    Notes: SYNOPSIS. Rabbit antisera to strains of different mating types of Chlamydomonas moewusii and to one strain of C. eugametos were tested against strains of C. reinhardti, C. eugametos, and C. moewussi, and against strains of 2 varieties and 6 mutant types of C. moewusii. Technics of double diffusion, absorption, and immunoelectrophoresis revealed marked serological differences between the sexually incompatible, distinct genetic species C. moewusii and C. reinhardti. Less distinct serological differences were resolved between C. moewusii and the so-called “species”C. eugametos, which is sexually compatible with the former, thus reconfirming the conspecificity between the 2 strains as suggested by Gowans. Marked serological differences were noted between C. moewusii and 2 of its varieties (C. moewusii var. tenuichloris and C. moewusii var. rotunda) which constitute 2 additional genetic species because of sexual incompatibility between themselves and with C. moewusii. Wild types and certain mutants of C. moewusii were compared serologically and could be distinguished on this basis. Strains of different mating types as well as certain mutant strains (e.g., paralyzed flagella, flagella-less, twins and monsters) could be differentiated serologically altho differences were often very subtle. Some antigens were common to organisms tested.

    Notes: Ochromonas danica, a freshwater, planktonic chrysophyte, is capable of sensing the light conditions of its environment. This biflagellate alga has a swelling near the base of the short flagellum and a chloroplastidic stigma in close association with it. A procedure is described for the isolation of this three dimensional flagellar swelling, the presumed photoreceptor. In contrast to an earlier method developed for the isolation of the paraflagellar swelling from Euglena gracilis, the protocol reported here for Ochromonas results in higher yields that should facilitate future biochemical investigations and could open avenues of investigation for the isolation and purification of the presumptive receptor protein. To verify the hypothesis that a rhodopsin-like protein might be present in this alga, we applied a standard extraction procedure successfully used in the identification of retinal. We here report the purification and identification of all-trans retinal in Ochromonas cells by column chromatography, HPLC and GC-MS. Since retinal is the chromophore of rhodopsin-like proteins, this finding may suggest that in these unicellular algae, too, a rhodopsin-like protein could be the photoreceptor pigment.

    Notes: The “mucigenic” or “muciferous” bodies of Peranema trichophorum are further characterized here as unique extrusive organelles, the mucocysts. Intracellular and ejected mucocysts have characteristic shapes that may represent different developmental stages. Mucocysts found near the Golgi apparatus are membrane-bounded, elongate, tubular structures with amorphous contents of low electron density. Subpellicular mucocysts are often aligned with pellicular striae and have dense contents, which are separated by an electron-lucent zone from granular material at the tips. Ejected mucocysts are uniform in structure and consist of an inner tube with helical striations, an outer tube with a diamond-shaped pattern, and a dense middle band. Fine fibrils, visible only after mucocyst discharge, emanate from the tips. Mucocysts may also protrude through the pellicle and discharge mucilaginous materials into the medium. Acid phosphatase activity is localized within the subpellicular mucocysts, suggesting that they may be involved in release of hydrolytic enzymes into the medium.

    Notes: Envelope development in Trachelomonas lefevrei (Deflandre) begins with the production of short, coarse mucilaginous strands, morphologically similar to the mucilage produced by non-enveloped euglenoids. These initial mucilaginous secretions subsequently become impregnated with manganese (Mn) and/or iron (Fe). Continued mucilage secretion and mineralization results in a mature envelope that is characteristic for the species. When these mature envelopes are treated with oxalic acid to remove the Mn and Fe, the envelopes collapse and are composed only of short, coarse mucilaginous strands similar to those present during early stages of development, prior to their mineralization. Brief treatment with 10 mM EDTA renders dark envelopes colorless, and our EM-EDS analyses show that this corresponds to a loss of Mn from the envelope; however, if Fe is present in the envelope, it is unaffected by the brief treatment. The mucilage present during early stages of envelope development and that remaining after complete demineralization is also morphologically similar to the mucilage in the plug at the anterior end of the envelope.

    Notes: Summary The structure of Euglena granulata and Chlamydomonas eugametos has been studied using polarization and electron microscopy, cinematography, and chemical extraction procedures, with the main focus on the structure of the eyespot. The 50–60 granules which form the extrachloroplastic eyespot of E. granulata are large bodies, up to 1200 mμ in diameter. They are found in the cytoplasm near the base of the reservoir and are associated with the parabasal body which contains a large crystal. The eyespot granules are contained within membranes having a unit membrane structure; 2 or 3 are usually present in a single eyespot packet; microtubules are also contained within the packet. The eyespot granules have the structure of a positive spherite and clearly exhibit birefringence; this structure is modified by fixation. The granules of the chloroplastic eyespot of C. eugametos are about 75 mμ in diameter and are contained within the chloroplast in an ordered array. Occasionally, the eyespot contains elongate or helical bodies mixed with the granules. Extraction with organic solvents caused the removal of materials which formed the eyespot granules as well as that of the osmophilic globules in the chloroplasts. Several hypotheses which concern the function of the eyespots in these and other species are discussed in the light of our results. the possible origin and demise of the eyespot granules are also discussed.

    Notes: Abstract Winthout close phylogenetic ties to any of the other algae, the Euglenophyta are a taxonomic enigma. The argument is made here that the euglenophytes have extensive morphological homology with the zooflagellte trypanosomatids, bodonids, andIsonema. A phylogenetic sequence is postulated linking these group; the available data suggest that the euglenophytes had their origin in these zooflagellates and that the introduction of the chloroplast and a phototrophic mode of nutrition was a relatively late step in this evolutionary process.

    Notes: Summary Envelopes of the euglenoid flagellate,Trachelomonas lefevrei may range in color from hyaline to golden to dark brown and may be variously ornamented with spines, papillae and puncta. Elemental composition affects both envelope color and microarchitecture, which may be granular and/or microcrystalline. TEM-EDS microanalyses of granular envelopes or regions show Fe as the predominant element, whereas Mn is the predominant element in envelopes or regions with high concentrations of needle-like microcrystallites. In envelopes composed of both structural components, there is a spatial segregation of Fe and Mn that corresponds to the distribution of the granular and/or microcrystalline entities. Extraction of the needle-like components with leucoberbelin blue indicates that they are Mn oxides. Quantitative elemental analyses show shifting ratios of Mn: Fe that correlate with envelope color and microarchitecture. This is the first direct demonstration both of such a spatial segregation of elements and of the presence of Mn oxides in envelopes ofTrachelomonas.

    Notes: Summary The structure or granules associated with the pellicle strips ofEuglena granulata (Klebs) Schmitz has been studied using light and electron microscopy. Two types of bodies can be distinguished in this association on the basis of their staining reactions, distribution, ultrastructure, and other characteristics. The first type, called pellicle pores, is distributed with regularity along certain pellicle strips; the pores are very uniform in size (about 300 mΜ in diameter) and in content. They consist of an aperture and a compartment, the latter lined by the plasmalemma; in each compartment a dense, osmiophilic body and a series of tubules occur. The structure of the pellicle strips is modified at the points where the pellicle pores are present; the aperture extends to the cell surface in the groove between two pellicle strips. The second type of body, which undergoes vital staining with neutral red, is called a pellicle vacuole. These are variable in size (ca. 1 to 2 microns in diameter) and contain a variety of components including myelin figures, proliferated membranes, and sheets of electrondense material. The structure of these elements as well as their possible functional significance are discussed in some detail.
