The Synthesis of Secretin, VI. Purification of Synthetic SecretinThe preparation of the pure heptacosapeptideamideH-His-Ser-Asp-Gly-Thr-Phe-Thr-Ser-Glu-Leu-Ser-Arg-Leu-Arg-Asp-Ser-Ala-Arg-Leu-Gln-1 2 3 4 5 6 7 8 9 10 11 12 13 14 15 16 17 18 19 20Arg-Leu-Leu-Gln-Gly-Leu-Val-NH2 21 22 23 24 25 26 27from crude secretin, which was obtained by demasking of the overall protected heptacosapeptide derivative by means of trifluoroacetic acid, is possible by ionexchange chromatography on SP-Sephadex C-25. The amorphous product being isolated by lyophilisation displays the full biological activity of the natural product. In addition to unobjectionable values of amino acid analysis, the synthetic heptacosapeptideamide behaves identical to natural secretin in chromatographic and electrophoretic tests as well as under the conditions of a trypsin degradation.

Die Reindarstellung des Heptacosapeptid-amidsH-His-Ser-Asp-Gly-Thr-Phe-Thr-Ser-Glu-Leu-Ser-Arg-Leu-Arg-Asp-Ser-Ala-Arg-Leu-Gln-1 2 3 4 5 6 7 8 9 10 11 12 13 14 15 16 17 18 19 20Arg-Leu-Leu-Gln-Gly-Leu-Val-NH2 21 22 23 24 25 26 27aus dem durch Trifluoressigsäure-Demaskierung des allseits geschützten Heptacosapeptid-Derivats erhaltenen Rohsekretin gelingt zweckmäßig durch Ionenaustausch-Chromatographie an SP-Sephadex C-25. Das durch Gefriertrocknung erhaltene amorphe Produkt zeigt volle biologische Aktivität des Naturstoffs; bei einwandfreien Aminosäure-Analysenwerten verhalten sich synthetisches Heptacosapeptid-amid und natürliches Sekretin in den chromatographischen und elektrophoretischen Tests sowie unter den Bedingungen des Trypsin-Abbaus identisch.
