    Notes: The effect of hydroxyapatite (HA) coating on cortical bone apposition around press-fit inserted implants and implants surrounded by a gap was investigated. Uncoated and HA-coated titanium implants were inserted in burr holes with three different diameters in the tibia of rabbits. Implantation time was four months. The histological evaluation demonstrated that after four months implantation the interfacial bone reaction appeared to be identical for HA-coated and non-coated implants with various degrees of surgical fit. Although after four months the interface showed the same response, there still might be an initial advantage of the HA-interface with bony tissue.

    Notes: Abstract Composite films made of poly L/D copolymer and hydroxylapatite (HA) as filler material were made in two different volume percentages. As references pure copolymer films and composite films with magnesium oxide (MgO) were prepared to serve as strongly basic model filler material. All these materials were immersed in phosphate buffered saline to study the effect of filler materials on the degradation rate of the copolymer. Filled films showed less molecular weight decrease than the unfilled films, while lactate release was higher for the filled films than for the unfilled films. This effect implies that the filler materials influence the degradation mechanism by preventing the occurrence of the autocatalytic effect of the acidic endgroups, resulting from hydrolysis of the polymer chains. The polymer chains at the surface of the material are less protected by the filler materials, which causes a more rapid degradation of these chains and a higher lactate release. At the same time composites seem to show an erosion type of degradation rather than bulk degradation in unfilled materials.

    Notes: The aim of this in vitro study was to examine the physico-chemical behaviour of hydroxylapatite/poly(l-lactide) (HA/PLLA) composites in solution tests. The polymer PLLA, the composites 30 wt% HA/PLLA (C30) and 50 wt% HA/PLLA (C50) and a one-side HA-coated PLLA (HAcP) were evaluated. Rectangular specimens were incubated in various acellular aqueous buffer solutions [citrate, Gomori's and phosphate-buffered saline (PBS)] up to 24 weeks. Data for cumulative release of calcium, phosphate and l-lactate release in solutions containing C30 or C50 showed linear patterns. Release data for solutions containing HAcP combined with scanning micrographs, X-ray microanalysis and X-ray diffraction patterns of the specimens in time showed that the plasma-sprayed HA coating on PLLA dissolves significantly, progressively in the first weeks and almost completely within the tested period of 24 weeks in vitro. A precipitate of scaly crystallites (calcium phosphates) was observed at the HA coating-PBS interface. After 24 weeks incubation all materials were still above their initial weight, indicating that swelling still exceeded dissolution. Application of C30, C50 and HAcP as implant materials seems interesting where initial stabilization through bone bonding is needed or where the linear release of constituents is a requirement. HAcP has the advantage that the HA coating acts as a hydrolysis barrier and consequently delays the degradation of PLLA in vitro.

    Notes: In in vivo experiments the gap healing of uncoated grooves and grooves coated with HA, HAH (heat treated hydroxylapatite) and FA was studied histologically and histometrically. Cylinders with grooves (0.25 mm, 0.50 mm, 0.75 mm or 1.0 mm depth) were implanted in cortical bone of goats for 3, 6 or 12 weeks. In the uncoated gaps after 3 weeks of implantation no bone ingrowth could be observed. Afer 6 weeks of implantation about 35% bone ingrowth was seen in the uncoated gap independent of gap depth. There was mostly no bone contact with the titanium surface. The presence of a coating enhanced bone ingrowth and initiated more bone contact. After 3 weeks of implantation there was more bone ingrowth and after 6 weeks the bone ingrowth and bone contact was increased, independent of the coating type. HAH showed the highest bone ingrowth. The bone ingrowth started from the periphery of the host bone.

    Notes: Abstract Radio-frequency magnetron sputtering deposition was used to produce calcium phosphate sputter coatings with three different thicknesses (0.1, 1 and 4 μm) on titanium discs. Half of the as-sputtered coatings were subjected to an additional heat treatment for 2 h at 500°C. X-ray diffraction demonstrated that annealing at 500°C changed the amorphous 1 and 4 μm sputtered coatings into an amorphous–crystalline structure, while the amorphous 0.1 μm changed in a crystalline apatite structure. Further, scanning electron microscopy (SEM) inspection demonstrated that annealing of the 1 and 4 μm coatings resulted in the appearance of some cracks. The dissolution behaviour of these Ca–P coatings was determined in a simulated body fluid. It was found that after incubation for 4 weeks the dissolution was determined by the crystallinity of the deposited coating. SEM and Fourier transform infrared evaluation showed that all the heat-treated sputter coating appeared to be stable under the test conditions and a Ca–P precipitate was always deposited on the coating surface. On the other hand, the amorphous 0.1 and 1 μm coatings dissolved completely within 4 weeks, while the amorphous 4 μm coating showed only signs of surface dissolution. Scratch testing demonstrated that there is a linear correlation between the critical load, L c, and the coating thickness. A heat treatment for the CaP-4 coating resulted in an additional decrease in the critical load. On the basis of these findings, we conclude that already a 0.1 μm heat-treated Ca-P sputter coating is of sufficient thickness to show in-vitro adequate bioactive and adhesive properties.

    Notes: Abstract Four different implant materials were installed into the mandibular corner of goats to investigate the trabecular bone response in a mainly unloaded model. The implants were installed using a standardized technique and were left in situ for 12 weeks. One goat had to be sacrificed after surgery because of a broken rib; the other animals healed uneventfully. After sacrifice of the animals, the bone response to the uncoated and the three different Ca–P implants was evaluated histologically and histomorphometrically. Four sections of each implant were evaluated; two were located in the cortical and two in the trabecular bone. Of the 44 retrieved implants, 20 implants appeared to be installed partially in the mandibular canal, as clearly visible on the X-rays. These implants were not used in the histomorphometrical measurements. Histological evaluation showed that the trabecular and cortical bone reactions were similar; there was no significant difference in the percentage of bone contact nor in the amount of bone in contact with the implants. In conclusion this study showed that the mandibular corner is an unsatisfactory model for the installation of implants because of anatomical restrictions. Also, the experiment remained inconclusive about the influence of loading conditions on bone behaviour. Nevertheless, the histological results confirmed the bioactive properties of Ca–P coatings.

    Notes: The aim of this study was to investigate the bone response to three different types of titanium (Ti) alloys and hydroxyapatite (HA) coated titanium alloy by histological and histomorphometrical analysis. Therefore, implants made of these materials were inserted into the tibia of rabbits. Implantation times were 6 and 16 weeks. The histological evaluation included measurement of the amount of bone apposition and analysis of the bone reaction and interface characteristics around the implants. The results demonstrated no marked differences in bony reaction to the different implant materials. In addition, the HA coatings showed loss of thickness. © 1993 John Wiley & Sons, Inc.

    Notes: In order to study the interaction of calcium phosphate coatings with bone tissue, coated titanium cylinders with a standard size were implanted in dog femora. Coatings were made by plasma spraying powders of hydroxylapatite, β-whitlockite, and tetracalciumphosphate particles. The plasma spraying process tuns β-whitlockite into α-TCP. Bone bonding and bone formation were evaluated by mechanical push-out tests and histological observations. Hydroxylapatite and tetracalciumphosphate coatings show an interface strength after 3 months of implantation of 34.3 ± 6.5 MPa and 26.8 ± 3.9 MPa, respectively, while α-TCP and blanco titanium lead to an interface strength of 10.0 ± 3.5 MPa and 9.7 ± 1.3 MPa, respectively. Histological examinations revealed that hydroxylapatite and tetracalciumphosphate give rise to an excellent bone formation, while α-TCP and blanco titanium evoked remodeling and less bone contact.
