Description: Many human cancers originate from defects in the DNA damage response (DDR). Although much is known about this process, it is likely that additional DDR genes remain to be discovered. To identify such genes, we used a strategy that combines protein-protein interaction mapping and large-scale phenotypic analysis in Caenorhabditis elegans. Together, these approaches identified 12 worm DDR orthologs and 11 novel DDR genes. One of these is the putative ortholog of hBCL3, a gene frequently altered in chronic lymphocytic leukemia. Thus, the combination of functional genomic mapping approaches in model organisms may facilitate the identification and characterization of genes involved in cancer and, perhaps, other human diseases.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Boulton, Simon J -- Gartner, Anton -- Reboul, Jerome -- Vaglio, Philippe -- Dyson, Nick -- Hill, David E -- Vidal, Marc -- 5R01HG01715-02/HG/NHGRI NIH HHS/ -- 7 R33 CA81658-02/CA/NCI NIH HHS/ -- P01CA80111-02/CA/NCI NIH HHS/ -- New York, N.Y. -- Science. 2002 Jan 4;295(5552):127-31.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Dana-Farber Cancer Institute and Department of Genetics, Harvard Medical School, Boston, MA 02115, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/11778048" target="_blank"〉PubMed〈/a〉

Description: Deletions or mutations of the retinoblastoma gene, RB1, are common features of many tumors and tumor cell lines. Recently, the RB1 gene product, p105-RB, has been shown to form stable protein/protein complexes with the oncoproteins of two DNA tumor viruses, the adenovirus E1A proteins and the simian virus 40 (SV40) large T antigen. Neither of these viruses is thought to be associated with human cancer, but they can cause tumors in rodents. Binding between the RB anti-oncoprotein and the adenovirus or SV40 oncoprotein can be recapitulated in vitro with coimmunoprecipitation mixing assays. These assays have been used to demonstrate that the E7 oncoprotein of the human papilloma virus type-16 can form similar complexes with p105-RB. Human papilloma virus-16 is found associated with approximately 50 percent of cervical carcinomas. These results suggest that these three DNA viruses may utilize similar mechanisms in transformation and implicate RB binding as a possible step in human papilloma virus-associated carcinogenesis.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Dyson, N -- Howley, P M -- Munger, K -- Harlow, E -- CA13106/CA/NCI NIH HHS/ -- New York, N.Y. -- Science. 1989 Feb 17;243(4893):934-7.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Cold Spring Harbor Laboratory, NY 11724.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/2537532" target="_blank"〉PubMed〈/a〉

Description: Antibodies that modulate receptor function have great untapped potential in the control of stem cell differentiation. In contrast to many natural ligands, antibodies are stable, exquisitely specific, and are unaffected by the regulatory mechanisms that act on natural ligands. Here we describe an innovative system for identifying such antibodies by...

Notes: Abstract To control environmental pollution, a biodegradation technique involving a biomass is used to degrade the liquid wastes from certain industrial processes before disposal. (d, p) reactions were used to study the degradation process and the extent of removal of carbon and nitrogen. The PIXE-PIGME technique was used for the simultaneous study of the compositions of the biomass, the feed and the treated material. Measurements were obtained using protons of 2.5 MeV (for the PIXE-PIGME measurements) and deuterons of 2.0 MeV using currents of the order of 25nA. The X-ray and γ-ray data were recorded simultaneously. Absolute concentrations were obtained by comparison with measurements of calibration standards. Results show that 54–58% of the carbon and 45–50% of the nitrogen is removed from the wastes and that the biomass is concentrating Cl, Ti, Ni and Fe.

Notes: The thick-target yield of characteristic x-rays from a series of gold-silver binary alloys has been determined experimentally (relative to the yield from the pure elements) under proton bombardment, using proton energies of 1. 5, 2. 0 and 2. 5 MeV. The yield per unit concentration of gold or silver varies with alloy composition because of the progressively varying effects of proton stopping power (specific energy loss) and x-ray self-absorption in the target materials. Theoretical yields have been calculated using tabulated data for stopping power and x-ray absorption. When both of these effects are taken into account, agreement with experiment is satisfactory. These results can be made the basis of stopping power and self-absorption corrections in proton-induced x-ray emission analysis.

Notes: The self-absorption effect for characteristic x-rays produced by proton bombardment of thick targets has been studied experimentally by the ‘tracer-layer’ and ‘angular distribution’ techniques. Four proton energies have been used. The experimental data have been converted into f(χ) curves which can be used to apply a self-absorption correction in PIXE microanalysis.
