    Notes: Bactericidal/permeability-increasing protein [BPI] is a cationic antimicrobial protein from neutrophils that specifically binds to the surfaces of Gram-negative bacteria via the lipid A component of lipopolysaccharide. To obtain information about the responses of Salmonella typhimurium to cell-surface damage by BPI, two-dimensional gel electrophoresis and N-terminal microsequencing were used to identify proteins that were induced or repressed following BPI treatment. The majority of the affected proteins are involved in central metabolic processes. Upon addition of BPI, the β-subunit of the F1 portion of Escherichia coli ATP synthase was repressed threefold whereas six proteins were induced up to 11-fold. Three of the latter were identified as lipoamide dehydrogenase, enoyl-acyl carrier protein reductase, and the heat-shock protein HtpG. Additionally, a novel protein, BipA, was identified that is induced over sevenfold by BPI; sequence analysis suggests that it belongs to the GTPase superfamily and interacts with ribosomes. A conserved direct-repeat motif is present in the regulatory regions of several BPI-inducible genes, including the bipA gene. Only one of the BPI-responsive proteins was induced when cells were treated with polymyxin B, which also binds to lipid A. We therefore conclude that BPI and polymyxin B affect different global regulatory networks in S. typhimurium even though they bind with high affinity to the same cell-surface component.

    Notes: The probability distribution of the four-phase invariants in the case of single isomorphous replacement has been developed to estimate some individual phases. An example of its application to obtain the phases having special values of 0, π or ±π/2 is given for a known protein structure in space group P212121. The phasing procedure includes the determination of starting phases and an iterative calculation. The initial values of starting phases, which are required by the formula, can be obtained from the estimate of one-phase seminvariants and by specifying the origin and enantiomorph. In addition, the calculations lead to two sets of possible phases for each type of reflection by assigning arbitrarily an initial phase value. The present method provides a possibility for the multisolution technique to increase greatly the number of known phases while keeping the number of the trials quite small.

    Notes: The probability distribution of the four-phase structure invariants (4PSIs) involving four pairs of structure factors is derived by integrating the direct methods with isomorphous replacement (IR). A simple expression of the reliability parameter for 16 types of invariant is given in the case of a native protein and a heavy-atom derivative. Test calculations on a protein and its heavy-atom derivative using experimental diffraction data show that the reliability for 4PSI estimates is comparable with that for the three-phase structure invariants (3PSIs), and that a large-modulus invariants method can be used to improve the accuracy.

    Notes: One of the most talked-about topics among Chinese intellectuals and Western China observers in the past few years has been the six-part television series Heshang (River Elegy). After its first broadcast in June 1988 through the national television network Zhongyang dianshitai (China Central Television, or CCTV in short), the film caused immediate shockwaves throughout China, marking the climax of the so-called ‘wenhua re’ (cultural fever). It was re-played through the CCTV in August, and some local stations aired it even more than twice. twice. Videotapes of the series have been circulating widely among overseas Chinese circles. Total number of viewers was reported to be over 400 million [Lee 1989]. The script appeared in various editions, and much attention has been paid to the ideological dimension of the film.

    Notes: The importance of international trade to the Chinese economy has been growing since the formal approval of the open-door policy at the Third Plenary Session of the 11th Central Committee of the Chinese Communist Party in 1978. However, trade expansion in China faces three constraints. First, there is the theoretical problem that orthodox ideology makes it illegitimate for a socialist country to have a large foreign trade sector. Secondly, there is the institutional problem that the Soviet-type foreign trade mechanism, characterized by a state monopoly of foreign trade, a centralized foreign trade plan, and insulation of domestic from foreign prices, is incapable of handling trade expansion in an efficient manner. Thirdly, there is the economic problem that the lack of competitiveness of domestic goods in the international market limits the country's export and thus import capacity. While the new Chinese leaders are making immense efforts to remove these constraints, this article will focus only on the first. The anti-trade attitude of Communist China is the combined result of China's historical heritage and Marxist ideology. The long history of self-sufficiency in “feudal” China meant that trade was never an imperative economic need. When contact with the west increased in the 19th century, international trade was associated with an influx of opium, an outflow of silver, and a series of unequal treaties. Such an unhappy, early experience of contact with the west has left China sensitive to any increase in international trade.

    Notes: In recent years the concept of civil society has gained scholarly attention world-wide. It has found numerous advocates in the West, such as John Keane who suggested democratizing European socialism by defending the distinction between civil society and the state; Michael Walzer who proposed synthesizing socialist, capitalist and nationalist ideals under the rubric of civil society; and Daniel Bell, who called for a revival of civil society in the United States as a protection against the expanding state bureaucracies. In 1992 alone, at least three books on the subject appeared. In Eastern Europe, proponents of the civil society concept – like Vaclav Havel, George Konrad and Adam Michnik – have been credited with developing an extremely useful theoretical tool for overthrowing Stalinist authoritarianism. A volume consisting of case studies of seven former or present socialist countries found that the notion of civil society is generally applicable to the study of Communist systems, as long as the influence of different cultures and traditions of individual countries are fully acknowledged. The civil society paradigm, despite its basic European orientation, has also been recognized as applicable to the study of developing countries.

    Notes: Summary We have previously described defined mutants of the TraT protein, an outer membrane lipoprotein specified by F-like plasmids, which sensitize Escherichia coli and Salmonella typhimurium to antibiotics that are normally excluded from the cell. In this paper, the isolation, characterization and molecular cloning of suppressors of one such mutant (pDOC40) is reported. The suppressors, which were isolated by selection for vancomycin-resistant revertants, also restored resistance to several hydrophobic antibiotics although there were no detectable changes in lipopolysaccharides (LPS), phospholipids or outer membrane proteins. Three suppressor loci, provisionally designated sip, for suppression of increased permeability, were cloned in cosmids and mapped by a novel approach involving random sequencing of cloned DNA to identify flanking genes with known map positions. Our results indicate that the sipB locus is located in the 11 min region (485–510 kb) whereas sipC and sipD both map to 82 min (3850–3885 kb). Additionally, the previously sequenced nlpA gene was also mapped to the 82 min region. The cloned suppressor loci were specific for the permeability phenotype caused by the mutant R6-5 TraT protein and had no effect on the permeability phenotype caused by a related TraT mutant of S. typhimurium.
