    Description / Table of Contents: The main focus of the book is the geological and geophysical interpretation of sedimentary basins along the South, Central and North Atlantic conjugate margins, but concepts derived from physical models, outcrop analogues and present-day margins are also discussed in some chapters. There is an encompassing description of several conjugate margins worldwide, based on recent geophysical and geological datasets. An overview of important aspects related to the geodynamic development and petroleum geology of Atlantic-type sedimentary basins is also included. Several chapters analyse genetic mechanisms and break-up processes associated with rift-phase structures and salt tectonics, providing a full description of conjugate margin basins based on deep seismic profiles and potential field methods.

    Description / Table of Contents: This volume covers the linkage between new transform margin research and increasing transform margin exploration. It offers a critical set of predictive tools via an understanding of the mechanisms involved in the development of play concept elements at transform margins. It ties petroleum systems knowledge to the input coming from research focused on dynamic development, kinematic development, structural architecture and thermal regimes, together with their controlling factors. The volume does this by drawing from geophysical data (bathymetry, seismic, gravity and magnetic studies), structural geology, sedimentology, geochemistry, plate reconstruction and thermo-mechanical numerical modelling.     Description: Many genes familiar from Drosophila development, such as the so-called gap, pair-rule, and segment polarity genes, play important roles in the development of other insects and in many cases appear to be deployed in a similar fashion, despite the fact that Drosophila -like "long germband" development is highly derived and confined to a subset of insect families. Whether or not these similarities extend to the regulatory level is unknown. Identification of regulatory regions beyond the well-studied Drosophila has been challenging as even within the Diptera (flies, including mosquitoes) regulatory sequences have diverged past the point of recognition by standard alignment methods. Here, we demonstrate that methods we previously developed for computational cis -regulatory module (CRM) discovery in Drosophila can be used effectively in highly diverged (250–350 Myr) insect species including Anopheles gambiae , Tribolium castaneum , Apis mellifera , and Nasonia vitripennis . In Drosophila , we have successfully used small sets of known CRMs as "training data" to guide the search for other CRMs with related function. We show here that although species-specific CRM training data do not exist, training sets from Drosophila can facilitate CRM discovery in diverged insects. We validate in vivo over a dozen new CRMs, roughly doubling the number of known CRMs in the four non- Drosophila species. Given the growing wealth of Drosophila CRM annotation, these results suggest that extensive regulatory sequence annotation will be possible in newly sequenced insects without recourse to costly and labor-intensive genome-scale experiments. We develop a new method, Regulus, which computes a probabilistic score of similarity based on binding site composition (despite the absence of nucleotide-level sequence alignment), and demonstrate similarity between functionally related CRMs from orthologous loci. Our work represents an important step toward being able to trace the evolutionary history of gene regulatory networks and defining the mechanisms underlying insect evolution.

    Description: Drug resistance is a consequence of how most modern medicines work. Drugs exert pressure on cells that causes death or the evolution of resistance. Indeed, highly specific drugs are rendered ineffective by a single DNA mutation. In this study, we apply the drug methotrexate, which is widely used in cancer and rheumatoid arthritis, and perform evolution experiments on Baker’s yeast to ask the different ways in which cells become drug resistant. Because of the conserved nature of biological pathways between yeast and man, our results can inform how the same mechanism may operate to render human cells resistant to treatment. Exposure of cells to small molecules and drug therapies imposes a strong selective pressure. As a result, cells rapidly acquire mutations in order to survive. These include resistant variants of the drug target as well as those that modulate drug transport and detoxification. To systematically explore how cells acquire drug resistance in an unbiased manner, rapid cost-effective approaches are required. Methotrexate, as one of the first rationally designed anticancer drugs, has served as a prototypic example of such acquired resistance. Known methotrexate resistance mechanisms include mutations that increase expression of the dihydrofolate reductase (DHFR) target as well as those that maintain function yet reduce the drug’s binding affinity. Recent evidence suggests that target-independent, epistatic mutations can also result in resistance to methotrexate. Currently, however, the relative contribution of such unlinked resistance mutations is not well understood. To address this issue, we took advantage of Saccharomyces cerevisiae as a model eukaryotic system that combined with whole-genome sequencing and a rapid screening methodology, allowed the identification of causative mutations that modulate resistance to methotrexate. We found a recurrent missense mutation in SEC21 (orthologous to human COPG1), which we confirmed in 10 de novo methotrexate-resistant strains. This sec21 allele (S96L) behaves as a recessive, gain-of-function allele, conferring methotrexate resistance that is abrogated by the presence of a wild-type copy of SEC21 . These observations indicate that the Sec21p /COPI transport complex has previously uncharacterized roles in modulating methotrexate stress.

    Notes: Abstract This study involves the integration of information interpreted from data sets such as LandsatTM, Airborne magnetic, geochemical, geological, and ground-based data of Rajpura—Dariba,Rajasthan, India through GIS with the help of (1) Bayesian statistics based on the weights ofevidence method and (2) a fuzzy logic algorithm to derive spatial models to target potentialbase-metal mineralized areas for future exploration. Of the 24 layers considered, five layers(graphite mica schist (GMS), calc-silicate marble (CALC), NE-SW lineament 0–2000 mcorridor (L4-NESW), Cu 200–250 ppm, and Pb 200–250 ppm) have been identified from theBayesian approach on the basis of contrast. Thus, unique conditions were formed based onthe presence and absence of these five map patterns, which are converted to estimate posteriorprobabilities. The final map, based on the same data used to determine the relationships, showsfour classes of potential zones of sulfide mineralization on the basis of posterior probability.In the fuzzy set approach, membership functions of the layers such as CALC, GMS, NE-SWlineament corridor maps, Pb, and Cu geochemical maps have been integrated to obtain thefinal potential map showing four classes of favorability index.
