Trimethoxyampicillin, Syringa-ampicillin, and TrimethoxyphenylalanylpenicillinIn dieser Arbeit werden folgende Bezeichnungen verwendet: Syringa-ampicillin = α-Aminosyringylpenicillin = α-Amino-4-hydroxy-3,5-dimethoxybenzylpenicillin, TMO = 3,4,5-Trimethoxy.The syntheses of derivatives of ampicillin 〈6-[D-(-)-α-amino-α-pheny1acetamido]penicillanic acid, 1〉 as TMO-ampicillins 8a and 8b, syringa-ampicillin 8c, and TMO-phenylalanyl-penicillin 14 are described. The substituted α-aminophenylacetic acids 5a, 5b, and 12 are prepared by Strecker as well as by the malonic ester synthesis. After protection of the α-amino groups the N-protected amino acids are coupled with 6-aminopenicillanic acid (6-APA) via the procedure of mixed anhydrides; subsequent deblocking is carried out by catalytic hydrogenation. Circular dichroism (CD) curves have been used for the determination of the configurations of the TMO-phenylglycines and the corresponding TMO-ampicillins. The antibacterial activity of the new ampicillins 8a, 8b, 8c and 14 against gram-positive and grarnnegative bacteria is reported.

Die Synthesen von Derivaten des Ampicillins 〈6-[D-(-)-α-Amino-α-phenylacetamido]penicillansäure, 1〉 nämlich der TMO-ampicilline 8a und 8b, des Syringa-ampicillins 8c und des TMO-phenylalanylpenicillins 14 werden beschrieben. Die substituierten a-Aminophenylessigsäuren 5a, 5b und 12 werden nach Strecker bzw. nach der Malonester-Methode synthetisiert. Nach Schutz der α-Aminofunktion werden die N-geschützten Aminosäuren nach dem Verfahren der gemischten Anhydride mit 6-Aminopenicillansäure (6-APS) kondensiert und die Produkte einer hydrogenolytischen N-Demaskierung unterworfen. Zur Konfigurationsbestimmung der TMO-phenylglycine und der entsprechenden TMO-ampicilline wird der Circulardichroismus herangezogen. Die mikrobiologische Wirksamkeit der neuen Ampicilline 8a, 8b, 8c und 14 gegen grampositive und gramnegative Bakterien wird mitgeteilt.
