    Description: Hair cells possess a single primary cilium, called the kinocilium, early in development. While the kinocilium is lost in auditory hair cells of most species it is maintained in vestibular hair cells. It has generally been believed that the primary role of the kinocilium and cilia-associated genes in hair cells is in the establishment of the polarity of actin-based stereocilia, the hair cell mechanotransduction apparatus. Through genetic screening and testing of candidate genes in zebrafish (Danio rerio) we have found that mutations in multiple cilia genes implicated in intraflagellar transport ( dync2h1 , wdr35 , ift88 , and traf3ip) , and the ciliary transition zone ( cc2d2a , mks1 , and cep290 ) lead to resistance to aminoglycoside-induced hair cell death. These genes appear to have differing roles in hair cells, as mutations in intraflagellar transport genes, but not transition zone genes, lead to defects in kinocilia formation and processes dependent upon hair cell mechanotransduction activity. These mutants highlight a novel role of cilia-associated genes in hair cells, and provide powerful tools for further study.

    Description: ITER cryolines (CLs) are complex network of vacuum-insulated multi and single process pipe lines, distributed in three different areas at ITER site. The CLs will support different operating loads during the machine life-time; either considered as nominal, occasional or exceptional. The major loads, which form the design basis are inertial, pressure, temperature, assembly, magnetic, snow, wind, enforced relative displacement and are put together in loads specification. Based on the defined load combinations, conceptual estimation of reaction loads have been carried out for the lines located inside the Tokamak building. Adequate numbers of embedded plates (EPs) per line have been defined and integrated in the building design. The finalization of building EPs to support the lines, before the detailed design, is one of the major design challenges as the usual logic of the design may alter.     Description: System of Cryoline and Cryo-distribution for ITER has matured to a stage of preliminary design with the advent of industrial associates. Starting from the cold power source, the system of Cryoline and Cryo-distribution transfers the controlled cold power through a large network to the superconducting magnets and cryopumps. The functional responsibility also includes very high reliability and availability with respect to the operation of the ITER machine. Following the completion of conceptual design, it was necessary to perform a detailed engineering study of the complete network of distribution system in totality, before entering in to the industrial phase.     Description: The first experimental evidence of detecting the neutrons co-related with the natural atmospheric lightning discharges (NALD) was obtained with Lead-Free Gulmarg Neutron Monitor (LFGNM) operating at High Altitude Research Laboratory(HARL), Gulmarg, Kashmir, India and was reported in the year 1985 [ Shah et al. , 1985]. The neutron observations still continue with LFGNM. However, the current configuration of LFGNM is the upgraded version of the system used earlier to record neutron bursts (in the recording period of 320 μ s in four successive electronic gates of 80 μ s each) supposedly originating from an NALD. In the current system the neutron recording time period/interval has been extended to 1260 μ s with 63 successive gates of 20 μ s each. The system also simultaneously records the differential times -maximum upto fourteen- between the consecutive strokes of a multi-stroke lightning flash. The distance between an NALD channel and LFGNM setup is determined empirically by making use of the time delay ( t d )/time of flight (TOF) measurement of the first detected neutron subsequent to the sensing of the electrostatic field variation caused due to the initiation of an NALD in the ambient atmosphere of the LFGNM setup. Assuming a priori incident energy as 2.45 MeV of the detected neutrons supposedly generated due to the fusion of deuterium ions in the lightning discharge channel leads to quantifying the neutron emission flux if the NALD channel distance with respect to the LFGNM setup is established. In this paper we discuss the experiment and the time profiles of several of the large number of the major neutron burst events recorded with LFGNM in association with NALDs. Moreover, a rare and an extra-ordinary neutron burst event, in terms of its associated ' t d /TOF' of first detected neutron after triggering, recorded by this system is specifically discussed. In this event, the recorded TOF of 14 μ s of the escaping neutron detected by the system immediately after getting triggered by the NALD that struck a nearby tree found located just around 300 meters (physically measured) away from the detector position indicates the energy of the detected neutron ϵ n ≈2.45 MeV. In the light of this only event we, therefore, cautiously suggest deuteron-deuteron fusion reaction, 2 H ( 2 H , n ) 3 H e , as one of the possible mechanism of the neutron generation co-related with an NALD. Nonetheless, the observations so far have reconfirmed production of neutrons in an NALD.

    Description: The autism spectrum disorders (ASDs) are a group of conditions characterized by impairments in reciprocal social interaction and communication, and the presence of restricted and repetitive behaviours. Individuals with an ASD vary greatly in cognitive development, which can range from above average to intellectual disability. Although ASDs are known to be highly heritable ( approximately 90%), the underlying genetic determinants are still largely unknown. Here we analysed the genome-wide characteristics of rare (〈1% frequency) copy number variation in ASD using dense genotyping arrays. When comparing 996 ASD individuals of European ancestry to 1,287 matched controls, cases were found to carry a higher global burden of rare, genic copy number variants (CNVs) (1.19 fold, P = 0.012), especially so for loci previously implicated in either ASD and/or intellectual disability (1.69 fold, P = 3.4 x 10(-4)). Among the CNVs there were numerous de novo and inherited events, sometimes in combination in a given family, implicating many novel ASD genes such as SHANK2, SYNGAP1, DLGAP2 and the X-linked DDX53-PTCHD1 locus. We also discovered an enrichment of CNVs disrupting functional gene sets involved in cellular proliferation, projection and motility, and GTPase/Ras signalling.     Description: We report an improved draft nucleotide sequence of the 2.3-gigabase genome of maize, an important crop plant and model for biological research. Over 32,000 genes were predicted, of which 99.8% were placed on reference chromosomes. Nearly 85% of the genome is composed of hundreds of families of transposable elements, dispersed nonuniformly across the genome. These were responsible for the capture and amplification of numerous gene fragments and affect the composition, sizes, and positions of centromeres. We also report on the correlation of methylation-poor regions with Mu transposon insertions and recombination, and copy number variants with insertions and/or deletions, as well as how uneven gene losses between duplicated regions were involved in returning an ancient allotetraploid to a genetically diploid state.
