    Description: There is a critical need for techniques that directly monitor protein synthesis within cells isolated from normal and diseased tissue. Fibrotic disease, for which there is no drug treatment, is characterized by the overexpression of collagens. Here, we use a bioinformatics approach to identify a pair of glycine and proline isoacceptor tRNAs as being specific for the decoding of collagen mRNAs, leading to development of a FRET-based approach, dicodon monitoring of protein synthesis (DiCoMPS), that directly monitors the synthesis of collagen. DiCoMPS aimed at detecting collagen synthesis will be helpful in identifying novel anti-fibrotic compounds in cells derived from patients with fibrosis of any etiology, and, suitably adapted, should be widely applicable in monitoring the synthesis of other proteins in cells.     Description: To characterize the precursor of mammalian thyrotropin-releasing hormone (TRH), a rat hypothalamic lambda gt11 library was screened with an antiserum directed against a synthetic peptide representing a portion of the rat TRH prohormone. The nucleotide sequence of the immunopositive complementary DNA encoded a protein with a molecular weight of 29,247. This protein contained five copies of the sequence Gln-His-Pro-Gly flanked by paired basic amino acids and could therefore generate five TRH molecules. In addition, potential cleavage sites in the TRH precursor could produce other non-TRH peptides, which may be secreted. In situ hybridization to rat brain sections demonstrated that the pre-proTRH complementary DNA detected neurons concentrated in the parvocellular division of the paraventricular nucleus, the same location as cells detected by immunohistochemistry. These findings indicate that mammalian TRH arises by posttranslational processing of a larger precursor protein.     Description: The formation of the Earth, was mainly from sizeable bodies: perhaps moon sized. Models of interaction among small planetesimals which take into account only close encounters all lead to the formation of moon sized objects, thus leading to several 100 in the inner solar system. Longer term interactions, such as secular resonance sweepings, are needed to get these planetesimals together to form the observed terrestrial bodies. After the accumulation of the Earth, during which core formation certainly occurred, further impacts probably influenced the locations of rifting centers in the system of mantle convection and crustal differentiation. They may have affected craton stabilization by promoting lateral heterogeneity, but had little influence on the key problem of early recycling of sial.

    Description: Reduced Instruction Set Computer (RISC) workstations and Personnel Computers (PC) are very popular tools for office automation, command and control, scientific analysis, database management, and many other applications. However, when using Input/Output (I/O) intensive applications, the RISC workstations and PC's are often overburdened with the tasks of collecting, staging, storing, and distributing data. Also, by using standard high-performance peripherals and storage devices, the I/O function can still be a common bottleneck process. Therefore, the high-performance mass storage system, developed by Loral AeroSys' Independent Research and Development (IR&D) engineers, can offload a RISC workstation of I/O related functions and provide high-performance I/O functions and external interfaces. The high-performance mass storage system has the capabilities to ingest high-speed real-time data, perform signal or image processing, and stage, archive, and distribute the data. This mass storage system uses a hierarchical storage structure, thus reducing the total data storage cost, while maintaining high-I/O performance. The high-performance mass storage system is a network of low-cost parallel processors and storage devices. The nodes in the network have special I/O functions such as: SCSI controller, Ethernet controller, gateway controller, RS232 controller, IEEE488 controller, and digital/analog converter. The nodes are interconnected through high-speed direct memory access links to form a network. The topology of the network is easily reconfigurable to maximize system throughput for various applications. This high-performance mass storage system takes advantage of a 'busless' architecture for maximum expandability. The mass storage system consists of magnetic disks, a WORM optical disk jukebox, and an 8mm helical scan tape to form a hierarchical storage structure. Commonly used files are kept in the magnetic disk for fast retrieval. The optical disks are used as archive media, and the tapes are used as backup media. The storage system is managed by the IEEE mass storage reference model-based UniTree software package. UniTree software will keep track of all files in the system, will automatically migrate the lesser used files to archive media, and will stage the files when needed by the system. The user can access the files without knowledge of their physical location. The high-performance mass storage system developed by Loral AeroSys will significantly boost the system I/O performance and reduce the overall data storage cost. This storage system provides a highly flexible and cost-effective architecture for a variety of applications (e.g., realtime data acquisition with a signal and image processing requirement, long-term data archiving and distribution, and image analysis and enhancement).

    Description: The dynamics of iron, its thermal state and its phase in the accreting Earth probably played a major role in the Earth's early thermal evolution. Plausible impact thermal histories make it possible that pure iron was molten in the accreting Earth after it was about 10% grown. Hence, iron eutectic alloys (FeS, FeO) certainly were. Additionally, the initial temperature of the core is an important constraint on the secular cooling of the early Earth and on the strength of the early geodynamo. Whether iron is solid or molten would influence geochemical equilibria in the upper and lower mantle; the mode of core formation, by spherical or near-spherical blobs, stalk-like instabilities, or something more catastrophic would influence the partitioning of siderophiles between silicate and iron phases. Early descent of iron (during accretion) favors partitioning according to low-pressure phase equilibria, whereas late descent favors higher pressure. The later core formation occurs, the greater the heat pulse, due to the strong dependence of gravitational potential energy on planetary radius. The heat may homogenize the mantle if core formation is global; otherwise, heterogeneity of iron differentiation may leave some of the pre-archean mantle unaffected. The larger the chunks of proto-core (and hence smaller surface/volume ratios) the greater the heterogeneity.

    Description: The minimum mantle viscosity in an earth accreting from planetesimals is estimated. A plausible distribution of planetesimal sizes deposits enough energy to melt the outer nine-tenths of earth's mass; however, vigorous convection keeps temperatures near the solidus. Viscosity is significantly lower than prevails now. The temperature-dependent viscosity provides self-regulation so there is a continuing balance between accretional energy input and heat transfer out. This allows calculation of the minimum viscosity necessary to transfer out heat by a Nu/Ra-number relation. Typical viscosities are 0.1 to a million sq m/sec, lowest at mid-accretion when the mass growth rate is largest. Terrestrial planets are compared, and minimum iron descent times to central lithospheres are calculated.
