Notes: Summary Aroclor 1242 is inhibitory to long-term batch growth ofE uglena gracilis at 10 ppm. Exposure to PCB's, subsequent to an initial drop of 50% in the first 30 minutes, does not appear to inhibit photosynthesis on a per cell basis over the time span of four hours. Consequently our experimental findings suggest that inhibition of population growth does not lie directly in the photosynthetic pathway despite the initial inhibition by Aroclor 1242 onEuglena gracilis in “light”. “Dark” absorption of bicarbonate appears to decrease with increased PCB exposure. This decrease may be causally related to the population growth inhibition observed.

Notes: Abstract The heats of fusion for crystalline uridine and its phosphate derivative, uridine-3′-(2′)-phosphate, as well as the enthalpic and entropic values for the polymerization of a mixture of these compounds, have been determined by Differential Scanning Calorimetry. An analog computer analysis of the exotherm associated with heating of the mixture indicated a minimum of five processes, which could be rationalized on the basis of previously published experimental findings.

Notes: Abstract Differential scanning calorimetry (DSC) has provided a universal tool for ascertaining the effects of preparation and thermal history on the physical characteristics of polymers. When applied to the study of nonpolymeric compounds, however, the interpretation of thermal analysis data may be complicated by the possible occurrence of chemical as well as physical transformations under thermal stress. In the present study involving 2'-deoxynucleosides, this difficulty has been resolved by utilizing a DSC-TLC combination to correlate thermal phenomena with corresponding chemical and physical changes. This combination shows great promise in extending the application of DSC to the investigation of thermal reactions.

Notes: Abstract Heat may be a mutagenic agent which could be more important to evolution than are the ordinary errors in DNA replication. Therefore, it is necessary to know the stability of heredity determinants to thermal energy. Consequently, a thermal analysis, via differential scanning calorimetry (DSC), has been undertaken, using model compounds (crystalline 2'-deoxyribonucleosides and their 5-halo derivatives) to provide information regarding: 1. bond free energies maintaining the fibrous structure in nucleic acids e.g. H-bonding,π-complexing and dipole induced dipole interaction. 2. susceptibility to thermal degradation e.g. thermolytic cleavage of the glycosidic bond and deamination of the bases. Interpretation of the thermal curves has been facilitated and enhanced by utilization of a DSC-TLC analytical technique. Based on these data, a mechanism for the thermolysis of the glycosidic bond in the melt has been considered.

Notes: Differential enthalpic analysis of a series of 3′,5′-cyclic nucleotides indicates that homolytic cleavage of the six-membered phosphate ring occurs either immediately prior to or concurrent with decomposition of the crystal lattice. Homolysis is followed by a rapid polymerization to yield oligonucleotides. The enthalpies of these reactions have, with the exception of guanosine 3′,5′-cyclic phosphate, almost identical values of 25 kcal/mole. The anomalous case is attributed to a more stabilized phosphate ring as a result of hydrogen bonding through the two position of the purine ring. The pair of overlapping exothermic peaks observed in each of the thermograms for cAMP and cIMP is related to the presence of two conformational arrangements within the unit cell of each compound.
