    Notes: Abstract. Sexually mature spawning and prespawning adult and ‘jack’ coho salmon, Oncorhynchus kisutch, were collected during their potamodromous migration from Lakes Ontario, Erie and Michigan in late autumn. In addition, sexually immature spring salmon were collected from Lakes Ontario and Erie. Serum total thyroxine (TT4), serum free thyroxine (FT4), serum total triiodothyronine (TT3) and serum free triiodothyronine (FT3) levels showed marked seasonal variability in salmon from Lakes Ontario and Erie with peak hormone levels evident in the spring fish and lowest levels in the sexually mature adults and ‘jacks’. Seasonal changes in serum triiodothyronine uptake (T3U) values were inverse of those of serum TT4, FT4, TT3 and FT3 levels. When data of the whole collection were combined significant positive correlation coefficients were evident as follows: serum TT3 v. serum TT4, FT4 and TT3; serum TT4 v. serum FT4 and FT3; serum FT4 v. serum FT3, and significant negative correlations as follows: serum T3U v. serum TT3, TT4, FT4 and FT3. When individual groups were examined significant correlations were either absent, or in some instances the reverse of those of whole data. The data provide further evidence of interlake differences in dysfunction of thyroid hormone metabolism in Great Lakes coho salmon; the possible involvement of environmental agents is discussed.

    Notes: A progressive fall in serum thyroid hormone levels was found in Lake Ontario coho salmon, Oncorhynchus kisutch, between May 1978 and January 1979 concomitant with an increase in goitre frequency and thyroid hyperplasia. Despite marked differences in overt goitre frequency in sexually mature adult salmon collected from Lakes Ontario (43.7%), Erie (93.5%), or Michigan (0%), serum thyroid hormone levels and thyroid histology were similar in fish from the three lakes. Two gonadotrop-like basophil cells (‘globular’and‘vacuolar’) were found in the pituitary, particularly in sexually maturing and mature adults and‘jacks’. Whereas in salmon from Lakes Ontario or Michigan, the globular cells were vacuolated in Lake Erie salmon they were not; this observation may be correlated with the absence of obvious secondary sexual characteristics in Lake Erie salmon. Serum free fatty acids serum, triglycerides and cholesterol concentrations in Lake Ontario salmon fell progressively from the condition in fish collected from the Lake in May 1978, to that of fish collected during their potamodrous migration in November 1978 and January 1979.

    Notes: Abstract. Rainbow trout were fed diets contaminated with either PCB (Aroclor 1254, 500 μg/g dry food) or Mirex (500 μg/g dry food). There was no apparent effect of PCB on thyroid histology or on serum thyroid hormone levels. Mire effected significant reductions in serum T4 (P〈0.05) and T3 levels (P〈0.01) without apparent change om the histological appearance of the thyroi. Liver weights and total liver lipid content were significantly (P〈0.001) greater in PCB fed fish compared with controls as was the carcass lipid content (P〈0.01). Mirex had no apparent effect on liver size or liver lipid content but there was a decline in carcass lipid content (P〈0.02) which persisted 25 days after returning Mirex-fed fish to the control diet (P〈0.05).

    Notes: Abstract. Serum thyroxine (T4) and triiodothyroninc (T3) levels were significantly lower in fish fed a high PCB diet (500 mg/kg) in comparison to those fed the control diet. The differences in serum thyroid hormone levels between the PCB-fed group and those fed the control diet were not evident after 14 days food deprivation nor after testosterone-injection. The low PCB diet (50 mg/kg) was without effect on serum T4 and T3 levels regardless of the treatment. Trout fed the highest Mirex diet (50 mg/kg) had T3 levels which were significantly lower and a T4 T3 ratio which T3was still evident after 14 days food deprivation but after testosterone administration the serum T3 levels in the control fish had fallen to levels similar to those in the Mirex-fed group. The low Mires diet (5 mg/kg) was without effect on serum T4 and T3 levels regardless of treatment. Serum T4 and T3 levels in fish fed a mixed PCB (50 mg/kg) and Mirex (5 mg/kg) diet were not significantly different from controls but T3 levels were significantly higher than in control fish after 14 days food deprivation and subsequent testosterone administration. There were no appearence difference in anterior pituitary or thyroid histology between the different groups. The hepatosomatic index (HSI) in PCB-fed trout was significantly larger than in controls in the fed groups, but not after 14 days food deprivation and subsequent testosterone administration. There was a five-fold difference in carcass PCB hioaccumulation between fish fed the two PCB-conlaminated diets, despite a 10-fold difference in dietary levels of the organochlorine. Similarly despite 10-fold differences in dietary Mirex levels in fish fed the 5 and 50 mg/kg and the 50 and 500 mg/kg diets, there were only 4–56–and 1–35-fold increases in carcass Mirex content, respectively. Trout fed the mixed Mirex-PCB diet had PCB levels of only 60.2% of those fed comparable levels of PCB alone (50 mg/kg) but contained similar levels of Mirex to those fed Mirex alone (a mg/kg).

    Notes: The hepatosomatic index (HS1) of female sexually mature coho salmon, Oncorhynchus kisutch, from Lake Superior was significantly higher (P〈0.01) than in males, whereas that of females from Lake Erie was significantly lower (P〈0.05) than in males; there was no significant sex differences in coho salmon from Lakes Ontario or Michigan. Hepatic nuclear areas (HNA) were significantly higher (P〈0.01) in males compared with females in fish collected from Lakes Ontario and Erie, but there was no sex differences in HNA of coho salmon from Lakes Superior or Michigan. In all four lakes, hepatocyte cytoplasmic volume was significantly higher (P〈0.01) in males than in females from the same lake, and the cytoplasmic volume: nuclear volume ratio of males was corresponding significantly higher (P〈 0.05) than in females from the same lake.Two hepatocytic cell types were evident, termed' light and dark cells, based on the electron density of their cytoplasmic matrix. There were considerable intersex and interlake differences in the relative populations of light ‘and dark’ cells in coho salmon from Lakes Ontario, Michigan, Erie and Superior, and also in the hepatocytic content of lipid, glycogen, mitochondria and ribosomal and smooth endoplasmic reticulum.There were no sex differences with regard to in vitro hepatic monodeiodination of L-thyroxine (T4) to triiodo-L-thyronine (T3) in salmon from any of the lakes. However, T4 to T3 conversion was significantly lower (P〈 0.05) in liver homogenates prepared from Lake Ontario coho salmon than in comparable preparations of liver from Lake Superior, Lake Erie, or Lake Michigan salmon.

    Notes: Whereas spring salmon, Oncorhynchus kisutch, from Lake Ontario had no overt thyroid goiters, comparable Lake Erie spring salmon had a 44% goitre incidence. Goitre incidence in sexually precocious ‘jacks’ from Lakes Ontario and Michigan was 43–45% and 100% in ‘jacks’ from Lake Erie.All fish displayed thyroid hyperplasia. The degree of hyperplasia (as indicated by thyroid follicle colloid content, thyroid follicle size and shape and thyroid follicle epithelial cell size) in Lake Ontario fish tended to be greater in the ‘jacks’ than in the spring fish; the reverse was true in Lake Erie fish. Serum total L-thyroxine (T4) concentrations in Lake Ontario spring fish were higher than in any other group. There were no differences in serum T, levels between the groups of ‘jacks’, nor were serum T, levels in ‘jacks’ and spring salmon from Lake Ontario significantly different. Serum total triiodo-thyronine (T) concentrations in Lake Ontario spring salmon were significantly higher than in all other groups and that in Lake Erie spring salmon greater than in all groups of ‘jacks’. Serum T, levels in Lake Ontario ‘jacks’ were higher than in Lake Erie ‘jacks’. Serum free fatty acid (FFA) and cholesterol levels showed significant interlake differences with lower levels present in Lake Erie salmon. Lake Erie ‘jacks’ appeared to have fewer and less well vacuolated pituitary gonadotropic basophils than ‘jacks’ from Lakes Michigan and Ontario. Putative thyrotrop cells appeared to be more numerous and more active in the Lake Erie spring salmon compared with Lake Ontario spring salmon. Prolactin, corticotrop and melanotrop cells appeared less active in spring salmon than in the ‘jacks’.
