    Description: Biogenesis of cytochrome c oxidase (CcO), the terminal enzyme of the mitochondrial respiratory chain, is a complex process facilitated by several assembly factors. Pathogenic mutations were recently reported in one such assembly factor, COA6 , and our previous work linked Coa6 function to mitochondrial copper metabolism and expression of Cox2, a copper-containing subunit of CcO. However, the precise role of Coa6 in Cox2 biogenesis remained unknown. Here we show that yeast Coa6 is an orthologue of human COA6, and like Cox2, is regulated by copper availability, further implicating it in copper delivery to Cox2. In order to place Coa6 in the Cox2 copper delivery pathway, we performed a comprehensive genetic epistasis analysis in the yeast Saccharomyces cerevisiae and found that simultaneous deletion of Coa6 and Sco2, a mitochondrial copper metallochaperone, or Coa6 and Cox12/COX6B, a structural subunit of CcO, completely abrogates Cox2 biogenesis. Unlike Coa6 deficient cells, copper supplementation fails to rescue Cox2 levels of these double mutants. Overexpression of Cox12 or Sco proteins partially rescues the coa6 phenotype, suggesting their overlapping but non-redundant roles in copper delivery to Cox2. These genetic data are strongly corroborated by biochemical studies demonstrating physical interactions between Coa6, Cox2, Cox12 and Sco proteins. Furthermore, we show that patient mutations in Coa6 disrupt Coa6–Cox2 interaction, providing the biochemical basis for disease pathogenesis. Taken together, these results place COA6 in the copper delivery pathway to CcO and, surprisingly, link it to a previously unidentified function of CcO subunit Cox12 in Cox2 biogenesis.

    Description: The eye is a complex organ with highly specialized constituent tissues derived from different primordial cell lineages. The retina, for example, develops from neuroectoderm via the optic vesicle, the corneal epithelium is descended from surface ectoderm, while the iris and collagen-rich stroma of the cornea have a neural crest origin. Recent work with pluripotent stem cells in culture has revealed a previously under-appreciated level of intrinsic cellular self-organization, with a focus on the retina and retinal cells. Moreover, we and others have demonstrated the in vitro induction of a corneal epithelial cell phenotype from pluripotent stem cells. These studies, however, have a single, tissue-specific focus and fail to reflect the complexity of whole eye development. Here we demonstrate the generation from human induced pluripotent stem cells of a self-formed ectodermal autonomous multi-zone (SEAM) of ocular cells. In some respects the concentric SEAM mimics whole-eye development because cell location within different zones is indicative of lineage, spanning the ocular surface ectoderm, lens, neuro-retina, and retinal pigment epithelium. It thus represents a promising resource for new and ongoing studies of ocular morphogenesis.     Notes: A 3D quasiperiodic pattern by projection from an nD lattice can be defined by an orthonormal n × n lattice matrix which produces basis vectors in pattern space with a prescribed arrangement and basis vectors in perpendicular or test space satisfying the quasicrystallographic condition. A 16 × 16 lattice matrix is derived which produces basis vectors in pattern space as a combination or mixing of dually positioned dodecahedral star and icosahedral star. It is shown that the mixed star constitutes a eutactic star. Since the module generated by the mixed eutactic star is totally irrational, patterns generated by projection using the lattice matrix are quasicrystallographic and the equilateral pattern is generated for 1: 1 mixing.

    Notes: A modification of Beenker's pattern is considered that is generated by the transformation matrix obtained by applying the rotation matrix to that for Beenker's pattern. The symmetry of the modified pattern is discussed based on the transformation matrix. It is well known that Beenker's pattern, a two-dimensional eightfold quasiperiodic pattern, is characterized by the transformation matrix, the column vectors of which are the projected basis vectors in four-dimensional cubic lattice space.

    Notes: A new quasiperiodic (QP) pattern with dodecagonal symmetry defined by a self-similar inflation/deflation rule is presented. The pattern consists of three kinds of base tiles, thin rhombus, regular triangle and square, and their self-similar inflation/deflation rule is shown to be derived from a regular zonogon with 12-fold symmetry. The self-similar transformation matrix for this pattern is derived and the quasiperiodicity is discussed.
