Zusammenfassung Bestimmungsmethoden, die für die Stabilitätsprüfung von pharmazeutischen Darreichungsformen angewendet werden, müssen spezifisch sein. Dazu eignen sich vor allem chromatographische Methoden, und zwar speziell die Dünnschicht- und die Flüssigkeits-Chromatographie. Die Dünnschicht-Chromatographie (TLC) hat durch die Einführung methodischer Varianten, wie der circularen bzw. anticircularen Entwicklung eine Bereicherung erfahren. Gerade die anticirculare Entwicklungstechnik eröffnet der Dünnschicht-Chromatographie einen größeren Anwendungsbereich, da sie reproduzierbarer, schneller und kostengünstiger als die anderen Dünnschicht-Chromatographietechniken arbeitet. Die Flüssigkeits-Chromatographie (HPLC) wird in der Stabilitätsprüfung zum einen dann eingesetzt, wenn sehr hohe Anforderungen an die Reproduzierbarkeit des Verfahrens gestellt werden, ferner bei der simultanen Bestimmung mehrerer Wirkstoffe in einem Produkt oder bei der gleichzeitigen Bestimmung von Wirkstoffen und ihren Zersetzungsprodukten. Zusätzlich kann die Flüssigkeits-Chromatographie zur Strukturaufklärung von Zersetzungsprodukten benutzt werden; durch Kopplung mit MS- und NMR-Geräten können zukünftig im „on-line“-Verfahren Spektren aufgenommen werden, ohne daß zuvor eine aufwendige Isolierung der Substanz vorgenommen werden muß. Anhand einiger Beispiele wird gezeigt, daß bei der Stabilitätsprüfung pharmazeutischer Darreichungsformen beide chromatographischen Verfahren ihre Existenzberechtigung haben und sich auch ergänzen können. An einem Herzglykosid wird schließlich demonstriert, daß die Dünnschicht-Chromatographie auch als Pilotmethode zur Optimierung der mobilen Phase für die Säulenflüssigkeits-Chromatographie angewandt werden kann.

Summary Methods of determination employed for stability tests of pharmaceutical products must be specific if they are to be acceptable for this purpose. Particularly suitable are chromatographic methods, especially those involving thin-layer and liquid chromatography. The scope of thin-layer chromatography (TLC) has been widened through the introduction of new techniques such as the circular and anti-circular development of plates. Especially the anti-circular development techniques opens up a wider field of application for thin-layer techniques. Liquid chromatography (HPLC) is the method of choice in stability testing when high demands are made on the reproducibility of the method; moreover, when several active ingredients of a product are to be determined simultaneously or when the simultaneous determination of an active ingredient and its decomposition products is required. Furthermore, liquid chromatography can be employed for structure elucidation of decomposition products; coupling with MS and NMR instruments will, in the future, enable the on-line aquisition of spectra without the necessity of an extensive isolation of the substance in question. It will be shown by means of examples that in the field of stability testing of pharmaceutical products, both chromatographic procedures have the right to exist and that they can also complement one another. Finally, it will also be demonstrated in the case of a cardiac glycoside that thin-layer chromatography can also be used as a pilot method for optimising the mobile phase for use in liquid column chromatography.
