    Notes: Four soluble types of polymers have been synthesized by grafting 4- and 4′-chalkonyl glycidyl ethers onto an aromatic polyamine (aniline-formaldehyde linear resin) and a polyamide (condensation product of dimerized linseed oil and an aliphatic polyamine) to obtain photo-crosslinkable chalkone structure in the side chains which are also expected to contain one —OH group each. The completion of grafting reactions has been followed by the disappearance of the epoxide absorption around 918 cm-1. The characterization of the resins was done by melting point, solubility, and IR measurements. Irradiation was carried out with a 100-watt Hanovia high-pressure mercury are lamp. Irradiated samples were studied by UV spectral measurements and their insolubility in CHCl3. It was observed that polymers grafted with 4′-chalkonyl glycidyl ether possibly undergo both isomerization and crosslinking reactions under UV irradiation while those grafted with 4-chalkonyl glycidyl ether undergo crosslinking reaction predominantly.

    Notes: Nine photocrosslinkable unsaturated polyesters containing pendent chalconyl and cinnamylidenemalonyl moieties have been synthesized by a route involving no water elimination. The resins are insolubilized by visible and UV irradiation. They can also be cured with styrene, MEK peroxide, and cobalt naphthenate. A study of the photoreactions reveals that the resins containing 4′-chalconyl groups crosslink and isomerize simultaneously, whereas those containing 4-chalconyl groups undergo crosslinking only, due to the steric suppression of isomerization. Photoinsolubility is also achieved by introducing the cinnamylidenemalonyl group into the resin structure. It is found that the resins containing both the chalconyl and cinnamylidenemalonyl groups in the repeat units do not exhibit any pronounced photofastness.

    Notes: Syntheses, characterization and photoreactions of six diglycidyl ethers obtained by condensing 2,2′, 2′,4, and 4,4′-dihydroxychalcones with epichlorohydrin in two different molar proportions have been reported. It has been observed that the well known cyclodimerization and trans-cis isomerization of the chalcone moiety under photoirradiation could be significantly affected to the extent of completely supressing the isomerization in 2,2′, and 2′,4-chalconic resins by steric factors. Increase in molecular weight made the photodimerization difficult in the 2,2′, and 2′,4-chalocnic resins whereas the 4,4′-chalconic resin remained unaffected. It has been suggested that the chalconic resin could be used in photomechanical processes mixed with aryldiazonium fluoroborates. These additives are known to decompose under photoirradiation yielding BF3 which is a crosslinking agent for the epoxide group.

    Description: NADPH-cytochrome P450 oxidoreductase (CYPOR) is essential for electron donation to microsomal cytochrome P450-mediated monooxygenation in such diverse physiological processes as drug metabolism (approximately 85–90% of therapeutic drugs), steroid biosynthesis, and bioactive metabolite production (vitamin D and retinoic acid metabolites). Expressed by a single gene, CYPOR’s role with these multiple redox partners renders it a model for understanding protein–protein interactions at the structural level. Polymorphisms in human CYPOR have been shown to lead to defects in bone development and steroidogenesis, resulting in sexual dimorphisms, the severity of which differs significantly depending on the degree of CYPOR impairment. The atomic structure of human CYPOR is presented, with structures of two naturally occurring missense mutations, V492E and R457H. The overall structures of these CYPOR variants are similar to wild type. However, in both variants, local disruption of H bonding and salt bridging, involving the FAD pyrophosphate moiety, leads to weaker FAD binding, unstable protein, and loss of catalytic activity, which can be rescued by cofactor addition. The modes of polypeptide unfolding in these two variants differ significantly, as revealed by limited trypsin digestion: V492E is less stable but unfolds locally and gradually, whereas R457H is more stable but unfolds globally. FAD addition to either variant prevents trypsin digestion, supporting the role of the cofactor in conferring stability to CYPOR structure. Thus, CYPOR dysfunction in patients harboring these particular mutations may possibly be prevented by riboflavin therapy in utero, if predicted prenatally, or rescued postnatally in less severe cases.
