    Description: This volume explores the meaning of republicanism in contemporary Ireland. While this has often been identified simply with nationalism, the book examines the connections, comparisons and contrasts between Irish republicanism and other strands of republican politics: the ideology and practice of official French republicanism, the broader European and American civic republican tradition and the contemporary revival of this tradition of citizenship. Academics from different disciplines, along with statesmen and politicians from different political perspectives, are brought together to examine the relationship of historical and contemporary Irish republicanism to the wider republican theoretical tradition. The book analyses political positions among those parties describing themselves as republican in Ireland in the twenty-first century and examines the possible relevance of the ideas of the broader republican tradition for future politics in Ireland.

    Notes: Design considerations for areal densities beyond 20 Gb/in.2 are discussed, and in particular a demonstration at 24.8 Gb/in.2 is shown. The demonstrations used a single combined write and read head (Merge, Notched head). In this article we will discuss the tradeoffs in kilobytes per inch (KBPI) and tracks-per-inch in obtaining areal densities at 25 Gb/in.2 densities. The KBPI is limited by both the total signal-to-noise ratio and the nonlinear-transition shift. Simple estimates of the increase in channel performance from class IV partial response (PR4), extended partial response 4 (EPR4), and modified EEPR4, as well as the addition of codes which eliminate bit shift and tribit errors are discussed as large contributors to the increase in the areal density. © 2000 American Institute of Physics.

    Notes: : This paper reviews the processes that occurred during an application of the Metropolitan Water District (MWD)-MAIN water use forecasting system for the City of Salinas, California. The review includes an analysis of sources of available data, methods for estimating input data, calibration, and verification of the MWD-MAIN System, and an evaluation of the reliability of system output. We found that inexperienced users can have difficulty understanding the level of skill, knowledge, and amount of data that are required to produce reliable forecasts.Some of the issues associated with application of the MWD-MAIN System include the following:〈list xml:id="l1" style="custom"〉•All input data needed for accurate forecasts simply are not available for many cities and towns.•The data requirements are more extensive than many users anticipate.•Substantial requirements for manipulation of input data produces opportunity for error that creates major time demands in troubleshooting.•Calibration and verification for specific uses can be substantially more difficult than is readily apparent from the guidance manual.•Independent validity checks need to be done to validate system output.•If specified calibrating procedures do not produce reasonable results, reestimating slope coefficients is an option, but this requires resources and expertise that can easily exceed the limits of most users.These are problems typical of most complex models. Reviews such as this can help users to appreciate the level of data required, and to use the MWD-MAIN System in a more effective and efficient manner.

    Notes: : The power of computers has increased in recent decades, and one might expect improved management to result because decisions can be made with understanding available only via models. However, there is potential for quite the opposite: poor decisions due to unrealistic model output generated by users without access to appropriate training in the use of models. We discuss and, by reference to water demand models (IWR-MAIN, MWD-MAIN), illustrate three areas in which unintended errors of judgment by untrained personnel may cause difficulty:〈list xml:id="l1" style="custom"〉*Attributes of management models; if output from any type of model has no measure of confidence, then results may be over- or undervalued*Input data; with complex models, problems here typically will be difficult to detect.*Calibration and history-matching (verification); if these steps or data are combined, then users should be less trustful of model output than otherwise.Because all models have weaknesses and because there always is uncertainty about output from any model, we end with suggestions for coping with complex models. Monitoring programs play a central role in such efforts because they can identify discrepancies between model predictions and actual events and because they can ensure time is available to develop solutions for problems unanticipated in the modeling effort.

    Notes: Proaerolysin is an extracellular dimeric protein that is secreted across the inner and outer membranes of Aeromonas spp. in separate steps. To investigate the role of protein folding in the second step, one or more cysteine residues were introduced and the mutant proaerolysins were expressed in Aeromonas hydrophila and Aeromonas salmonicida, as well as Vibrio cholerae. Replacing Met-41 with Cys resulted in expression of a protein that could form a dimer in which the monomers were linked together by a disulphide bridge. A double mutant was also made, in which Gly-202 and Ile-445 were replaced with cysteine in order to allow the formation of an intrachain disulphide bridge when the molecule was correctly folded. The M41C covalent dimer and G202C/I445C proaerolysin with the new intrachain bridge were both easily detected inside the bacteria, and they later appeared in the culture supernatants. Small amounts of incorrectly folded proaerolysin were also observed in the cells, but they were not secreted. We conclude that proaerolysin folds and dimerizes before being released from the cell, and that correct folding is a requirement for secretion to occur. The proton ionophore CCCP reduced release of the folded proteins. Unoxidized protein was secreted by cells grown in β-mercaptoethanol and by a dsbA mutant of V. cholerae, indicating that disulphide bond formation may not be essential for release.

    Notes: Aerolysin is one of a large group of bacterial proteins that can kill target cells by forming discrete channels in their plasma membranes. The toxin has many properties in common with the porins of the Gram-negative bacterial outer membrane, including an extensive amount of β-structure, a high proportion of hydrophilic amino acid side-chains and no hydrophobic stretches in the primary structure. It also oligomerizes to produce an insertion-competent state. Aerolysin is secreted as a dimer by members of the Aeromonas family. It binds to a high-affinity receptor on the target cell that has recently been shown to be a glycosylphosphatidylinositol-anchored glycoprotein. Binding is followed by heptamerization to form a structure that we propose contains a β-barrel which can insert into the membrane and produce a channel.

    Notes: Human theta-class glutathione S-transferases (GST's) appear to play a critical role in the metabolism of a variety of environmental pollutants but in some cases the products of the reaction are carcinogenic. Crystals of a human theta-class GST, namely hGSTT2-2, have been grown from polyethylene glycol by the hanging-drop vapour-diffusion method. The crystals belong to the trigonal space group P3121 with cell dimensions of a = b = 94.0 and c = 120.5 Å. They contain two monomers in the asymmetric unit and diffract to 3.0 Å resolution.
