    Notes: Abstract Rapid progress in rice genomics is making it possible to undertake detailed structural and functional comparisons of genes involved in various biological processes among rice and other plant species, such as Arabidopsis. In this review, we summarize the current status of rice genomics. We then select two important areas of research, reproductive development and defense signaling, and compare the functions of rice and orthologous genes in other species involved in these processes. The analysis revealed that apparently orthologous genes can also display divergent functions. Changes in functions and regulation of orthologous genes may represent a basis for diversity among plant species. Such comparative genomics in other plant species will provide important information for future work on the evolution of higher plants.

    Notes: Abstract Aminopterin-resistant cell lines of maize were isolated by two different procedures of callus selection and by plating suspension cultures on drugcontaining medium after mutagen treatment. Efficiencies of different methods of variant selection were compared. Four aminopterin-resistant cell lines were shown to be 10–40 times more resistant than the parental cell line, and they were also resistant to another folate analog, methotrexate. The results suggest that alterations in at least three different cell properties could be responsible for resistance; 1) increased dihydrofolate reductase activity, 2) altered aminopterin sensitivity of dihydrofolate reductase, and 3) reduced drug uptake. One of the resistant cell lines showed more than one alteration, but its resistance proved to be unstable. The results suggest that stable changes which may or may not be of genetic origin and also unstable physiological changes or a combination of both could lead to aminopterin resistance in maize cell cultures.

    Notes: Abstract Two amino-acid auxotrophic cell clones ofHyoscyamus muticus, VA5 (His-) and VIIIB9 (Trp-), isolated in a previous experiment have been characterised quantitatively. Studies of growth in the presence and absence of histidine and tryptophan and an examination of dose-response relationships for the two amino acids confirmed the strict auxotrophy of both cell lines. No revertants to prototrophy were detected in either cell line after more than two years in culture. N-methyl-N′-nitro-nitrosoguanidine did not induce reversion in VA5 cell populations. Wild-type aggregates mixed in various combinations with VA5 and VIIIB9 cells could be recovered after plating in selective conditions. No cross-feeding was detected, either between wild-type and auxotrophic cells or between the auxotrophic lines themselves. Both variants were recloned by protoplast culture. All protoplast-derived clones were auxotrophic. The auxotrophic phenotypes behaved as recessive traits in protoplast-fusion experiments.

    Notes: Abstract Wheat (Triticum aestivum L. cv Chinese Spring) protoplasts were isolated from immature embryos or embryogenic calli (3–4 weeks of culture on MS medium with 32 mg/1 dicamba) and cultured in R2 medium containing 2 mg/1 2,4-D by the nurse culture methods originally developed for rice protoplasts (Kyozuka et al. 1987). Protoplasts isolated from embryogenic calli started to divide within 3–5 days and formed colonies at frequencies up to 2% after 3–4 weeks of culture, while protoplasts isolated from immature embryos formed colonies at much lower frequency (less than 0.1%). Some of these colonies were “embryogenic”, and they appeared at a frequency of approximately 0.5% of colonies formed when callus-derived protoplasts were used. From two of those “embryogenic” colonies, calli were regenerated and albino shoots and roots were obtained.
