    Notizen: The physical processes involved in the formation of a spheromak are studied. Using one-fluid axisymmetric resistive magnetohydrodynamic equations together with a temperature equation, investigations of the essential physics aspects of the formation specifically for the new Maryland Spheromak (MS) [J. Antoniades, C. Chin-Fatt, A. DeSilva, G. Goldenbaum, R. Hess, and R. Shaw, in Proceedings of the Sixth U. S. Symposium on Compact Torus Research, Princeton, 1984 (Princeton Plasma Physics Laboratory, Princeton, NJ, 1985), p. 65] have been performed. These studies elucidate the role of differential rotation in accelerating the penetration of toroidal field Bφ and in forcing I≡ rBφ to be a flux function. Another important aspect of the formation is the magnetic reconnection that occurs at the X point between the two reversal coils. A strong toroidal current is generated by the reconnection, and the X point is converted into an O point that finally becomes the magnetic axis of the spheromak. In this latter phase of reconnection, the plasma is heated very strongly. Studies have also been done on the effect of the difference in phase and time scales for the reversal and Iz banks for the purpose of optimization. The effect of radiation from carbon and oxygen is examined and found to have very little effect on the proposed formation scheme. The results are also relatively insensitive to changes in the anomalous electron thermal conduction models used.

    Notizen: The effectiveness of myoglobin and its derivatives as photosensitizers in singlet oxygen (1O2) generation was determined by electron paramagnetic resonance (EPR) spectroscopy using a spin trapping technique. A stable nitroxide radical adduct, 2,2,6,6,-tetramethyl-4-piperidone-N-oxyl (TAN), was formed and detected by EPR. Native myoglobin and apomyoglobin exhibited no photosensitizing function, whereas hematin appeared to be a weak sensitizer. Protoporphyrin IX ring showed strong photosensitizing activity. A 1O2 quencher, N,N N′N′-tetramethylethylene diamine (TMEDA) and the combination of TMEDA and a free radical scavenger, butylated hydroxyanisole (BHA) drastically reduced the EPR signal of TAN when protoporphyrin IX solution was illuminated indicating that 1O2 was produced using protoporphyrin IX ring as a sensitizer.

    Notizen: The initiative role of singlet oxygen (1O2) in hydroperoxide production in ground pork and turkey was studied. Ground meats treated with or without various antioxidants and illumination (350 ft-c, 4°C) were monitored for lipid peroxidation by peroxide value (POV). Ground turkey breast meat developed higher (P 〈 0.01) POV during illumination than did thigh meat. In both pork and turkey, light exposure resulted in higher (P 〈 0.01) POV than samples kept in the dark. The prooxidant effect of light was reduced (P 〈 0.01) by a 1O2 quencher (2,2,6,6-tetramethy1-4-piperidone) in turkey meat, and by another 1O2 quencher (N,N,N′,N′-tetramethylethylene diamine, TMEDA), a free radical scavenger (butylated hydroxyanisole, BHA) and TMEDA + BHA in pork.

    Notizen: Electron paramagnetic resonance (EPR) spectroscopy was used to detect the photosensitized generation of singlet oxygen (1O2) by chlorophyll b. Illumination of a chlorophyll b solution containing a spin trap (2,2,6,6-tetramethyl-4-piperidone, TMPD) yielded pronounced EPR signals for TAN (2,2,6,6-tetramethyl-4-piperidone-N-oxyl) suggesting the presence of 1O2. N,N,N′,N′-tetramethylethylene diamine (TMEDA, a 1O2quencher) and a combination of TMEDA with a free radical scavenger (butylated hydroxyanisole, BHA) completely eliminated the TAN signal. BHA alone did not exert this inhibitory effect.

    Notizen: Abstract Temperature distributions were measured during the irradiation of a CO2 laser beam at one end of a rod-shaped specimen and at the centre of a thin plate-shaped specimen. Regarding the measured temperature distributions as one-dimensional and two-dimensional unsteady state heat transfer solutions, CO2 laser beam absorptances were calculated using a modified finite difference method. Temperature dependence of thermal properties, heat loss due to convection and latent heat during melting of the specimen were taken into account in this numerical calculation. Increasing the specimen temperature from room temperature to melting point, absorptances of STS304 stainless steel and SM45C steel were calculated as 8 ∼ 40% and 6 ∼ 41% for the one-dimensional calculation, and as 9.3 ∼ 41% and 5 ∼ 41% for the two-dimensional calculation, respectively. These calculated absortances were very close to theoretical values at relatively low temperature, which were calculated from the electrical resistivities of the specimens. Increasing the temperature of the specimens, absorptances increased considerably due to oxidation of the specimens. Regardless of specimen composition and specimen dimension, both absorptances showed nearly the same value of 41% at their melting points, in which the structures of both metals became amorphous phases.

    Notizen: Abstract Human keratinocytes proliferate and differentiate in an epidermal environment where induction of apoptosis can be triggered by ultraviolet radiation (UVR), activated lymphocytes and cytokines. The purpose of this study was to determine whether keratinocytes were susceptible to apoptosis induced by ionophore, ultra-violet radiation, cytokines or crosslinking of CD95 (Fas/APO-1). In normal human skin exposed to two minimal erythema doses of ultraviolet radiation, suprabasal cells were the first keratinocytes to demonstrate apoptotic nuclei, and by 48 h apoptotic cells were identified throughout the mid to upper epidermis. However, most keratinocytes resisted apoptosis and UVR-induced apoptosis was not observed in basal cells, or in the most differentiated epidermis. Human keratinocytes and keratinocyte cell lines cultured in vitro developed maximal apoptosis 48 h after radiation. Human keratinocytes cultured in full growth factor supplements were resistant to UVR-induced apoptosis compared to keratinocyte cell lines or to a lymphoid cell line (HL60) susceptible to apoptosis. Keratinocyte cell lines were completely resistant to apoptosis induced by interferon-γ, interferon-α, IL-2, IL-6, TNF-α, IL-1Ra, and GM-CSF. A subset of the cells in cultures of keratinocytes and transformed keratinocyte cell lines died by apoptosis in response to anti-Fas, IL-1α and TNF-α plus IFN-γ and ionophore. Second passage freshly isolated human keratinocytes were much more resistant to apoptosis induced by ionophore, anti-Fas and cytokines than were transformed keratinocyte cell lines. Calcium shift to induce differentiation in second-passage keratinocyte cultures made keratino-cytes even more resistant to UVR-induced apoptosis. This parallels the lack of UVR-induced apoptosis observed in the most differentiated keratinocytes in irradiated human skin. Both keratinocytes and kerati-nocyte cell lines express rather low levels of the anti-apoptotic proteins bcl-2 and bcl-x compared to other apoptosis-resistant cell types. The differences between keratinocytes and keratinocyte cell lines in suscepti-bility to apoptosis are not explained by difference in expression of bcl-2 or bcl-x. Finally, withdrawal of growth factors from keratinocytes decreased cell survival following UVR and increased the induction of apoptosis. Inhibition of protein synthesis with cyclo-heximide also made keratinocytes more susceptible to UVR-induced apoptosis, indicating that anti-apop-totic defences in cultured keratinocytes are dependent on active protein synthesis. These experiments show that the strong keratinocyte defences against apoptosis are stratified within the epidermis, and can be altered by differentiation and growth factor withdrawal.
