Summary A method for the long-term storage of hybridoma cells in liquid nitrogen is described. Hybridoma cells are harvested in logarithmic phase of growth and suspensed in freeze medium containing dimethylsulfoxide. Using a programmable freezer, the cells are frozen very slowly (1°C/min) and then transferred rapidly to liquid nitrogen refrigerators. Consequently, the hybridoma cells can be stored for long periods without substantial loss of viability and with minimal biochemical changes.
