    Notes: Summary Restriction sites on the chloroplast genome of Pinus monticola have been mapped, and the gene for the large subunit of ribulose bisphosphate carboxylase/oxygenase, the genes for the photosystem II polypeptides psbA, psbD and psbC, and the 16S and 23S ribosomal RNA genes have been located. The genome lacks the large inverted repeat characteristic of most angiosperms. The gene order is similar to that found in P. radiata. The presence of dispersed repeated sequences is likely. Two structural features, lack of a large inverted repeat and the presence of dispersed repeats, may confer a degree of variability on the genome which will prove useful in studies of population structure.

    Notes: Abstract   A probe for chalcone synthase (CHS) was generated by PCR using chalcone synthase conserved sequences. The cloned PCR product has high similarity to both chalcone synthase and stilbene synthase sequences. The probe was used to examine the organization of chalcone synthase and stilbene synthase genes in Abies procera, Pinus lambertiana, P. monticola, Picea glauca, P. sitchensis, Pseudostuga menziesii, Taxus brevifolia, and Thuja plicata. A large number of hybridizing bands were found in all species except T. plicata which did not cross hybridize. The hybridization patterns are highly polymorphic between the species and are also polymorphic within several of them.

    Notes: Summary Within-species variability of a restriction site in the chloroplast (cp) DNA in Pinus monticola has been surveyed. Frequencies of two variants of the cp genome are significantly different in interior versus coastal populations. Paternal inheritance of the cp genome predominates, though some individuals have both variants of the genome. The presence of heteroplasmic individuals indicates occasional biparental inheritance.

    Notes: Abstract Root induction on in vitro adventitious Pinus monticola Dougl. shoots from mature embryos was improved after 8 weeks co-cultivation with Agrobacterium rhizogenes Conn. strain A4 or the pRi transconjugant strain R1000 as compared to controls, and to naphthaleneacetic acid and indoleacetic acid treatments. An improvement in the number and quality of roots induced was observed on co-cultivated shoots as well as co-cultivated Pinus banksiana Lamb and Larix laricina (DuRoi) K. Koch de-rooted seedlings. Changes in substrate nutrient solution did not influence rooting as much as the presence of Agrobacterium in the rhizosphere. Rooting of cuttings from 3-year-old P. monticola trees was improved in two different seedlots with A. rhizogenes A4 strain.

    Description: Resistance of lodgepole pine (Pinus contorta Dougl. ex Loud. var. latifolia Engelm.) to western gall rust (Endocronartium harknessii (J.P. Moore) Y. Hiratsuka) was compared between 21-year field infection trials and inoculation of 2-month-old seedlings. The seedlings were produced from seed stored since the original field plantings. Virtually all of the seedlings from families identified as susceptible in the field produced galls in the inoculation trial, only 1% of seedlings from these families did not develop galls. Nearly half the seedlings in families classed as resistant in the field trials did not form galls 13 months after inoculation. Only one of these seedlings developed galls in the following 2 years. Seedling inoculation provides a simple early test to assess field resistance to gall rust of lodgepole pine seedlots, and an efficient way to cull at least the most susceptible ones.
