The mutants were characterized by their plating efficiencies, growth in suspension culture, and uptake of isotopic precursors of DNA, RNA, and protein. A range in phenotypes was observed, and there appeared to be some differences between the mutants obtained by the two types of isolation procedures. In uptake experiments the most marked reductions in the rates of precursor incorporation were seen with 3H-TdR, rather than 3H-UR or 3H-Leu. Different mutant lines showed considerable variation in the rate of cessation of DNA synthesis as well as the time required for termination of cell division. These experiments suggest that both types of isolation procedures are feasible for obtaining temperature-sensitive mutants having a range of phenotypes.

    Notes: Hydroxyurea and guanazole were used as selective agents in tissue culture to obtain independent Chinese hamster ovary cell lines resistant to the cytotoxic effects of hydroxyurea or guanazole. In all cases tested a cell line selected for resistance to one of the antitumor agents exhibited resistance to both drugs. This result supports the view that these two drugs act at a common site.
