Summary The DNA of the phage ϕ80iλdhis, which carries the histidine operon (his) of Escherichia coli K12, has been used as template for transcription in vitro catalyzed by E. coli RNA polymerase. The product obtained has been characterized through hybridization to the DNA of the phages ϕ80iλdhis and ϕ80iλ. Hybridization to the separated strands and hybridization competition show that the his operon is actually transcribed in vitro on the correct DNA strand.
