    Description: Innate immunity provides the first line of defence against invading pathogens and provides important cues for the development of adaptive immunity. Type-2 immunity-responsible for protective immune responses to helminth parasites and the underlying cause of the pathogenesis of allergic asthma-consists of responses dominated by the cardinal type-2 cytokines interleukin (IL)4, IL5 and IL13 (ref. 5). T cells are an important source of these cytokines in adaptive immune responses, but the innate cell sources remain to be comprehensively determined. Here, through the use of novel Il13-eGFP reporter mice, we present the identification and functional characterization of a new innate type-2 immune effector leukocyte that we have named the nuocyte. Nuocytes expand in vivo in response to the type-2-inducing cytokines IL25 and IL33, and represent the predominant early source of IL13 during helminth infection with Nippostrongylus brasiliensis. In the combined absence of IL25 and IL33 signalling, nuocytes fail to expand, resulting in a severe defect in worm expulsion that is rescued by the adoptive transfer of in vitro cultured wild-type, but not IL13-deficient, nuocytes.     Description: Little is known about the integral membrane proteins that participate in the early secretory pathway of mammalian cells. The complementary DNA encoding a 28-kilodalton protein (p28) of the cis-Golgi was cloned and sequenced. The protein was predicted to contain a central coiled-coil domain with a carboxyl-terminal membrane anchor. An in vitro assay for endoplasmic reticulum-Golgi transport was used to show that p28 participates in the docking and fusion stage of this transport event.     Description: The study of cell-population heterogeneity in a range of biological systems, from viruses to bacterial isolates to tumor samples, has been transformed by recent advances in sequencing throughput. While the high-coverage afforded can be used, in principle, to identify very rare variants in a population, existing ad hoc approaches frequently fail to distinguish true variants from sequencing errors. We report a method (LoFreq) that models sequencing run-specific error rates to accurately call variants occurring in 〈0.05% of a population. Using simulated and real datasets (viral, bacterial and human), we show that LoFreq has near-perfect specificity, with significantly improved sensitivity compared with existing methods and can efficiently analyze deep Illumina sequencing datasets without resorting to approximations or heuristics. We also present experimental validation for LoFreq on two different platforms (Fluidigm and Sequenom) and its application to call rare somatic variants from exome sequencing datasets for gastric cancer.     Description: Accurate and precise measurements of the Hubble constant are critical for testing our current standard cosmological model and revealing possibly new physics. With Hubble Space Telescope ( HST ) imaging, each strong gravitational lens system with measured time delays can allow one to determine the Hubble constant with an uncertainty of ~7 per cent. Since HST will not last forever, we explore adaptive-optics (AO) imaging as an alternative that can provide higher angular resolution than HST imaging but has a less stable point spread function (PSF) due to atmospheric distortion. To make AO imaging useful for time-delay-lens cosmography, we develop a method to extract the unknown PSF directly from the imaging of strongly lensed quasars. In a blind test with two mock data sets created with different PSFs, we are able to recover the important cosmological parameters (time-delay distance, external shear, lens-mass profile slope, and total Einstein radius). Our analysis of the Keck AO image of the strong lens system RXJ 1131–1231 shows that the important parameters for cosmography agree with those based on HST imaging and modelling within 1 uncertainties. Most importantly, the constraint on the model time-delay distance by using AO imaging with 0.09 arcsec resolution is tighter by ~50 per cent than the constraint of time-delay distance by using HST imaging with 0.09 arcsec when a power-law mass distribution for the lens system is adopted. Our PSF reconstruction technique is generic and applicable to data sets that have multiple nearby point sources, enabling scientific studies that require high-precision models of the PSF.

    Notes: An existing aetated lagoon treating piggery waste was converted into an aerobic sequencing batch reactor (SBR). After the commissioning period, the SBR plant treated piggery wastewater containing BOD and suspended solids (SS) concentrations of 2881 mg/l and 1419 mg/1 respectively, producing an effluent having an average BOD and SS of 18.7 mg/1 and 12.3 mg/1 respectively. The volumetric loadings on the SBR and the process were similar. However, the SBR process operated at a lower mixed liquor suspended solids (MLSS) concentration and shorter sludge retention period. Although the capital and operating costs of the SBR are higher than for the aerated lagoon, the simplicity of operation, the high BOD and SS removal efficiency, and the small land requirement make this type of process an attractive treatment option, particularly in places where land supply is limited and expensive.

    Notes: Abstract Three hydrographic surveys were carried out in Deep Bay, which is located in the eastern part of Pearl estuary between Shenzhen, the most successful special economic zone of PRC, and Hong Kong. Data on current, size distributions of bottom sediment, suspended solids, and some water quality parameters were obtained. This information is of value for mathematical modeling of tidal circulation and sediment transport in the bay, and also useful in the planning of further development in this area. The sedimentation rate in Deep Bay was estimated by two different approaches, viz. comparison of historical navigation maps and210Pb dating. Information obtained from the maps indicated that average sedimentation rate between 1898 and 1949 was about 8 mm/yr, while a 15 mm/yr figure was obtained from the210Pb dating for recent years.
