    Description: A single-channel 3 mm interferometer has been developed for plasma density diagnostics in the Sino-UNIted Spherical Tokamak (SUNIST). The extremely compact microwave interferometer utilizes one corrugated feed horn antenna for both emitting and receiving the microwave. The beam path lies on the equatorial plane so the system would not suffer from beam path deflection problems due to the symmetry of the cross section. A focusing lens group and an oblique vacuum window are carefully designed to boost the signal to noise ratio, which allows this system to show good performance even with the small-diameter central column itself as a reflector, without a concave mirror. The whole system discards the reference leg for maximum compactness, which is particularly suitable for the small-sized tokamak. An auto-correcting algorithm is developed to calculate the phase evolution, and the result displays good phase stability of the whole system. The intermediate frequency is adjustable and can reach its full potential of 2 MHz for best temporal resolution. Multiple measurements during ohmic discharges proved the interferometer’s capability to track typical density fluctuations in SUNIST, which enables this system to be utilized in the study of MHD activities.

    Description: An attempt was done to improve the quality of deformation texture predictions by statistical models through the introduction of "clusters" of N grains thus defining a third, intermediate length scale. The interaction between each cluster and the macroscopic length scale is of the Taylor type, whereas inside each cluster a VPSC scheme is used. Predictions of cold rolling deformation textures were quantitatively compared with experimental results for a steel alloy. The results are encouraging.

    Description: Motivation: Identifying drug–target protein interaction is a crucial step in the process of drug research and development. Wet-lab experiment are laborious, time-consuming and expensive. Hence, there is a strong demand for the development of a novel theoretical method to identify potential interaction between drug and target protein. Results: We use all known proteins and drugs to construct a nodes- and edges-weighted biological relevant interactome network. On the basis of the ‘guilt-by-association’ principle, novel network topology features are proposed to characterize interaction pairs and random forest algorithm is employed to identify potential drug–protein interaction. Accuracy of 92.53% derived from the 10-fold cross-validation is about 10% higher than that of the existing method. We identify 2272 potential drug–target interactions, some of which are associated with diseases, such as Torg-Winchester syndrome and rhabdomyosarcoma. The proposed method can not only accurately predict the interaction between drug molecule and target protein, but also help disease treatment and drug discovery.     Description: 〈p〉〈i〉Artemisia annua〈/i〉 produces the valuable medicinal component, artemisinin, which is a sesquiterpene lactone widely used in malaria treatment. AaORA, a homolog of CrORCA3, which is involved in activating terpenoid indole alkaloid biosynthesis in 〈i〉Catharanthus roseus〈/i〉, is a jasmonate (JA)–responsive and trichome-specific APETALA2/ETHYLENE-RESPONSE FACTOR that plays a pivotal role in artemisinin biosynthesis. However, the JA signaling mechanism underlying AaORA-mediated artemisinin biosynthesis remains enigmatic. Here, we report that AaORA forms a transcriptional activator complex with AaTCP14 (TEOSINTE BRANCHED 1/CYCLOIDEA/PROLIFERATING CELL FACTOR 14), which is also predominantly expressed in trichomes. AaORA and AaTCP14 synergistically bind to and activate the promoters of two genes, 〈i〉double bond reductase 2〈/i〉 (〈i〉DBR2〈/i〉) and 〈i〉aldehyde dehydrogenase 1〈/i〉 (〈i〉ALDH1〈/i〉), both of which encode enzymes vital for artemisinin biosynthesis. AaJAZ8, a repressor of the JA signaling pathway, interacts with both AaTCP14 and AaORA and represses the ability of the AaTCP14-AaORA complex to activate the 〈i〉DBR2〈/i〉 promoter. JA treatment induces AaJAZ8 degradation, allowing the AaTCP14-AaORA complex to subsequently activate the expression of 〈i〉DBR2〈/i〉, which is essential for artemisinin biosynthesis. These data suggest that JA activation of the AaTCP14-AaORA complex regulates artemisinin biosynthesis. Together, our findings reveal a novel artemisinin biosynthetic pathway regulatory network and provide new insight into how specialized metabolism is modulated by the JA signaling pathway in plants.〈/p〉

    Description: Obesity is causally linked to osteoarthritis, with the mechanism being not fully elucidated. MicroRNAs are pivotal regulators of various diseases in multiple tissues, including inflammation in the chondrocytes. Here we for the first time identified the expression of miR-26a in mouse chondrocytes. Decreased level of miR-26a was correlated to increased chronic inflammation in the chondrocytes and circulation in obese mouse model. Mechanistically, we demonstrated that miR-26a attenuated saturated free fatty acid-induced activation of NF-κB (p65) and production of proinflammatory cytokines in chondrocytes. Meanwhile, NF-κB (p65) also suppressed miR-26a production by directly binding to a predicted NF-κB binding element in the promoter region of miR-26a. Finally, we observed a negative correlation between NF-κB and miR-26a in human patients with osteoarthritis. Thus, we identified a reciprocal inhibition between miR-26a and NF-κB downstream of free fatty acid signaling in obesity-related chondrocytes. Our findings provide a potential mechanism linking obesity to cartilage inflammation.

    Description: Ta 2 O 5 /TaO x heterostructure has become a leading oxide layer in memory cells and/or a bidirectional selector for resistive random access memory (RRAM). Although atomic layer deposition (ALD) was found to be uniquely suitable for depositing uniform and conformal films on complex topographies, it is hard to use ALD to grow suboxide TaO x layer. In this study, tantalum oxide films with a composition of Ta 2 O 5 were grown by ALD. Using Ar + ion irradiation, the suboxide was formed in the top layer of Ta 2 O 5 films by observing the Ta core level shift toward lower binding energy with angle-resolved X-ray photoelectron spectroscopy. By controlling the energy and irradiation time of an Ar + ion beam, Ta 2 O 5 /TaO x heterostructure can be reliably produced on ALD films, which provides a way to fabricate the critical switching layers of RRAM.

    Description: Two polycrystalline plasticity models, namely the ALAMEL model and the viscoplastic self-consistent (VPSC) model were used to study the effect of the grain shape on the evolution of plastic anisotropy. The latter was characterized by the q-value in this paper. The two models both predict the evolution of grain shape and texture and then can predict the effect of the initial grain shape on the texture. The final anisotropy of the materials was also assessed with the help of these models. Data of tensile tests in three directions for three low carbon steel sheets with either equiaxed or a pancake grain shape were used to evaluate the model predictions. Both models give the same trend of the grain shape effect on the evolution of the q-values. Results demonstrate that the effect of the grain shape can be clearly reflected in the x-value distribution as defined and discussed in this paper.     Description: Atrial septal defects (ASDs) are a common human congenital heart disease (CHD) that can be induced by genetic abnormalities. Our previous studies have demonstrated a genetic interaction between Tbx5 and Osr1 in the second heart field (SHF) for atrial septation. We hypothesized that Osr1 and Tbx5 share a common signaling networking and downstream targets for atrial septation. To identify this molecular networks, we acquired the RNA-Seq transcriptome data from the posterior SHF of wild-type , Tbx5 +/ – , Osr1 +/– , Osr1 –/– and Tbx5 +/– /Osr1 +/– mutant embryos. Gene set analysis was used to identify the Kyoto Encyclopedia of Genes and Genomes pathways that were affected by the doses of Tbx5 and Osr1 . A gene network module involving Tbx5 and Osr1 was identified using a non-parametric distance metric, distance correlation. A subset of 10 core genes and gene–gene interactions in the network module were validated by gene expression alterations in posterior second heart field (pSHF) of Tbx5 and Osr1 transgenic mouse embryos, a time-course gene expression change during P19CL6 cell differentiation. Pcsk6 was one of the network module genes that were linked to Tbx5. We validated the direct regulation of Tbx5 on Pcsk6 using immunohistochemical staining of pSHF, ChIP-quantitative polymerase chain reaction and luciferase reporter assay. Importantly, we identified Pcsk6 as a novel gene associated with ASD via a human genotyping study of an ASD family. In summary, our study implicated a gene network involving Tbx5 , Osr1 and Pcsk6 interaction in SHF for atrial septation, providing a molecular framework for understanding the role of Tbx5 in CHD ontogeny.
