    Notes: The physiology of hyperhydricity in relation to oxidative stress, mineral nutrients, antioxidant enzymes and ethylene has been studied in three micropropagated carnation cultivars under experimentally induced hyperhydricity. A marked increase in Fe content in comparison with normal tissues was observed in the hyperhydric tissues from the three cultivars. The levels of ethylene, solute leakage and malondialdehyde content were also significantly higher in the hyperhydric tissues. In relation to the time course of H2O2 production measured by fluorescence quenching, a similar trend could be observed for the three cultivars, with a clear increase in the generation of hydrogen peroxide in hyperhydric tissues. The activities of all the antioxidative enzymes studied, except lipoxygenase, were higher in the hyperhydric shoots. Phenylalanine ammonia-lyase (PAL) showed a significant decrease in activity in the hyperhydric tissues in comparison with the controls for the three cultivars. Soluble guaiacol peroxidase had a strong increase in activity in hyperhydric shoots of the three cultivars. These results provide, for the first time, direct evidence of H2O2 generation in hyperhydric tissues, characterize the response of the antioxidant system to an oxidative stress during hyperhydricity in carnation leaves and point to the accumulation of toxic forms of oxygen as the inducer of some of the abnormalities observed.

    Notes: Abstract Electron microscopy observations of salt-tolerant embrogenic calli of Citrus limon [(L.) Burm. f.] showed several changes in cell ultrastructure when compared with control calli. Both types of calli comprised clusters of meristematic cells, but salt-tolerant calli had several structural differences: thick cell walls, ring-shaped mitochondria, an increased content of lipid bodies, microbodies and parallel accumulation of rough endoplasmatic reticulum. These structural features seem to be related with salt tolerance in Citrus limon cells.

    Notes: Abstract Embryogenic callus cultures of lemon (Citrus limon L. Burm f. cv Verna), were selected for resistance to salt stress (170 mM NaCl). Inorganic analysis showed that selected callus accumulated more Na+ and Cl- ions than the non-selected one. Moreover, the salt-tolerant C. limon callus exhibited an increase in the activity of antioxidant enzymes involved in oxygen metabolism, with the induction of a new superoxide dismutase isozyme and an increase of the peroxidase activity while the catalase activity was unchanged. Proline and total sugar, mainly sucrose, concentrations increases significantly in salt-tolerant cells as compared to control cells. On the other hand, the selected cell line also showed an increase in choline and glycine betaine, but to lesser extent.

    Notes: Abstract Uptake and metabolism of sucrose in micropropagatedRosa multiflora using the double layer technique was investigated. In the multiplication as well as the root induction stage, hydrolysis of sucrose in the culture medium was observed. A mathematical model was developed to quantify sucrose hydrolysis and the uptake of sucrose, glucose and fructose, based on the time series for the different sugars in the culture medium. These data were linked to a study of the sugar metabolism in the microshoots. After 48 h of incubation on14C-[U]-glucose containing medium, the incorporated label was mainly detected in the ethanol soluble fraction; within this fraction sucrose was the most important compound. This indicates a significant re-synthesis of sucrose in the plant material after the uptake of hexose. To assess the extent that different enzymes of sucrose metabolism (invertases, sucrose synthase and sucrose-P-synthase) were involved, their activity in different plant parts (of final stage III microshoots) were assayed. A decreasing gradient for sucrose metabolising enzymes from the roots toward the leaves gave a good indication of how the different tissues depend on sucrose absorbed from the medium.

    Notes: Abstract Catalase and superoxide dismutase (SOD) activities were followed in leaves during ex vitro acclimatization and growth of micropropagated Spathiphyllum floribundum Schott ‘Petite’ and Calathea louisae Gagnep ‘Maui Queen’. During acclimatization of both plants catalase activity increased, reaching a maximum 4 weeks after transplantation, while total superoxide dismutase activity increased with plant growth reaching a maximum in the 24th week. Variations in the pattern of catalase and SOD isoforms were observed; a second Mn-SOD band appeared in Spathiphyllum 12 weeks after transfer from in vitro, while in Calathea plants an additional Mn-SOD form was present from the second until the fourth week after transplantation. The observed changes reflect the plants' capacity to develop antioxidant mechanisms during acclimatization. These findings indicate that the adaptation of micropropagated plants to ex vitro conditions is more extended in time than generally accepted.

    Description: In this study, the functioning of antioxidant metabolism and photosynthesis efficiency during the acclimatisation of Stevia rebaudiana plants to ex vitro conditions was determined. A high percentage of acclimatised plants (93.3%) was obtained after four weeks. According to the extent of lipid peroxidation, an oxidative stress occurred during the first hours of acclimatisation. A lower activity of monodehydroascorbate reductase (MDHAR) than dehydroascorbate reductase (DHAR) was observed after 2 days of acclimatisation. However, after 7 days of acclimatisation, stevia plants activated the MDHAR route to recycle ascorbate, which is much more efficient energetically than the DHAR route. Superoxide dismutase and catalase activities showed a peak of activity after 7 days of acclimatisation, suggesting a protection against reactive oxygen species. Peroxidase activity increased about 2-fold after 2 days of acclimatisation and remained high until day 14, probably linked to the cell wall stiffening and the lignification processes. In addition, a progressive increase in the photochemical quenching parameters and the electronic transport rate was observed, coupled with a decrease in the non-photochemical quenching parameters, which indicate a progressive photosynthetic efficiency during this process. Taken together, antioxidant enzymes, lipid peroxidation, and chlorophyll fluorescence are proven as suitable tools for the physiological state evaluation of micropropagated plants during acclimatisation to ex vitro conditions.
