    Description: The data set presents more than 32,000 of about 40,000 trees, which were surveyed during several Russian-German expeditions by the North-Eastern Federal University Yakutsk and the Alfred-Wegener-Institute Potsdam in the North-East of the Russian Federation between the years 2011 and 2021. The purpose was to gather information on trees and forests in this region, which was then used to understand tree line migration, stand infilling and natural disturbance and succession processes and to initialize and validate a forest model. Trees are located on more than 160 vegetation plots, each of which has a size of several hundred square meters. For every tree, height was estimated, and the species recorded, while a few individuals were subject to more detailed inventory. This table contains every standing tree of at least 40 cm height that was encountered on the vegetation plots described in the Plot Data Base. It partially overlaps with the dataset “Tree data set from forest inventories in north-eastern Siberia - Tree measurements.” (doi: 10.1594/PANGAEA.949861), which additionally contains details on small or lying deadwood.

    Description: Die Rekonstruktion von Paläoumwelt und Paläoklima ist ein aktuelles Forschungsgebiet sowohl in der Biologie als auch in der Geowissenschaft, von dem man sich Hinweise auf Ursachen und Auswirkungen heutiger Klimaveränderungen erhofft. Häufig verwendete Klimaproxies sind dabei Pollen, Makrofossilien und zunehmend auch DNA-Reste. In dieser Arbeit sollte geprüft werden, ob sich aus pleistozänen sibirischen Torfen mittels der Analyse von aDNA Hinweise auf die zu dieser Zeit vorherrschende Flora und Fauna finden lassen. Zunächst wurde aus drei Torfproben im Alter von 200.000 und 20.000 Jahren alte DNA extrahiert und mittels Polymerase-Kettenreaktion unter Verwendung von spezifischen Primern für Gefäßpflanzen, Pilze, Moose, Säugetiere, Diatomeen und Ringelwürmer das Vorhandensein von alter DNA nachgewiesen. Dabei lieferte die jüngste Probe die besten Ergebnisse. Mittels Klonierung, Sangersequenzierung und Sequenzanalyse konnten die in dieser Probe amplifizierten PCR-Produkte bis auf die taxonomische Ebene der Familie identifiziert werden. Die Ergebnisse für die Pflanzen wurden mit einer an der gleichen Probe durchgeführten Pollenanalyse verglichen, welche ebenfalls eine Bestimmung der Taxa auf Familienebene lieferte. Beide Analysen lieferten ähnliche Taxa, wobei bei der Pollenanalyse eine etwas höhere Diversität erreicht wurde. Neben der Identifizierung von Paläoindikatoren wie Diatomeen, deren Analyse auch mikroskopisch durchführbar wäre, lieferte die DNA-Analyse auch Hinweise auf Organismen wie Säugetiere, Pilze und andere Eukaryoten, die allein anhand von morphologischen Methoden schwer oder gar nicht nachzuweisen sind. Die gefundenen Taxa der untersuchten Eukaryotengruppen deuten auf raue Umweltbedingungen und eine freie, tundrenartige Landschaft hin. Dies ist in Übereinstimmung mit zahlreichen Untersuchungen von Sedimenten, welche aus derselben Region und demselben Ablagerungszeitraum stammen. Es wurde somit gezeigt, dass die Analyse der in sibirischen Torfen enthaltenen DNA sich zum Nachweis verschiedener Eukaryotengruppen eignet, welche teilweise nicht mit mikroskopischen Methoden nachgewiesen werden können und somit detailliertere Hinweise auf die Umweltbedingungen geben können. Um allerdings mehr Informationen über die Vielfalt der taxonomischen Zusammensetzung zu erhalten, sollte in Zukunft neben einer Pollenanalyse die gesamte genomische DNA sequenziert werden. Dadurch entstehen größere Datenmengen, welche mittels bioinformatischer Auswertung genauere Umweltrekonstruktionen zulassen würden.
