    Notes: Summary Tryptophan (Trp) residues within copoly(L-glutamic acid-co-L-Trp) (GAT) form a tricyclic structure when treatment with trifluoroacetic acid (TFA) and show reversible color change from yellow (above pH 8.0) to red (below pH 7.0) when the nitrogen atom in an indole ring dissociates positively in the neutral pH region. The interaction between TFA-treated GAT (GAT-T) and an anionic molecule such as warfarin was investigated using VIS and fluorescence spectrophotometry. Trp residues within GAT-T were found to interact with warfarin electrostatically besides hydrophobic interaction resulting in a color change from red to yellow. Release behavior of warfarin from GAT-T microspheres (MS) was also investigated. Binding amount and retention time of warfarin in GAT-T MS were found to be superior to those in untreated GAT MS.

    Notes: Summary Effects of anionic lipids and heparin on the conformation of plasma proteins, such as albumin, γ-globulin, and fibrinogen were investigated by evaluating both α-helix and β-structure contents from circular dichroism (CD) spectra. Sodium dodecyl sulfate and sodium dodecylbenzenesulfonate increased the α-helix content of γ-globulin, and β-structure content of fibrinogen accompanied with decreasing α-helix content, while they stepwise disrupted the α-helix of albumin with increasing their concentration. It was concluded that the hydrophobic interaction was predominant for binding albumin, while the electrostatic interaction affected the structural changes of γ-globulin more than hydrophobic interaction.

    Notes: The rate constants of hydrolysis of the enantiomers of amino acid nitrophenyl esters by catalytic domains composed of cationic surfactant aggregates and dipeptide catalysts or Ndecanoyl-L-histidine have been determined at pH 7.30. The dipeptide catalysts shows large rate enhancement and stereoselectivity in aggregate domains. The surfactant structural effects are examined by investigation of the rate constants and stereoselectivities, and the nature of stereoselective catalysis is discussed.

    Notes: This paper is concerned with optimization of the operating mode of a fermentor. Combining the various modes of operation - batch, semibatch, and continuous - the operating pattern which maximizes the desired metabolic product in a single fermentor is determined by using Kelley's transformation method with Pontryagin's maximum principle. Kelley's transformation method is a device which avoids the singular situation which occurs when the usual procedure of selecting the optimal control function by the maximum principle breaks down. This is the case in the problem considered in this paper. For lysine fermentation, the best operating mode depends on the fermentor capacity and operating time. The results of this study are summarized thus: (i) when the operating time is “long enough,” optimal conditions require that continuous operation follows either semibatch and/or batch operation, and (ii) when the fermentor capacity becomes “large enough,” semibatch operation becomes important.

    Notes: This work is concerned with the optimization study of the semibatch fermentation by which an amino acid is produced. The particular fermentation studied is the synthesis of lysine by the auxotrophic mutant. Applying Green's theorem to the maximization problem was proposed, and it succeeded in determining the feed rate of the substrate that maximized the production rate of the desired product.

    Notes: Summary Tryptophan within copoly(N-hydroxyethyl L-glutamine-co-L-tryptophan) treated with nitric acid (EGT-N) dissociates negatively above pH 10.5 by the electron-attractive effect of nitro groups introduced into the indole ring. The interaction between EGT-N and various metal ions was investigated using UV/VIS, ESCA, and ICP measurement. EGT-N was found to selectively form a complex with cobalt ions above pH 10.5. The formation of the cobalt ion complex of EGT-N was confirmed to be caused by the cooperative coordination of the tryptophan-introduced nitro groups and the OH groups of hydroxyethyl L-glutamine.

    Notes: Summary The binding of fibrinogen with heparin and dextran sulfates, and of antithrombin III with heparin and dextran sulfate were investigated by sedimentation velocity method. From the measurement of fibrinogen-acidic polysaccharide systems, it was confirmed that heparin and low molecular weight dextran sulfate(DSC) forms soluble complexes with fibrinogen, though the amount bound of the latter was rather small compared with high molecular weight dextran sulfate(DSD). Antithrombin III was also found to be bound by heparin and DSC. The numbers of heparin molecules bound to one molecule of fibrinogen and antithrombin III were estimated to be 4.2 and 0.3, respectively.

    Notes: Summary Retardation effect of acidic polysaccharides on the interactions of fibrinogen with thrombin was kinetically investigated by measuring the turbidity as a measure of the concentration of fibrin polymer formed in the system. To this aim, the fibrinogenfibrin conversion curve was analyzed kinetically based on the elementary reactions, and the slope and the induction period of the conversion curve were expressed as functions of the concentrations of fibrinogen, thrombin, and polysaccharide. The strong retardation effect of dextran sulfate and heparin was quantitatively interpreted. Seven kinds of polysaccharides were examined.
