    Notizen: The genetic diversity of 66 Phaseolus genotypes was investigated, which included 14 local varieties of Phaseolus vulgaris and nine local varieties of P. coccineus, collected in Marche, central Italy. Their genetic diversity was assessed using three types of molecular marker: inter simple sequence repeats (ISSRs), nuclear gene-tagged simple sequence repeats (SSRs) and chloroplast simple sequence repeats (CpSSRs). Phaseolus vulgaris shows a higher genetic diversity than P. coccineus for the SSRs and CpSSRs, but not for the putative neutral ISSR markers. These data suggest that selection by farmers and adaptation to heterogeneous environments has maintained the diversity in landraces of the common bean. Comparing genetic diversity in Marche with that of the American controls reveals that 71% of the local P. vulgaris varieties in Marche are of Andean origin. The two gene pools of the common bean can be found on the same farm, and there is some evidence of past hybridization events between these two gene pools.

    Notizen: The genetic structure of wild populations of Colletotrichum lindemuthianum was evaluated for virulence and molecular markers. Forty-five isolates were collected from five wild common bean populations located in their South-Andean centre of origin. The five pathogen populations were monomorphic in their ITS regions, but 45 polymorphic markers were identified using RAPDs. Polymorphism for virulence was also observed; 15 pathotypes were characterized on an international set of 12 differentials. A molecular variance analysis (AMOVA) showed that a very large part of the total genetic variation was within populations. Statistical analysis showed that there was a weak though significant differentiation among the five populations for the RAPD and virulence markers. A positive and significant correlation was found between geographic distance and the distances from RAPD and virulence data, suggesting migration between adjacent populations along the Argentinian transect. Our results suggest that the Andean wild isolates of C. lindemuthianum do not reflect all the putative diversity found in the isolates from cultivated common bean.

    Notizen: Abstract  Diversity was analyzed in wild and cultivated Lactuca germplasm using molecular markers derived from resistance genes of the NBS-LRR type. Three molecular markers, one microsatellite marker and two SCAR markers that amplified LRR-encoding regions, were developed from sequences of resistance gene homologs at the main resistance gene cluster in lettuce. Variation for these markers were assessed in germplasm including accessions of cultivated lettuce, Lactuca sativa L. and three wild Lactuca spp., L. serriola L., L. saligna and L. virosa L. Diversity was also studied within and between natural populations of L. serriola from Israel and California; the former is close to the center of diversity for Lactuca spp. while the latter is an area of more recent colonization. Large numbers of haplotypes were detected indicating the presence of numerous resistance genes in wild species. The diversity in haplotypes provided evidence for gene duplication and unequal crossing-over during the evolution of this cluster of resistance genes. However, there was no evidence for duplications and deletions within the LRR-encoding regions studied. The three markers were highly correlated with resistance phenotypes in L. sativa. They were able to discriminate between accessions that had previously been shown to be resistant to all known isolates of Bremia lactucae. Therefore, these markers will be highly informative for the establishment of core collections and marker-aided selection. A hierarchical analysis of the population structure of L. serriola showed that countries, as well as locations, were significantly differentiated. These differences may reflect local founder effects and/or divergent selection.

    Beschreibung: The Nakaseomyces clade consists of a group of six hemiascomyceteous yeasts ( Candida glabrata, Nakaseomyces delphensis, C. nivarensis, C. bracarensis, C. castelli, N. bacillisporus ), phylogenetically close to the yeast Saccharomyces cerevisiae , their representative being the well-known pathogenic yeast C. glabrata . Four species had been previously examined for their carbon assimilation properties and found to have similar properties to S. cerevisiae (repression of respiration in high glucose—i.e. Crabtree positivity—and being a facultative anaerobe). We examined here the complete set of the six species for their carbon metabolic gene content. We also measured different metabolic and life-history traits (glucose consumption rate, population growth rate, carrying capacity, cell size, cell and biomass yield). We observed deviations from the glycolytic gene redundancy observed in S. cerevisiae presumed to be an important property for the Crabtree positivity, especially for the two species C. castelli and N. bacillisporus which frequently have only one gene copy, but different life strategies. Therefore, we show that the decrease in carbon metabolic gene copy cannot be simply associated with a reduction of glucose consumption rate and can be counterbalanced by other beneficial genetic variations.

    Beschreibung: Severe and extreme surface charging on geosynchronous spacecraft is examined through the analysis of 16 years of data from particles detectors on-board the Los Alamos National Laboratory spacecraft. Analysis shows that high spacecraft frame potentials are correlated with 10 to 50 keV electron fluxes, especially when these fluxes exceed 1×10 8 cm -2 s -1 sr -1 . Four criteria have been used to select severe environments: 1) large flux of electrons with energies above 10 keV, 2) large fluxes of electrons with energies below 50 keV and above 200 keV, 3) large flux of electrons with energies below 50 keV and low flux with energies above 200 keV, and 4) long periods of time with a spacecraft potential below - 5kV. They occur preferentially during either geomagnetic storms or intense isolated substorms, during the declining phase of the solar cycle, during equinox seasons and close to midnight local time. The set of anomalies reported in Choi et al. [2011] is concomitant with a new database constructed from these events. The worst-case environments exceed the spacecraft design guidelines by up to a factor of 10 for energies below 10 keV. They are fitted with triple Maxwellian distributions in order to facilitate their use by spacecraft designers as alternative conditions for the assessment of worst-case surface charging.

    Beschreibung: Saccharomyces cerevisiae and S. uvarum are two domesticated species of the Saccharomyces sensu stricto clade that diverged around 100 Ma after whole-genome duplication. Both have retained many duplicated genes associated with glucose fermentation and are characterized by the ability to achieve grape must fermentation. Nevertheless, these two species differ for many other traits, indicating that they underwent different evolutionary histories. To determine how the evolutionary histories of S. cerevisiae and S. uvarum are mirrored on the proteome, we analyzed the genetic variability of the proteomes of domesticated strains of these two species by quantitative mass spectrometry. Overall, 445 proteins were quantified. Massive variations of protein abundances were found, that clearly differentiated the two species. Abundance variations in specific metabolic pathways could be related to phenotypic traits known to discriminate the two species. In addition, proteins encoded by duplicated genes were shown to be differently recruited in each species. Comparing the strain differentiation based on the proteome variability to those based on the phenotypic and genetic variations further revealed that the strains of S. uvarum and some strains of S. cerevisiae displayed similar fermentative performances despite strong proteomic and genomic differences. Altogether, these results indicate that the ability of S. cerevisae and S. uvarum to complete grape must fermentation arose through different evolutionary roads, involving different metabolic pathways and duplicated genes.
