    Notes: Wall-to-wall linkage may help maintain cell integrity and polarity, and focus mechanical stress from wall to mech-anotransductive ion channels within the plasm a lemma. When cells of onion bulb scale epidermis shrink during plasmolysis with CaCl2, the plasmalemma remains attached to the cell wall by Hechtian strands which we hypothesize might possibly be drawn out from linkages fulfilling the above functions. We show that at least many of the attachment loci are independent of the plasmodesmata. A priori, wall glycoproteins seem good candidates for the wall-to-membrane linkers; therefore, we investigated the distribution in wall and plasmalemma of antigen recognized by antibody to hydroxyproline-rich glycoprotein (HRGP). Using fluorescent secondary antibodies, we showed that polyclonal antibodies prepared against wall HRGP from soybean bind to the onion walls (following mild depectination), but also bind to the plasmalemma after the wall is enzymatically digested. The distribution of the antibodies is punctate. On the plasmalemma, the points tend to be scattered more or less uniformly, but can cluster at termini of large streaming strands (which rarely form in wall-constrained cells.) These streaming strands can be seen to exert tension on the membrane. We hypothesize that (1) the antigen on the surface of the protoplast may correspond to the antigen in the walls, (2) such antigen may be responsible for adhesion of membrane to wall at the linkage sites visualized by CaCl2 plasmolysis, and (3) the linkage sites may be transmembrane proteins to which cytoskeleton can attach at the inner surface.

    Description: Introduction. The higher plant is a heterogeneous, mechanically prestressed structure continually subject to shifting forces. When a cell grows in a plant at gravitropic equilibrium, it must create localized maxima of shear in walls of neighboring cells. Such mechanical stress and strain are likely detected in a variety of ways. However, tension-sensitive ion channels are of particular interest because it appears that they are elaborately evolved for sensory function. We hypothesize that 1) the patchy patterns of high shear are focused via wall-to-membrane linkers onto the plasma membrane, where 2) they are translated by mechanosensory cation channels into corresponding patterns of high cytosolic Ca2+, which 3) initiate local enhancement of wall expansion. Further, we hypothesize that the local promotion of enhancement is achieved at least in part by local intensification of auxin transport across the plasma membrane. By implication, when an organ is asymmetrically pressed, rubbed, or bent or when it is displaced in the gravitational field, the net asymmetry of shear stress occurring across the organ would lead to asymmetric redistribution of auxin and corrective asymmetric growth. We shall describe a representative mechanosensitive Ca(2+) -selective cation channel (MCaC) with susceptibilities to xenobiotics implicating it as a force transducer in thigmo- and gravitropism. Then, we shall consider whether a putative wall-to-membrane linker (WML) could be a key feature of the molecular architecture permitting the stress distributed in the wall system to be focused on the channels.

    Description: In shoots of many plants, of which tomato (Lycopersicon esculentum Mill.) is an example, ethylene production is substantially increased during gravitropism. As a first step toward elucidating the role of ethylene in gravitropism, detailed time courses of ethylene production in isolated hypocotyl segments and whole plants were measured for gravistimulated and upright tomato seedlings. In the first experiment, seedlings were set upright or laid horizontal and then, at 15 min intervals, sets of hypocotyls were excised and sealed into gas tight vials. A steady long term rise in ethylene production begins after 15 min gravistimulation. It is possible that this increase is a consequence of the accumulation of indoleacetic acid (IAA) in the lower tissue of the hypocotyle. In a second kind of experiment, whole seedlings were enclosed in sealed chambers and air samples were withdrawn at 5 min intervals. Stimulated seedlings produced more ethylene than controls during the first 5 min interval, but not appreciably more during the second. This suggests the possibility that the ethylene production induced during the first 5 min occurs immediately rather than after a lag, and thus much too soon to be controlled by redistribution of IAA.

    Description: Decapitated veritcal stem segments were observed for 45 min, noting the number of transients mu V for each 5-min interval. Results for two sets of stem segments are plotted. The average frequencies were 0.72 + or -0.05 and 0.66 + or - 0.04 per 5 min. One set of segments was swiveled gently to the horizontal position; then, recording continued for another 90 min. Transients continued in the vertical controls at a closely similar average rate 0.78 + or - 0.04 per 5 min. However, after the first 5-min interval the horizontally placed plants exhibited transients at an increased average rate of 1.08 + or - 0.04 per 5 min. The frequency of transients also increased following horizontal placement of intact shoots. However, it appears that this increase had two components: one due to gravity reception, and one due to the redistribution of indolacetic acid (IAA) believed to mediate gravitropic curvature (or, more specifically, to the increase of IAA in the lower tissue.

    Description: The mechanism of gravitropic gravity reception by seedling shoots was examined. Gravitropism is mediated by lateral migration of the auxin IAA. The 10 min. lag for induction of growth by IAA helps to explain auxin action. Evidences for the rapicity of gravitropism are examined.
