    Notes: Ionspray ionization of solutions containing peptides (M) plus alkali metal ions X+ produces [M + X]+, [M + H + X]2+ and [M + 2X]2+ ions. Low-energy collision-induced dissociation of these species demonstrated a marked trend towards decreased strength of the M-X+ interaction with increasing size of X+. For example, the [M +2X]2+ species yield predominantly [M + X]+ plus X+ for X+ = K+, Rb+, Cs+, but rich spectra of fragment ions incorporating X+ when X+ = Li+ or Na+. In general, these fragmentation reactions correspond to those established previously for the corresponding [M + X]+ precursor, including formation of [bi + X + OH]+ and [bi + X - H]+ fragments. As a special case, complexes of Li+ or Na+ with peptides having Arg (but not Lys or other basic residues) at the C-terminus undergo a facile loss of 60 u. The resulting [M + X - 60]+ ions also fragment further to yield [yi′ + X - 60]+ fragments. Characterization of the 60 u neutral loss was undertaken by using tandem mass spectrometry plus hydrogen-deuterium exchange, and a mechanism is proposed to account for the characteristics of the process.

    Notes: Peptides formed as reaction products, of specific hydrolysis of proteins by trypsin, are characterized by a basic residue (Arg or Lys) as the C-terminus, which facilitates formation of bundant [M+2H]2+ ions under electrospray r ionspry conditions. These doubly charged ions readily dissociate upon collisional activation to y" and b fragment ions which are mass complemens of one another. The suggestion tht these fragments are formed by direct charge-separation disociatons must contend with the observation that the y" intensities are generaly appreciably larger than those of their b counterparts. However, it is shown that this can be accouned for by a greater susceptibility f the b ions to undergo further dissociaton to smaler fragments such as immonium ions. In addition no evidence could be found to support alternative mechanisms, including dissociative electron capture, for which equal intensities of the two fragment ion series are not obligatory. Initial protonation at the N-terminus was shown to be required for formation of these [M+2H]2+ ions via its suppression by mono-acetylation at the N-terminus. These findings, and others concerning formation of [y"′]2+ fragments from singly protonated peptides, via charge-site-induced cleavages and intramolecular proto transfers between nitrogen atoms, respectively.

    Notes: Oligodendrocytes, the myelinating cells of the central nervous system, are terminally differentiated cells that originate through asynchronous waves of proliferation and differentiation of precursors present at birth. Withdrawal from cell cycle and onset of differentiation are tightly linked and depend on an intrinsic program modulated by the action of growth factors. p27 plays a central and obligatory role in the initiation of oligodendrocyte differentiation and cessation of proliferation. In this paper, we have characterized the role of modulation of cdk2 and cdk5 kinase activity during the process of oligodendrocyte precursor differentiation. As rat primary oligodendrocytes differentiate in culture there is a fall in cdk2 activity and a rise in cdk5 activity as well as an increase in the cdk inhibitor, p27 protein. The decline in cdk2 activity is not accompanied by a drop in cdk2 protein level, suggesting that it results from inhibition of cdk2 activation rather than decreased protein expression. Taken together, these data suggest that oligodendrocytes may withdraw from the cell cycle at G1-S transition through inactivation of cdk2 activity, possibly initiated by increasing amount of p27, and that cdk5 may have a role until now unrecognized in the differentiation of oligodendrocytes.     Notes: Noncovalent interactions in soybean agglutinin (SBA) were studied on an electrospray ionization (ESI) time-of-flight mass spectrometer constructed recently at the University of Manitoba. The high m/z range and high sensitivity of the instrument together with mild ESI interface conditions turned out to be ideal for detecting this noncovalently bonded tetrameric protein (MW ∼ 116 000 Da) in low charge states (z=23 to 27). By altering the acetonitrile content of the SBA solutions it was shown that the observed SBA tetramers are due to structurally specific noncovalent associations in solution. Octamers and dodecamers (MW ∼ 350 000 Da) were also detected. Information on the quaternary structure of the tetramers was obtained by analyzing the fragment-ion spectrum resulting from the collision-induced dissociation of the tetramer ions.

    Notes: The study of large-scale rearrangements of [b′]2+ ions produced by electrospray ionization of Substance P (Tang et al., Anal. Chem. Vol. 65, p. 2824 (1993)) has been extended to 18 other peptides containing either a lysine or ornithine residue remote from the C-terminus. Evidence for wholesale transfer of one or more residues, from the C-terminus of the [b′]2+ precursor to the ω-amino group of the Lys (or Orn) residue, was observed for 12 of the 18 peptides studied. Unfortunately, no rigorous predictive rules, relating features of the peptide sequence to the propensity to undergo such rearrangements, could be discerned although a significant correlation with presence of a proline residue dose to the lysine or ornithine on the C-terminal side was apparent. The resulting mass-shifts can complicate derivation of peptide sequences from fragment-ion spectra of [M+2H]2+ peptide ions, for example, since the cyclized [b′]2+ ions responsible for the rearrangements are readily formed as intermediate species in the fragmentation mechanisms.

    Description: The PSMD6 variant rs831571 has been identified as a susceptibility locus for type 2 diabetes mellitus (T2DM). This study aimed to investigate the association of this variant with therapeutic effects of oral antidiabetic drugs in Chinese T2DM patients. 209 newly diagnosed T2DM patients were randomly assigned to treatment with repaglinide or rosiglitazone for 48 weeks, and the therapeutic effects were compared. In the rosiglitazone cohort, rs831571 showed significant associations with fasting plasma glucose (FPG), 2-h glucose and decrement of glycated haemoglobin (HbA1c) levels after 24 weeks of treatment (P = 0.0368, 0.0468 and 0.0247, respectively). The C allele was significantly associated with a better attainment of FPG at 24 and 32 weeks (P = 0.0172 and 0.0257, respectively). Survival analyses showed CC homozygotes were more likely to attain a standard FPG level (P = 0.0654). In the repaglinide cohort, rs831571 was significantly associated with decreased HbA1c levels after 24 weeks of treatment, the homeostatic model assessment of insulin resistance and fasting insulin level after 48 weeks of treatment with repaglinide (P = 0.0096, 0235 and 0.0212, respectively). In conclusion, we observed that the PSMD6 variant rs831571 might be associated with the therapeutic effects of rosiglitazone and repaglinide in Chinese T2DM patients.
